    Description: Europe faces a historic opportunity. We need to reimagine European unity as a collective endeavour, as the only way in which we can turn threats to our way of life into the possibility to create a better life for all. This report focuses on three core areas where EU-wide political ambition and investment are truly necessary: Rapid technological change amid increasing global competition, with digital technologies set to have dramatic impacts across all sectors of the economy. Europe needs to become more competitive through innovation and the fast absorption of new technologies if it is to reap the benefits of this wave of change. Increasing breakdown of the global climate and other ecosystems due to our economic system, with action on mitigation – and to adapt to already inevitable changes – now incredibly urgent. In part, this is because of the time lags built into natural systems. In part, it is because of the unavoidable path-dependency and pace of our own response. Growing threats to social cohesion and social sustainability from multiple trends, including how we manage technological change and automation, the climate transition, and societal ageing. This is happening against a background of stalling convergence across Europe, rising income inequality over several decades, rising burdens on households and persistent inequalities of opportunity that are also a drag on economic performance. Competitiveness, sustainability and inclusion have to be addressed together, holistically. This is why this report addresses all three with a focus on the complex interlinkages between them. The vital role of far-sighted investment is a key theme that emerges from this investigation. The critical role of European-level collaboration is another. Instead of merely adapting to change, we need to be proactive through timely reform and investment that sets us on a pathway towards a society that is productive, sustainable and inclusive, by design.

    Notes: Abstract Paracoccus halodenitrificans, grown anaerobically in the presence of nitrite, contained membrane and cytoplasmic nitrite reductases. When assayed in the presence of phenazine methosulfate and ascorbate, the membranebound enzyme produced nitrous oxide whereas the cytoplasmic enzyme produced nitric oxide. When both enzymes were assayed in the presence of methyl viologen and dithionite, the cytoplasmic enzyme produced ammonia. Following solubilization, the membrane-bound enzyme behaved like the cytoplasmic enzyme, producing nitric oxide in the presence of phenazine methosulfate and ascorbate, and ammonia when assayed in the presence of methyl viologen and dithionite. The cytoplasmic and membranebound enzymes were purified to essentially the same specific activity. Only a single nitrite-reductase activity was detected on electrophoretic gels and the electrophoretic behavior of both enzymes suggested they were identical. The spectral properties of both enzymes suggested they were cd-type cytochromes. These data suggest that the products of nitrite reduction by the cd-cytochrome nitrite reductase are determined by the location of the enzyme and the redox potential of the electron donor.

    Notes: Abstract The ability of Halobacterium vallismortis, Halobacterium mediterranei and Halobacterium marismortui (Ginzburg strain) to grow anaerobically and denitrify was determined. Each organism grew anaerobically only in the presence of nitrate. H. marismortui produced nitrite and dinitrogen from nitrate during exponential growth. However, as the culture entered stationary phase, dinitrogen production ceased and nitrous oxide was detected. H. vallismortis produced nitrous oxide and dinitrogen during exponential growth, with dinitrogen production ceasing at the onset of stationary phase. H. mediterranei produced dinitrogen during exponential growth and did not produce nitrous oxide. These results confirm the occurrence of denitrification in the halobacteria.

    Notes: Abstract Based upon arguments concerning properties of the environment and the energetics of nitrogen transformation reactions, new hypotheses regarding their evolution are presented. These hypotheses are supported by new calculations and observations germane to understanding the evolution of the nitrogen cycle. From calculations of shock production by meteor impact, we suggest that impact produced fixed nitrogen could have resulted in the entire reservoir of Earth's N2 being converted into fixed nitrogen at the end of accretion. We have significantly improved upon previous calculations of the abiotic fixation rate on the early earth and find a rate of fixation by lightning of ∼1−3 × 1016 Molecules NO/J, which is 2 to 3 times greater than previous estimates. This strengthens the suggestion, corroborated by the predominance of a single nitrogenase enzyme, that biological nitrogen fixation may have been a late evolutionary development, after the development of an aerobic atmosphere. In addition, we show for the first time that HNO, predicted to be the main product of atmospheric photochemical reactions involving NO on the primitive Earth by photochemical models, would eventually become NO2 − and NO3 − after reaching the Earth's surface. Based upon microbe-environment interactions on an ecological as well as a biochemical scale we suggest that denitrification arose prior to aerobic respiration and that nitrification arose after the advent of an aerobic atmosphere. We hypothesize the following evolutionary sequence for the biological transformation of nitrogen compounds: Ammonification → Denitrification → Nitrification → Nitrogen fixation.

    Notes: Abstract We studied the genome of an antarctic ice fish, Chionodraco hamatus, in order to detect highly repetitive DNAs that may play a role in heterochromatinization processes and sex chromosome differentiation. We used two different experimental approaches. Hybridization of a Bkm probe to genomic DNA showed slight differences between the two sexes. Using restriction enzymes, a Bgl II satellite (pIF) was isolated. In situ hybridization revealed a preferential localization of pIF on the centromeres and the telomeres of most chromosomes, as well as an interstitial band on the long arms of the neo-Y sex chromosome, where probably the hypothetical fusion took place. Dot-blot experiments showed that pIF is still present in species belonging to different families of the same suborder. Though preliminary, our results suggest a conservative nature of this DNA which might have played a definite functional role in the genome of these polar fishes.

    Notes: Abstract The nuclear DNA content of 23 selachian species (10 Batoidea, 11 Galeomorphii, and 2 Squalomorphii) was histophotometrically studied. Their genome sizes range from 7.5 pg/N in Raja fillae (Batoidea) to 34.1 pg/N in Oxynotus centrina (Squalomorphii). Results show slight differences in the pattern of quantitative variations between the superorders Batoidea and Galeomorphii; Squalomorphii preserve their peculiar wide interspecific variability at the intrafamilial level, with values sited between 13.1 and 34.1 pg/N. In 21 species also the DNA base composition was determined by means of DAPI. The study shows that in the species examined the DAPI positive fraction varies from a minimum of 27.7% in Oxynotus centrina, which possesses the largest genome size among all the Selachians studied, to a maximum of 72.5% in Carcharhinus limbatus. As a whole the data show an inverse correlation between the DNA content and the DAPI positive fraction, a condition common to all cold-blooded vertebrates. The low percentage of DAPI positive DNA found in Oxynotus centrina could be attributable to a lower stainability by the fluorochrome caused by a higher chromatin condensation in the erythrocytes. The validity of the DAPI method was verified by comparison with the biochemical assay according to the thermal denaturation method in 6 selachian species.
