    Description: To pursue a systematic approach to the discovery of functional connections among diseases, genetic perturbation, and drug action, we have created the first installment of a reference collection of gene-expression profiles from cultured human cells treated with bioactive small molecules, together with pattern-matching software to mine these data. We demonstrate that this "Connectivity Map" resource can be used to find connections among small molecules sharing a mechanism of action, chemicals and physiological processes, and diseases and drugs.     Description: Multiple myeloma is an incurable malignancy of plasma cells, and its pathogenesis is poorly understood. Here we report the massively parallel sequencing of 38 tumour genomes and their comparison to matched normal DNAs. Several new and unexpected oncogenic mechanisms were suggested by the pattern of somatic mutation across the data set. These include the mutation of genes involved in protein translation (seen in nearly half of the patients), genes involved in histone methylation, and genes involved in blood coagulation. In addition, a broader than anticipated role of NF-kappaB signalling was indicated by mutations in 11 members of the NF-kappaB pathway. Of potential immediate clinical relevance, activating mutations of the kinase BRAF were observed in 4% of patients, suggesting the evaluation of BRAF inhibitors in multiple myeloma clinical trials.     Notes: Summary The following paper is the second part of a study devoted to the only preserved work of Hipparchus, his Commentary to Aratus. The first part, published in 1984, was based on the hypothesis that Hipparchus wrote his treatise with the help of a mobile sphere and led us to the determination of some features of that instrument. The coordinates of 78 stars, which are given below, have been obtained on the basis of the same hypothesis and using results already published. The stars selected are those for which the text connects numerical data to at least two events out of rising, setting or culmination. An analysis of the positions shows that the stars have been located on the sphere in an equatorial frame slightly different from the rotation axis frame. Statistical data processing shows that the stars were plotted on the sphere as circles rather than as points: the celestial coordinates of the stars are then those of the center of the disc, whose radius was determined. The position of a few stars could be evaluated thanks to the determination of an approximate value of the cubit used by Hipparchus. Lastly, the time of the observations was confirmed to be −140±25 years. The study confirms the basic hypothesis and throws light on the part played by the mobile sphere when the heavens are being described. It gives a better idea of the difficulties of constructing the instrument and of the ways of using it; a more detailed model of Hipparchus' mobile sphere will allow a more thorough investigation of all stars mentioned in the Commentary.

    Notes: Résumé L'étude des données écliptiques contenues dans le Commentaire d'Hipparque aux Phénomènes d'Eudoxe et d'Aratos permet de décrire la sphère mobile qui a pu aider Hipparque dans la rédaction de son texte. Le traitement statistique des 165 couples de mesures de longitudes écliptiques, associé à l'utilisation d'une méthode par autocorrélation, conduit à déterminer une fonction d'appareil qui est négligeable, ce qui montre la perfection de l'instrument employé par Hipparque. La présente étude montre qu'Hipparque n'a pas tenu compte, dans le relevé des longitudes au méridien, de l'épaisseur de l'armille. Il a procédé à un arrondi scientifique au demi-degré pour la plupart des lectures, qu'elles soient faites d'un côté ou de l'autre du méridien. La valeur trouvée pour la latitude affichée sur l'instrument correspond parfaitement à celle du nord de l'île de Rhodes et l'obliquité de l'écliptique à celle qu'elle avait au 2ème siècle avant notre ère. Ce dernier résultat laisse penser qu'Hipparque aurait déterminé avec exactitude l'obliquité de l'écliptique mais que la valeur obtenue ne nous aurait pas été transmise par la tradition écrite.
