    Notes: Abstract Staphylococcus saprophyticus was shown to be agglutinated by wheat germ agglutinin, wheat germ agglutinin-biotin and bovine serum albumin-p-aninophenyl-N-acetyl-β-d-glucosaminide (GlcNAc-BSA), and sheep red blood cells. In these agglutinations, filamentous or amorphous structures radiating from the surface of S. saprophyticus were demonstrated by electron microscope observation. Cytochemical analyses of the agglutination revealed the binding sites of wheat germ agglutinin in S. saprophyticus and the binding sites of GlcNAc in the sheep red blood cells and S. saprophyticus. Since GlcNAc-BSA contains N-acetylglucosamine to which wheat germ agglutinin can bind, it is most likely that an interaction between a wheat germ agglutinin-bindable substance in S. saprophyticus and an N-acetylglucosamine-bindable substance in sheep red blood cells is involved in the agglutination.

    Notes: The polyene antibiotic, filipin, was used as the probe for demonstrating sterols in the freeze-fractured plasma- and cytomembranes of Pneumocystis carinii. The distribution of filipin-sterol complexes was homogeneous on the plasma membrane throughout all developmental stages from trophozoite to cyst; however, the density of the complexes gradually decreased with the progress of development. In the trophozoite, the density of the complexes was 485 ± 42/μm2 on the P face and 341 ± 27/μm2 on the E face. It was 249 ± 50 on the P face and 132 ± 48 on the E face in the precyst and 138 ± 24 and 59 ± 20, respectively, in the cyst. The membranes of nucleus, mitochondria, and small round bodies showed more or fewer complexes while no complexes were found in the membranes of one endoplasmic reticulum. In nuclear and mitochondrial membranes, some small scattered clusters of complexes were observed. Two types of vacuoles were distinguished: one having many complexes in its membrane and the other having none at all.

    Notes: The ultrastructure of Trypanosoma brucei gambiense was investigated by the freeze-fracture method. Three different regions of the continuous plasma membrane; cell body proper, flagellar pocket, and flagellum were compared in density and distribution of the intramembranous particles (IMP's). The IMP-density was highest in the flagellar pocket membrane and lowest in flagellum. Intra membranous particles of the cell body membrane were distributed uniformly on both the protoplasmic (P) and exoplasmic (E) faces. On the P face of the flagellar membrane, a single row of IMP-clusters was seen along the juncture of the flagllum to the cell body. Since the spacing of the IMP-clusters was almost equal to the spacing of the paired rivet structures observed in thin section, these clusters likely are related to the junction of flagellum and cell body. At the neck of the flagellar pocket, several linear arrays of IMP's were found on the P face of the flagellar membrane, while on the E face rows of depressions were seen. At the flagellar base, the clusters of IMP's were only seen on the P face. On the flagellar pocket membrane, particle-rich depressions and linear particle arrays were also found on the P face, while on the E face such special particle arrangements were not recognized. These particle-rich depressions may correspond to the sites of pinocytosis of the bloodstream forms which have been demonstrated in thin sections.

    Notes: Abstract Ultrastructure of the trophozoite of Pneumocystis carinii was studied by the freeze-fracture technique. Nuclei and cytoplasmic organelles such as the endoplasmic reticulum, mitochondria, cytoplasmic vacuoles and small round bodies were observed. The mean number of nuclear pores was 8 per Μm2, which is small compared with that reported for other human pathogenic protozoa. In general, the density of nuclear pores is considered to be related to the metabolic activity of the nucleus. This result, therefore, suggests that the nucleus of P. carinii may be less metabolically active than those of other protozoa thus far examined. Both the nuclear envelope and the endoplasmic reticulum showed a similar distribution of intramembraneous particles (IMPs): the P face was heterogeneous and the E face was homogeneous. However, the outer membrane of mitochondria was somewhat heterogeneous in IMP distribution on both P and E faces. The cytoplasmic vacuoles always showed a lower IMP density than that of the plasma membrane. This indicates that the vacuoles of P. carinii would not be phagosomes. By means of this technique, the tubular expansions could be divided morphologically into four types: tubules, lobopodia, branching and beaded structures. Furthermore, it was noted that the daughter trophozoite in the endogenous form was not different from the usual free trophozoites in the IMP distribution pattern.

    Notes: The ectethmoid-mandibular articulation in Melithreptus and Manorina (Meliphagidae: Aves) consists of the dorsal mandibular process fitting into and abutting against the ventral ectethmoid fossa; it forms a brace for the mandible. This articulation in Melithreptus is a typical diarthrosis with long folded capsular walls. The mandible, thus, has two separate articulations, each with a different axis of rotation. No other genus of Meliphagidae (except Ptiloprora) or any other avian family possesses a similar feature.The jaw and tongue musculature of Melithreptus are described. The two muscles opening the jaws are well developed, while those closing the jaws are small. The tongue muscles show no special developments.A large maxillary gland, presumably muscus secreting, covers the ventral surface of the jaw muscles. Its duct opens into the oral cavity just behind the tip of the upper jaw. The frilled tip of the tongue rests against the duct opening.The ectethmoid-mandibular articulation braces the adducted mandible against dorsoposteriorly directed forces. The mandible can be held closed without a compression force exerted by the mandible on the quadrate, permitting the bird to raise its upper jaw with greater ease and less loss of force. The tongue can be protruded through the slight gap between the jaws, moving against the duct opening and thus be coated with mucus. Presumably, these birds capture insects with their sticky tongue. Hence, the ectethmoid-mandibular articulation is an adaptation for this feeding method; it evolved independently in three genera of the Meliphagidae.The ectethmoid-mandibular articulation demonstrates that a bone can have two articulations with different axes of rotation, that the two articular halves can separate widely, and that articular cartilages can be flat and remain in contact over a large area. Its function suggests that the basitemporal articulation of the mandible found in many other birds has a similar function. And it demonstrates that in the evolution of the mammalian dentary-squamosal articulation, the new hinge did not have to lie on the same rotational axis as the existing quadrate-articular hinge.

    Description: The source location method using high-frequency seismic amplitudes based on the assumption of isotropic radiation of S waves has been used successfully to locate seismic events and tremor at volcanoes. This amplitude source location (ASL) method overcomes the limitations of traditional hypocenter determination methods that use onset arrival times, and has great potential to improve volcano-seismic monitoring and the investigation of source processes of volcano-seismic signals. However, theoretical justification of the basic assumption used in the ASL method has not been provided in previous studies. In this study, we tested the ASL method by using seismic waveforms simulated with a finite-difference method in realistic heterogeneous volcanic structures with intrinsic attenuation and topography. Our results showed that ASL determinations were successful when using waveforms characterized by multiple scattering that can be approximated by the diffusion model. We found that the energy solution of the diffusion model provided an amplitude-distance relation that is similar to that used in the ASL method. Our results suggest that the ASL method is applicable to high-frequency seismograms in highly heterogeneous media with transport mean free paths of 10 3 m or smaller and strong intrinsic attenuation. These medium parameters are consistent with those estimated at various volcanoes. Our study validates the basic assumption of the ASL method and justifies its use to locate the sources of high-frequency seismic signals observed at volcanoes.
