    Description: Harmonic multiplying operation in a gyrotron traveling-wave amplifier (gyro-TWA) permits for magnetic field reduction and frequency multiplication. Lowering a beam voltage is an important step toward miniaturization of a harmonic multiplying gyro-TWA. However, the additional degree of freedom that is provided by the multitude cyclotron harmonics in a low-voltage harmonic multiplying gyro-TWA still easily generates various competing modes. An improved mode-selective circuit, using circular waveguides with various radii, can provide the rejection points within the frequency range to suppress competing modes. Simulated results reveal that the mode-selective circuit can provide an attenuation of more than 14 dB to suppress the competing modes. Furthermore, the performance of the gyro-TWA is analyzed for studying the sensitivity of the saturated output power and full width at half maximum bandwidth of the gyro-TWA to the beam voltage and the magnetic field. A stable low-voltage harmonic multiplying gyro-TWA with the mode-selective circuit is predicted to yield a peak output power of 24 kW at 200.4 GHz, corresponding to a saturated gain of 56 dB at an interaction efficiency of 20%. The full width at half maximum bandwidth is 3.0 GHz.

    Description: The DED1 gene, which encodes a putative RNA helicase, has been implicated in nuclear pre-messenger RNA splicing in the yeast Saccharomyces cerevisiae. It is shown here by genetic and biochemical analysis that translation, rather than splicing, is severely impaired in two newly isolated ded1 conditional mutants. Preliminary evidence suggests that the protein Ded1p may be required for the initiation step of translation, as is the distinct DEAD-box protein, eukaryotic initiation factor 4A (eIF4A).     Description: It has been argued that the evolution of plant genome size is principally unidirectional and increasing owing to the varied action of whole-genome duplications (WGDs) and mobile element proliferation. However, extreme genome size reductions have been reported in the angiosperm family tree. Here we report the sequence of the 82-megabase genome of the carnivorous bladderwort plant Utricularia gibba. Despite its tiny size, the U. gibba genome accommodates a typical number of genes for a plant, with the main difference from other plant genomes arising from a drastic reduction in non-genic DNA. Unexpectedly, we identified at least three rounds of WGD in U. gibba since common ancestry with tomato (Solanum) and grape (Vitis).     Description: Signalling by Wnt proteins is finely balanced to ensure normal development and tissue homeostasis while avoiding diseases such as cancer. This is achieved in part by Notum, a highly conserved secreted feedback antagonist. Notum has been thought to act as a phospholipase, shedding glypicans and associated Wnt proteins from the cell surface. However, this view fails to explain specificity, as glypicans bind many extracellular ligands. Here we provide genetic evidence in Drosophila that Notum requires glypicans to suppress Wnt signalling, but does not cleave their glycophosphatidylinositol anchor. Structural analyses reveal glycosaminoglycan binding sites on Notum, which probably help Notum to co-localize with Wnt proteins. They also identify, at the active site of human and Drosophila Notum, a large hydrophobic pocket that accommodates palmitoleate.     Description: Motivation: Despite many attempts for algorithm development in recent years, automated identification of intact glycopeptides from LC-MS 2 spectral data is still a challenge in both sensitivity and precision. Results: We implemented a supervised machine learning algorithm, Random Forest, in an automated workflow to identify N-glycopeptides using spectral features derived from ion trap-based LC-MS 2 data. The workflow streamlined high-confident N-glycopeptide spectral data and enabled adaptive model optimization with respect to different sampling strategies, training sample size and feature set. A critical evaluation of the features important for glycopeptide identification further facilitated effective feature selection for model improvement. Using split sample testing method from 577 high-confident N-glycopeptide spectral data, we demonstrated that an optimal true-positive rate, precision and false-positive rate of 73, 88 and 10%, respectively, can be attained for overall N-glycopeptide identification Availability and implementation: The workflow developed in this work and the application suite, Sweet-Heart, that the workflow supports for N-glycopeptide identification are available for download at http://sweet-heart.glycoproteomics.proteome.bc.sinica.edu.tw/ .     Notes: Generation of a uniform density plasma has been a key consideration in the design of microwave/plasma processing systems. A circularly polarized wave can generate a plasma with good azimuthal uniformity as well as provide strong resonant interaction with the plasma electrons. In this article, we report the development of a three-port polarization converter which efficiently converts the TE10 wave of a standard rectangular waveguide into a circularly polarized TE11 wave of a cylindrical waveguide. Employing the same principle for mode conversion, the converter is also made to function as a protective device of the high power microwave source by providing a separate port for the return and damping of the reflected wave. Such a converter has been analyzed, constructed, and tested. At the operating frequency of 2.45 GHz, test results indicate a 25 dB return loss, 97% end-to-end polarization conversion efficiency, and better than 20 dB rejection of the reflected wave. The structural simplicity of the device allows high power operation as well as easy construction for system research and applications. © 1999 American Institute of Physics.
