Immunochemical Studies on the 0-Antigens of Cell- Wall Polysaccharides from Salmonella Bacteria. Synthesis of the p-Nitrophenyl-α-pyranosides of 2-0- and 6-O-Acetyl-D-galactose and of 2-0-Acetyl-abequoseTreatment of p-nitrophenyl-α-D-galactoside with acetone/copper sulphate gives p-nitrophenyl-4,6-O-isopropylidene-α-D-galactoside (5) and p-nitrophenyl-3,4-O-isopropylidene-α-D-galactoside (6). Tritylation and acetylation of 6 yield p-nitrophenyl-2-O-acetyl-3,4-O-isopropyl-idene-6-O-trityl-α-D-galactoside (8). After removal of all protecting residues by treatment with 80% acetic acid, p-nitrophenyl-2-O-acetyl-α-D-galactoside is obtained. Partial acetylation of 6 produces a mixture of p-nitrophenyl-2-O-acetyl-3,4-O-isopropylidene-α-D-galactoside (13)and p-nitrophenyl-6-O-acetyl-3,4-O-isopropylidene-α-D-galactoside (12) which are separated by column chromatography. Heating of 12 at 65% with 80% acetic acid leads to p-nitro-phenyl-6-O-acetyl-α-D-galactoside (15). - Partial acetylation of p-nitrophenyl-α-D-abequoside (16) yields p-nitrophenyl-2-O-acetyl-α-D-abequoside (17) and p-nitrophenyl-4-O-acetyl-α

Aus p-Nitro-phenyl-α-D-galaktosid (4) werden mit Aceton/Kupfersulfat p-Nitro-phenyl-4.6-O-isopropyliden-α-D-galaktosid (5) und p-Nitrophenyl-3.4-O-isopropyliden-α-D-galaktosid (6) dargestellt. Die Umsetzung von 6 mit Tritylchlorid und Acetanhydrid liefert p-Nitro-phenyl-2-O-acetyl-3.4-O-isopropyliden-6-O-trityl-α-D-galaktosid (8). Nach Abspaltung der Schutzgruppen mit 8Oproz. Essigsaure wird p-Nitro-phenyl-2-O-acetyl-α-d-galaktosid (9) erhalten. Monoacetylierung von 6 ergibt p-Nitro-phenyl-2-O-acetyl-3.4-O-isopropyliden-α-D-galaktosid (13) und p-Nitro-phenyl-6-O-acetyl-3.4-O-isopropyliden-α-d-galaktosid (12), die säulen chromatographisch getrennt werden. Durch Kochen von 12 mit 80proz. Essigsäureresultiert p-Nitro-phenyl-6-O-acetyl-α-D-galaktosid (15). - Durch Monoacetylierung von p-Nitro-phenyl-α-D-abequosid (16) werden p-Nitro-phenyl-2-O-acetyl-α-D-abequosid (17) und p-Nitro-phenyl-4-O-acetyl-α-D-abequosid (18) synthetisiert, die ebenfalls chromatographisch getrennt werden. Die Stellung der Acetylgruppe ist massenspektrometrisch an den aus 17 und 18 erhaltenen 4-0-Methyl-1.2.5-tri-0-acetyl- und 2-0-Methyl-1.4.5-tri-0-acetyl-abequiten (25 und 26) bestimmt worden.
