    Notes: Two sites for the β-oxidation of fatty acids in avocado (Persea americana L.) mesocarp exist. One site is the microbody, the other the mitochondrion. It is apparent that the mitochondrial membrane barrier, which remains intact after sucrose density gradient centrifugation, prevents rapid access of acyl CoA substrates to matrix β-oxidation sites. Thus, intact mitochondria showed little β-oxidation enzyme activity. Rupturing of the mitochondrial membrane allowed rapid access of the acyl CoA substrates to matrix sites. Consequently, in ruptured mitochondria, high O2-oxidation enzyme activities were measured. O2 uptake studies further distinguished the two organellar sites of β-oxidation. During palmitoyl CoA oxidation, O2 uptake was reduced by catalase and increased by KCN in the microbodies, whilst mitochondrial O2 uptake was unaffected by catalase and reduced by KCN. This reflected the differing fates of FADH2, produced during the first β-oxidation step, in the two organelles. In addition, only the mitochondrial β-oxidation of fatty acids was carnitine-dependent.

H14CO3 was not incorporated into fatty acids by isolated pea leaf chloroplasts, which, therefore, do not possess a self-contained pathway for the synthesis of fatty acids from early intermediates of the Calvin cycle. Citrate, pyruvate, acetate and L-acetylcarnitine were all shown to act as sources of acetyl groups for fatty acid synthesis by pea leaf chloroplasts. L-acetylcarnitine was the best substrate, being incorporated into fatty acids at rates that were at least five-fold higher than those achieved with the other substrates. Citrate was incorporated into fatty acids at the lowest rate, followed by pyruvate, with acetate being incorporated at the second highest rate of all. When the isolated chloroplasts were ruptured, an inhibition of L-acetylcarnitine incorporation into fatty acids was noted, whilst acetate incorporation remained unaffected. L-acetylcarnitine also increased the ratio of monoenoic: saturated fatty acids synthesized, compared with a 1:1 ratio observed when citrate, pyruvate and acetate were supplied as substrates. It is suggested that L-carnitine and carnitine acyltransferases play a central role in plant acyl CoA metabolism by facilitating the transfer of activated acyl groups across membranes (acyl CoA barriers).

    Notes: Abstract. Whilst L-acetylcarnitine acted as a substrate for fatty acid synthesis by isolated pea leaf chloroplasts, D-acetylcarnitine did not. This result, together with those obtained using the inhibitors D-carnitine and deoxycarnitine, indicated that L-acetylcarnitine was not being hydrolysed to free acetate prior to incorporation into chloroplast fatty acids. Seventy-five per cent and 66% inhibitions of L-acetylcarnitine incorporation into fatty acids, brought about by adding equimolar quantities of D-carnitine and deoxycarnitine, respectively, were suggestive of competitive inhibition at two points: an integral membrane translocator in the chloroplast envelope: and the carnitine acetyltransferase enzyme of the chloroplast stroma, which converts L-acetylcarnitine to acetyl CoA. Isotope competition experiments between acetate and L-acetylcarnitine confirmed that L-acetylcarnitine was the preferred substrate for pea chloroplast fatty acid synthesis.

    Description / Table of Contents: Zusammenfassung In drei Versuchen wurde beim Weißklee (Trifolium repens) die Zeit bis zur Knöllchenbildung sowie das Gewicht der Pflanze bzw. deren Entwicklungsgeschwindigkeit ermittelt. Bei Versuch 1 und 2 erfolgte die Kultur in Reagenzgläsern auf künstlichem Nährboden nach kontrollierter Impfung und bei Versuch 3 im Boden ohne Impfung. Zunächst wurde im Versuch 1 die Variabilität der Zeitdauer bis zur Knöllchenbildung nach Impfung mit Rhizobium trifolii untersucht, und es zeigte sich eine auffällige Beziehung zwischen der Entwicklungsgeschwindigkeit der Knollen und der Pflanze. Den wesentlichsten Teil der Ergebnisse lieferte Versuch 2, in dem die Nachkommenschaften dialleler Kreuzungen von 12 in Versuch 1 selektierten Pflanzen hinsichtlich der Kriterien Anzahl Tage bis zur Knöllchenbildung und Pflanzengewicht nach 80 Tagen geprüft wurden. Die Resultate ergaben ein recht kompliziertes Bild von der genetischen Veranlagung der beiden untersuchten Merkmale. Es wurden reziproke Unterschiede (sowohl allgemein wie spezifisch) sowie additive und nicht-additive Wirkungen festgestellt. Schließlich wurden im Versuch 3 die Nachkommenschaften aus 22 diallelen Kreuzungen in Erde ohne Impfung geprüft und die Ergebnisse mit denen aus Versuch 2 verglichen. Es wurde festgestellt, daß sie nicht übereinstimmten.
    Notes: Summary The results of experiments with white clover (Trifolium repens) in which time of nodulation and seedling plant weight or vigour were measured are reported. Experiments 1 and 2 were conducted in artificial growth medium in test tubes with controlled inoculation and experiment 3 in soil without controlled inoculation. Experiment 1 which was preliminary in nature showed the extent of the variation for time of nodulation after inoculation with Rhizobium trifolii. It was evident also that plant vigour and the number of days to nodulation were negatively correlated. Experiment 2 forms the major part of the results and is concerned with the analyses and interpretation of the diallel cross progeny of twelve plants selected from experiment 1. The results indicated a rather complex genetic picture for the two characters measured, namely days to nodulation and seedling plant weight at 80 days. Reciprocal (both general and specific) as well as additive (g.c.a.) and non-additive (s.c.a.) effects were present. Experiment 3, in which seed from 22 families of the diallel cross was sown in soil without controlled inoculation, indicated that the results obtained under the laboratory conditions of experiment 2 and those obtained in soil conditions were not correlated. The implications of these results in relation to selection of improved varieties of the host species are discussed.

    Notes: Abstract Mitochondria from pea (Pisum sativum L.) seeds were separated into two fractions, mitoplasts (intact inner membrane) and the outer-membrane fraction. The mitoplasts only oxidised palmitate in the presence of carnitine and added outermembrane fraction. Mitoplasts were able to oxidise palmitoylCoA in the presence of carnitine and added outer-membrane fraction had no effect on this oxidation. It was concluded that a long-chain acylCoA synthetase (EC 6.2.1.3) was located on the outer membrane and that the activity of this enzyme in assays was more than sufficient to account for any observed rate of O2 uptake during palmitate oxidation by pea mitochondria. The location of carnitine long-chain acyltransferase (carnitine palmitoyl transferase EC 2.3.1.21) would appear to be the mitoplast i.e. the inner mitochondrial membrane, and confirms the previous work at Newcastle.
