    Description: Convergence is a central concept in evolutionary studies because it provides strong evidence for adaptation. It also provides information about the nature of the fitness landscape and the repeatability of evolution, and can mislead phylogenetic inference. To understand the role of adaptive convergence, we need to understand the patterns of nonadaptive convergence. Here, we consider the relationship between nonadaptive convergence and divergence in mitochondrial and model proteins. Surprisingly, nonadaptive convergence is much more common than expected in closely related organisms, falling off as organisms diverge. The extent of the convergent drop-off in mitochondrial proteins is well predicted by epistatic or coevolutionary effects in our "evolutionary Stokes shift" models and poorly predicted by conventional evolutionary models. Convergence probabilities decrease dramatically if the ancestral amino acids of branches being compared have diverged, but also drop slowly over evolutionary time even if the ancestral amino acids have not substituted. Convergence probabilities drop-off rapidly for quickly evolving sites, but much more slowly for slowly evolving sites. Furthermore, once sites have diverged their convergence probabilities are extremely low and indistinguishable from convergence levels at randomized sites. These results indicate that we cannot assume that excessive convergence early on is necessarily adaptive. This new understanding should help us to better discriminate adaptive from nonadaptive convergence and develop more relevant evolutionary models with improved validity for phylogenetic inference.

    Description: The paper deals with the investigation of Lamb waves ultrasonic testing technique applied for evaluation of different stress-strain and damaged state of aluminum specimens at static and fatigue loading in order to develop a Structural Health Monitoring (SHM) approach. The experimental results of tensile testing of AA2024T3 specimens with welded joints are presented. Piezoelectric transducers used as actuators and sensors were adhesively bonded to the specimen's surface using two component epoxy. The set of static and cyclic tensile tests with two frequencies of acoustic testing (50 kHz and 335 kHz) were performed. The recorded signals were processed to calculate the maximum envelope in order to evaluate the changes of the stress-strain state of the specimen and its microstructure during static tension.     Description: Duchenne muscular dystrophy (DMD) is a lethal, X-linked muscle-wasting disease caused by lack of the cytoskeletal protein dystrophin. There is currently no cure for DMD although various promising approaches are progressing through human clinical trials. By pharmacologically modulating the expression of the dystrophin-related protein utrophin, we have previously demonstrated in dystrophin-deficient mdx studies, daily SMT C1100 treatment significantly reduced muscle degeneration leading to improved muscle function. This manuscript describes the significant disease modifying benefits associated with daily dosing of SMT022357, a second-generation compound in this drug series with improved physicochemical properties and a more robust metabolism profile. These studies in the mdx mouse demonstrate that oral administration of SMT022357 leads to increased utrophin expression in skeletal, respiratory and cardiac muscles. Significantly, utrophin expression is localized along the length of the muscle fibre, not just at the synapse, and is fibre-type independent, suggesting that drug treatment is modulating utrophin transcription in extra-synaptic myonuclei. This results in improved sarcolemmal stability and prevents dystrophic pathology through a significant reduction of regeneration, necrosis and fibrosis. All these improvements combine to protect the mdx muscle from contraction induced damage and enhance physiological function. This detailed evaluation of the SMT C1100 drug series strongly endorses the therapeutic potential of utrophin modulation as a disease modifying therapeutic strategy for all DMD patients irrespective of their dystrophin mutation.

    Description: This article presents results of mercury in surface waters from Hunza River basin, Northern Areas, Pakistan. Small-scale gold mining activities along Hunza and Gilgit rivers are long known to be discharging mercury in the amalgamation and roasting processes. Previous studies reported high mercury concentrations in soils close to mining operations as well as serious health problems for miners. However, none of the studies have focused on the level of contamination in aqueous environments. This is the first study on the investigation of source and fate of sediment and river-borne mercury in Hunza River. Samples collected near panning sites showed higher mercury concentrations than critical levels established by the U.S. Environmental Protection Agency. The observed dissolved mercury concentrations ranged from 5.10 to 25.25 ng/L, whereas particulate-bound mercury ranged from 4.85 to 154.62 ng/L. Particulate phase mercury corresponded to more than 75% of the total observed mercury concentrations for all of the sampled rivers. Thus, suspended sediments represented the major pathway of the riverine mercury transport. A mass balance calculation suggested a 48.6 g/km 2 of annual mercury flux into Hunza River basin. The samples collected from the most affected river, Shimsal River, averaged to have 108 ng/L total mercury concentration. This number was close to the average soil mercury data from PMDC (2001) of 151 ng/L. The dominant source of contamination was shown to be the leaching of large quantities of mercury from the Hg-rich sediment and flood plain soil into the rivers rather than the direct release from mining activities. Significant decrease in both dissolved and particulate-bound mercury concentration downstream of Attabad Lake suggested that mercury is being accumulated or consumed in the lake. Although minimization or elimination of mercury loses from mining process seems important for the wellbeing of the miners, preventing remobilization of accumulated mercury is equally important in mercury control in this region.     Description: Theileria annulata and T. parva are closely related protozoan parasites that cause lymphoproliferative diseases of cattle. We sequenced the genome of T. annulata and compared it with that of T. parva to understand the mechanisms underlying transformation and tropism. Despite high conservation of gene sequences and synteny, the analysis reveals unequally expanded gene families and species-specific genes.     Description: We report the genome sequence of Theileria parva, an apicomplexan pathogen causing economic losses to smallholder farmers in Africa. The parasite chromosomes exhibit limited conservation of gene synteny with Plasmodium falciparum, and its plastid-like genome represents the first example where all apicoplast genes are encoded on one DNA strand. We tentatively identify proteins that facilitate parasite segregation during host cell cytokinesis and contribute to persistent infection of transformed host cells. Several biosynthetic pathways are incomplete or absent, suggesting substantial metabolic dependence on the host cell.     Description: Diacylglycerol kinase catalyses the ATP-dependent phosphorylation of diacylglycerol to phosphatidic acid for use in shuttling water-soluble components to membrane-derived oligosaccharide and lipopolysaccharide in the cell envelope of Gram-negative bacteria. For half a century, this 121-residue kinase has served as a model for investigating membrane protein enzymology, folding, assembly and stability. Here we present crystal structures for three functional forms of this unique and paradigmatic kinase, one of which is wild type. These reveal a homo-trimeric enzyme with three transmembrane helices and an amino-terminal amphiphilic helix per monomer. Bound lipid substrate and docked ATP identify the putative active site that is of the composite, shared site type. The crystal structures rationalize extensive biochemical and biophysical data on the enzyme.     Description: G protein-coupled receptors (GPCRs) are responsible for the majority of cellular responses to hormones and neurotransmitters as well as the senses of sight, olfaction and taste. The paradigm of GPCR signalling is the activation of a heterotrimeric GTP binding protein (G protein) by an agonist-occupied receptor. The beta(2) adrenergic receptor (beta(2)AR) activation of Gs, the stimulatory G protein for adenylyl cyclase, has long been a model system for GPCR signalling. Here we present the crystal structure of the active state ternary complex composed of agonist-occupied monomeric beta(2)AR and nucleotide-free Gs heterotrimer. The principal interactions between the beta(2)AR and Gs involve the amino- and carboxy-terminal alpha-helices of Gs, with conformational changes propagating to the nucleotide-binding pocket. The largest conformational changes in the beta(2)AR include a 14 A outward movement at the cytoplasmic end of transmembrane segment 6 (TM6) and an alpha-helical extension of the cytoplasmic end of TM5. The most surprising observation is a major displacement of the alpha-helical domain of Galphas relative to the Ras-like GTPase domain.
