    Notes: Transgenic rice plants have been regenerated from kanamycin-resistant callus of Oryza sativa (cv. Taipei 309) derived from protoplasts electroporated with pCaMVNEO carrying the neomycin phosphotransferase II (nptII) gene. Of 6 randomly selected plants, all contained the nptll gene, but only 2 plants expressed NPTII activity. The transgenic plants were significantly shorter, produced fewer tillers, took longer to flower and had reduced fertility compared to non-transformed protoplastderived plants. Fifty-six seeds collected from one transgenic plant expressing NPTII activity germinated on medium containing kanamycin sulphate to give 16 green, first seed generation (R1) plants. The latter could be divided into 3 groups: (i) Plants which set seed, had normal floret morphology and produced a total of 76 seeds; (ii) Plants which flowered, but which failed to set seed; (iii) Plants which failed to flower, were shorter and had significantly fewer tillers than plants of groups (i) and (ii). The nptII gene was present in all transgenic R1 plants, but only 8 plants expressed the gene. Phenotypic characteristics, observed in transgenic R1 plants were also seen in the transforned R2 plants. These included reduced stature, a longer vegetative phase and reduced fertility compared to non-transformed plants.

    Notes: Summary An investigation was undertaken on the storage characteristics of pollen collected from two English rose cultivars. A rapid decline in viability was observed in pollen stored at +4° C and −20° C, whereas the viability of pollen, stored at ultra-low temperature (−196° C), remained constant. Cryopreserved pollen was shown to retain its ability for fertilisation. The effects of the stage of flower development and anther dehiscence were assessed on both pre-and post-cryopreservation viabilities. Successful long-term storage of pollen will facilitate hybridisation of rose species and cultivars that do not flower synchronously.

    Notes: Summary The phenotypes of seed progeny (R2 generation) of Oryza sativa L. cv. Taipei 309, which carried the neomycin phosphotransferase II (npt II) gene, were compared with those of non-transformed, protoplast-derived plants of the same generation and non-transformed, seed-derived plants under field and glasshouse conditions. Under both conditions the transgenic plants were generally smaller, took longer to flower and had reduced fertility. Significant differences were observed between individuals within the group of transgenic plants. The npt II gene was present in most of the transgenic plants, but NPT II activity was only detected in a minority of individuals.

    Notes: Abstract Shoot tips of in vitro grown plantlets of Rosa multiflora were cryopreserved using an encapsulation/dehydration procedure. The influence of sucrose and silica gel pretreatments on pre- and post-freeze shoot growth were examined. Shoot tips recovered from liquid nitrogen only grew after 24h pretreatment in medium containing 0.5 M sucrose, followed by 2 h drying with silica gel and rapid freezing.

    Notes: Abstract Brachypodium distachyon (L.) P. Beauv. has several features of its genome and growth habit reminiscent of Arabidopsis thaliana (L.) Heyn. that may allow it to be developed as a model molecular genetic system representative of the temperate grasses. In order for B. distachyon to be exploited in this way, it will be necessary to develop tissue culture procedures. This report details initial studies of the characteristics of mature seed-derived callus and the production of fertile plants from callus of three ecotypes of B. distachyon. Optimum development of embryogenic callus occurred on LS (Linsmaier & Skoog 1965) and N6 (Chu et al. 1975) media containing 3.0% w/v sucrose and 11.25 μM (2.5 mg l-1) 2,4-dichlorophenoxyacetic acid. Plants were recovered at a high frequency from embryogenic callus of ecotype B200 maintained on growth regulator-free N6 medium and were easy to establish in compost. A method was also developed for the in vitro clonal propagation of shoots using MS (Murashige & Skoog 1962) medium supplemented with 4 to 13 μM (1.0 to 3.0 mg l-1) benzyladenine. It was concluded that B. distachyon performed well in tissue culture and was suitable for further studies aimed at genetic transformation and its continued development as a model molecular genetic system.

    Notes: Summary Protoplasts isolated from celery cell suspension cultures, were mixed with fungal protoplasts, from either the saprophytic speciesAspergillus nidulans or the pathogenic speciesFusarium oxysporum. The incubation of protoplast mixtures with PEG caused close adhesion between plant and fungal protoplasts. Subsequent dilution of PEG resulted in the uptake of protoplasts from either fungal species into the plant protoplast cytoplasm. A range of PEG concentrations, incubation times and dilution rates were tested to maximise adhesion and uptake frequencies. Identification of uptake was achieved either by fluorescent staining of nuclei or by electron-microscopy. A maximum of 10% celery protoplasts had taken upA. nidulans protoplasts after PEG treatment. Fungal protoplasts were taken up into celery protoplast cytoplasm by endocytosis, and were maintained within vesicles; two bounding membranes were observed by electron microscopy. Plant protoplast viability was determined during prolonged incubation following fungal protoplast uptake. The presence ofA. nidulans protoplasts tended to maintain celery protoplast viability and although some morphological disintegration occurred intact celery protoplasts remained for at least 92 h after uptake. The uptake ofF. oxysporum protoplasts markedly depressed celery protoplast viability after 24 h incubation and greater celery protoplast disintegration occurred.
