    Notes: Abstract      Intact cells of the unicellular cyanobacterium Synechococcus UTEX 625 degraded exogenously supplied cyanate (as KOCN) to CO2 and NH3 in a light-dependent reaction. NH3 release to the medium was as high as 80 μmol(mgChl)–1h–1 and increased 1.7-fold in the presence of methionine sulfoximine, a glutamine synthetase inhibitor. Cyanate also supported photosynthetic O2 evolution to a maximum rate of 188 μmol O2(mgChl)–1h–1 at pH 8 and 30° C. Cyanate decomposition in cell-free extracts, measured by mass spectrometry as 13CO2 production from KO13CN, occurred in the soluble enzyme fraction, but not in the thylakoid/carboxysome fraction, and was enhanced by HCO3 – and inhibited by the dianion oxalate. CO2, rather than HCO3 –, was a product of cyanate decomposition. The ability to decompose cyanate was not dependent upon pre-exposure of cells to cyanate to induce activity. The collective results indicate that Synechococcus UTEX 625 possesses a constitutive, cytosolic cyanase (EC 4.3.99.1), similar in mechanism to that found in some species of heterotrophic bacteria. The reaction catalyzed was: OCN– + HCO3 – + 2 H+→ 2 CO2 + NH3. In intact cells, the CO2 produced by the action of cyanase on OCN– was either directly fixed by the Calvin cycle enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase, leading to O2 evolution, or leaked into the medium where it was returned to the cell by the active CO2/HCO3 – transport systems for fixation. However, leakage of CO2 from air-grown cells was only observed when the active CO2 transport system was inhibited by darkness or the CO2 analogue carbon oxysulfide.

    Notes: Abstract— The multiple specimen technique was developed to measure the crack growth fracture resistance of a through axial crack in a pressurised pipe and the results compared with data measured from test specimens. The comparison indicates that there is no significant difference between pipe and specimen behaviour. The results are also compared with elastic-plastic three-dimensional finite element analyses of the pipe and the R6 failure assessment curve. Reasonably good agreement was found between the experimental results and finite element analyses. The experimental results all lie outside the material specific failure assessment curve of the R6 structural integrity assessment procedure.

    Notes: Summary The mechanisms by which crystal size distributions affect the usual method of quantitative X-ray diffraction analysis of bone mineral have been determined on synthetic crystals. It was observed that each component of a crystal-size distribution diffracts independently. This independence causes systematic nonlinear behavior in the plot of integrated intensity vs. broadening parameter curves. The nonlinearity resulted in an overestimation of the amount of nondiffracting material present in bone mineral. Because crystal size distributions may vary for different crystallographic directions, it is strongly suggested that the usual practice of adding thec-axis anda-axis integrated intensities to estimate the crystallinity of the sample be discontinued. Methods of understanding the crystal size distribution function in bone mineral are discussed and evaluated.

    Notes: Summary The amount of 14C-glycolate excreted by Oscillatoria sp. and Anabaena flos-aquae is less than 1% of the 14C fixed by the algae during photosynthesis. Transfer of cells grown on 5% CO2 in air to a medium of low bicarbonate concentration or treatment of the cells with isonicotinyl hydrazide (INH) during photosynthesis, caused little increase in glycolate excretion. α-Hydroxysulfonates failed to stimulate massive excretion of glycolate. Although these blue-green algae excreted little glycolate, a significant proportion of the photosynthetically fixed carbon was excreted in the form of basic, neutral and acidic compounds, and such excretion was greater in 5% CO2-grown cells than in air-grown cells.

Pour déterminer le COD, on a procédé à une extension de la solution appliquéc à une coque mince pour obtenir le facteur d'intensité d'entaille selon Folias, et les équations obtenues ont été résolues par voie numérique, en suivant une procédure due à Erdogan. Les résultats peuvent être utilisés à la prédiction d'une rupture ainsi qu'au calcul de la vitesse de fuite d'un gaz contenu dans un récipient.
    Notes: Abstract The crack opening displacement at the centre of a surface crack in a pressurized sphere has been calculated using a strip yielding model. The shallow shell solution for the stress intensity factor derived by Folias has been extended to derive the COD, and the resulting equations have been solved numerically by a procedure due to Erdogan. The results are of relevance to fracture predictions and gas leakage rate calculations.

    Notes: This study was undertaken to investigate the possibility of measuring total body water in human subjects to better than ±0.5%. Accurate serial estimates of total body water were required to complement densitometric and anthropometric measurements used to monitor body compositional changes in obese patients undergoing dietary or surgical weight reduction therapy. The method required the oral administration of 1-2 g of deuterium oxide and the analysis of pre-dose and respective equilibrated samples of urine, plasma or saliva. The sample size required for analysis was 5 μl and the conversion of gaseous phase was accomplished using a uranium reduction furnace. Isotopic erichment of samples was measured using a mass spectrometer incorporating several features designed to cope with problems inherent in H2/H2H isootopic analysis. Reproducibility of sample preparation and accuracy of the mass spectrometer were tested using international standards and shown to give an overall sensitivity of 2 parts in 107 for the determination of deuterium in H2O/H2HO mixtures. This precision has enabled us to demonstrate that isotopic fractionation of deuterium with respect to hydrogen occurs within the body and expands the potential use of this isotope for quantitative biochemical studies in the human subject.
