Description: The 5'-leader of the HIV-1 genome regulates multiple functions during viral replication via mechanisms that have yet to be established. We developed a nuclear magnetic resonance approach that enabled direct detection of structural elements within the intact leader (712-nucleotide dimer) that are critical for genome packaging. Residues spanning the gag start codon (AUG) form a hairpin in the monomeric leader and base pair with residues of the unique-5' region (U5) in the dimer. U5:AUG formation promotes dimerization by displacing and exposing a dimer-promoting hairpin and enhances binding by the nucleocapsid (NC) protein, which is the cognate domain of the viral Gag polyprotein that directs packaging. Our findings support a packaging mechanism in which translation, dimerization, NC binding, and packaging are regulated by a common RNA structural switch.〈br /〉〈br /〉〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3335204/" target="_blank"〉〈img src="https://static.pubmed.gov/portal/portal3rc.fcgi/4089621/img/3977009" border="0"〉〈/a〉   〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3335204/" target="_blank"〉This paper as free author manuscript - peer-reviewed and accepted for publication〈/a〉〈br /〉〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Lu, Kun -- Heng, Xiao -- Garyu, Lianko -- Monti, Sarah -- Garcia, Eric L -- Kharytonchyk, Siarhei -- Dorjsuren, Bilguujin -- Kulandaivel, Gowry -- Jones, Simonne -- Hiremath, Atheeth -- Divakaruni, Sai Sachin -- LaCotti, Courtney -- Barton, Shawn -- Tummillo, Daniel -- Hosic, Azra -- Edme, Kedy -- Albrecht, Sara -- Telesnitsky, Alice -- Summers, Michael F -- 2T34 GM008663/GM/NIGMS NIH HHS/ -- R01 GM042561/GM/NIGMS NIH HHS/ -- R01 GM042561-21/GM/NIGMS NIH HHS/ -- R01 GM042561-22/GM/NIGMS NIH HHS/ -- R01 GM042561-23/GM/NIGMS NIH HHS/ -- R01 GM042561-24/GM/NIGMS NIH HHS/ -- R01 GM42561/GM/NIGMS NIH HHS/ -- R25 GM055036/GM/NIGMS NIH HHS/ -- R25 GM055036-14/GM/NIGMS NIH HHS/ -- R25 GM055036-15/GM/NIGMS NIH HHS/ -- R25 GM055036-16/GM/NIGMS NIH HHS/ -- R25 GM55036/GM/NIGMS NIH HHS/ -- T34 GM008663/GM/NIGMS NIH HHS/ -- Howard Hughes Medical Institute/ -- New York, N.Y. -- Science. 2011 Oct 14;334(6053):242-5. doi: 10.1126/science.1210460.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉Howard Hughes Medical Institute (HHMI) and Department of Chemistry and Biochemistry, University of Maryland Baltimore County (UMBC), Baltimore, MD 21250, USA.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/21998393" target="_blank"〉PubMed〈/a〉

Description: Multiple myeloma is an incurable malignancy of plasma cells, and its pathogenesis is poorly understood. Here we report the massively parallel sequencing of 38 tumour genomes and their comparison to matched normal DNAs. Several new and unexpected oncogenic mechanisms were suggested by the pattern of somatic mutation across the data set. These include the mutation of genes involved in protein translation (seen in nearly half of the patients), genes involved in histone methylation, and genes involved in blood coagulation. In addition, a broader than anticipated role of NF-kappaB signalling was indicated by mutations in 11 members of the NF-kappaB pathway. Of potential immediate clinical relevance, activating mutations of the kinase BRAF were observed in 4% of patients, suggesting the evaluation of BRAF inhibitors in multiple myeloma clinical trials. These results indicate that cancer genome sequencing of large collections of samples will yield new insights into cancer not anticipated by existing knowledge.〈br /〉〈br /〉〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3560292/" target="_blank"〉〈img src="https://static.pubmed.gov/portal/portal3rc.fcgi/4089621/img/3977009" border="0"〉〈/a〉   〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3560292/" target="_blank"〉This paper as free author manuscript - peer-reviewed and accepted for publication〈/a〉〈br /〉〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Chapman, Michael A -- Lawrence, Michael S -- Keats, Jonathan J -- Cibulskis, Kristian -- Sougnez, Carrie -- Schinzel, Anna C -- Harview, Christina L -- Brunet, Jean-Philippe -- Ahmann, Gregory J -- Adli, Mazhar -- Anderson, Kenneth C -- Ardlie, Kristin G -- Auclair, Daniel -- Baker, Angela -- Bergsagel, P Leif -- Bernstein, Bradley E -- Drier, Yotam -- Fonseca, Rafael -- Gabriel, Stacey B -- Hofmeister, Craig C -- Jagannath, Sundar -- Jakubowiak, Andrzej J -- Krishnan, Amrita -- Levy, Joan -- Liefeld, Ted -- Lonial, Sagar -- Mahan, Scott -- Mfuko, Bunmi -- Monti, Stefano -- Perkins, Louise M -- Onofrio, Robb -- Pugh, Trevor J -- Rajkumar, S Vincent -- Ramos, Alex H -- Siegel, David S -- Sivachenko, Andrey -- Stewart, A Keith -- Trudel, Suzanne -- Vij, Ravi -- Voet, Douglas -- Winckler, Wendy -- Zimmerman, Todd -- Carpten, John -- Trent, Jeff -- Hahn, William C -- Garraway, Levi A -- Meyerson, Matthew -- Lander, Eric S -- Getz, Gad -- Golub, Todd R -- K12 CA133250/CA/NCI NIH HHS/ -- R01 AG020686/AG/NIA NIH HHS/ -- R01 AG020686-07/AG/NIA NIH HHS/ -- R01 CA133115/CA/NCI NIH HHS/ -- R01 CA133115-04/CA/NCI NIH HHS/ -- R01 CA133966/CA/NCI NIH HHS/ -- R01 CA133966-03/CA/NCI NIH HHS/ -- England -- Nature. 2011 Mar 24;471(7339):467-72. doi: 10.1038/nature09837.〈br /〉〈span class="detail_caption"〉Author address: 〈/span〉The Eli and Edythe L. Description: In recent years, the role played by the stromal microenvironment has been given growing attention in order to achieve a full understanding of cancer initiation and progression. Because cancer is a tissue-based disease, the integrity of tissue architecture is a major constraint toward cancer growth. Indeed, a large contribution of...

Description: The Tuamotu Archipelago in French Polynesia is a Co-rich ferromanganese crust province. The NODCO I survey (1986) provided detailed data on Co-rich crusts in this environment through the exploration of a restricted zone in the vicinity of Niau Island on the southern flank of the archipelago. This flat zone is a fossil atoll which, under the action of subsidence and tectonic movements, has collapsed to a water depth of 1000 m. The plateau is partially filled with coralline sediments. Outcrops of ferromanganese crusts, associated with rare nodules and slabs, are located on the inner side of the coral reef which bounds the ancient lagoon. The successive episodes of plateau history have been recorded in the different growth periods of the ferromanganese crusts. The crusts, nodules and slabs belong to the same morphological, mineralogical and geochemical family. Cobalt contents vary from 0.7 to 1.3%. The highest values belong to the thinnest ferromanganese crusts which are located on the flanks of the plateau. Average Ni contents are about 0.5% and Cu contents about 0.1%; Pt contents vary from 0.2 to 1.3 ppm. Platinum and Co are enriched in the outermost oxide zone of the crusts. Poorly crystallized -MnO2 is the dominant mineralogical phase. Cobalt enrichment seems to be related to -MnO2 particle size. The greatest contents are located in the finest material where the particle size is less than 0.1 m. Cobalt-rich crusts of the Niau Zone have the same characteristics as the Co-rich crusts from the Equatorial North Pacific. They differ in original setting: the reefal environment in the Niau Zone is superficial, overlying a volcanic substrate.

Description: This work is about the experimentation of a new methodology using a proton magnetometer to find artificial sources (as a wrecked ship) at any depht with the highest resolution. Such methodology, called Nottom Reduction (BTM) (Faggioni et al.), adopts a "high definition" procedure in order to strongly decrease the indetermination of the point of measure. The BTM method includes three steps: high definition geomagnetism, temporal correction and bottom reduction. The signals sampled in space (x,y,z) and time (t), are subsequently analyzed in frequency fields: f(s,t)---f(s,w)

Description: The marine geomagnetism is often used to solve geological problems at a regional scale: structural, volcanological or mining problems, for instance (Faggioni et al., 1995). In recent time, the development of the “high definition (HD) metrology” (Faggioni et al., 2001) has made the marine geomagnetic method suitable for detecting short wavelength and low amplitude geomagnetic anomalies. This type of signal may be often related to environmental and/or archaeological sources.......

Description: Recently two marine magnetic surveys, combined with the use of a multi-beam sonar (Kongsberg Marittime EM 300 multibeam: 30 KHz frequency echosounder for hydrographic purposes; acoustic lobe composed of 128 beams able to cover a 150° sector) a side-scan sonar (Simrad MS 992 dual-frequency sidescan sonar with echo sounder transducers 150 Hz and 330 KHz) and a Remote Operated Vehicle (ROV – a mobile tools used in environments which are too dangerous for humans), were executed in two sites respectively in the Ligurian Sea and the Asinara Gulf. The aim of these investigations was to test modern instrumentations and set new working procedures for searching underwater cultural resources. The collected and processed magnetic data yielded very satisfactory results: we detected submerged and buried features of cultural interest at both sites, at depths of 40 m and 400 m respectively.
