    Notes: Abstract Rhodococcus strain I24 is able to convert indene into indandiol via the actions of at least two dioxygenase systems and a putative monooxygenase system. We have identified a cosmid clone from I24 genomic DNA that is able to confer the ability to convert indene to indandiol upon Rhodococcus erythropolis SQ1, a strain that normally can not convert or metabolize indene. HPLC analysis reveals that the transformed SQ1 strain produces cis-(1R,2S)-indandiol, suggesting that the cosmid clone encodes a naphthalene-type dioxygenase. DNA sequence analysis of a portion of this clone confirmed the presence of genes for the dioxygenase as well as genes encoding a dehydrogenase and putative aldolase. These genes will be useful for manipulating indene bioconversion in Rhodococcus strain I24.

    Notes: After a 2 h exposure of intact soybean nodules to high concentrations of NaCl (100mol m−3) or oxygen (8OkPa O2), morphometric computations carried out using an image analysis technique on semi-thin sections showed that both treatments induced a decrease in the area of the inner-cortex cells, which were then characterized by a tangential elongation. In contrast, no significant change in area occurred in the middle-cortex cells although their elongation decreased. Electron microscopic observations showed that in the inner-cortex cells changes included the presence of wall infoldings, an enlarged periplasmic space and a lobate nucleus whose chromatin distribution differed from that of the control. Structural changes also occurred in the endoplasmic reticulum, microbodies, mitochondria and plastids. From several of these changes, which are similar to those noted in osmocontractil cells in response to external stimuli, it can be hypothesized that the inner cortex may provide a potential mechanism for the control of oxygen diffusion through the nodules.

    Notes: Abstract When the leaves of Mimosa pudica are changed from their normal position in the gravitational field, they perform reversible compensatory movements by means of pulvini. These movements are not the result of growth processes but involve reversible turgor variations. These variation are concomitant with ion migrations within pulvini: during the gravitropic movement, K+ and Cl- shift towards the adaxial half of the motor organ whereas Ca2+ shifts towards the abaxial half. Compounds known to affect K+ transport, tetraethylammonium chloride and valinomycin, do not hinder the gravitropic movement but inhibit strongly the seismonastic reaction. The same general result is obtained with compounds affecting anion transport, disulfonic stilbenes and 9-anthracene carboxylic acid. Calcium chelators inhibit the gravitropic movement more efficiently than the seismonastic reaction and the calcium ionophore A 23 187 increases both movements. The data obtained with these various compounds indicate that ions do not have the same functional importance in the regulation of the two different pulvinar movements.

    Notes: Abstract To develop a transposable element-based system for mutagenesis in Rhodococcus, we used the sacB gene from Bacillus subtilis to isolate a novel transposable element, IS1676, from R. erythropolis SQ1. This 1693 bp insertion sequence is bounded by imperfect (10 out of 13 bp) inverted repeats and it creates 4 bp direct repeats upon insertion. Comparison of multiple insertion sites reveals a preference for the sequence 5′-(C/T)TA(A/G)-3′ in the target site. IS1676 contains a single, large (1446 bp) open reading frame with coding potential for a protein of 482 amino acids. IS1676 may be similar to an ancestral transposable element that gave rise to repetitive sequences identified in clinical isolates of Mycobacteriumkansasii. Derivatives of IS1676 should be useful for analysis of Rhodococcus strains, for which few other genetic tools are currently available.

    Notes: Summary InMimosa pudica the primary and secondary motor organs (pulvini) of fully grown leaves are capable of graviresponse. These organs possess sedimentable amyloplasts in their starch sheath cells. In the primary pulvinus these cells are characterized by a structural polarity induced by the localization of nucleus at their (morphologically) apical part and the localization of amyloplasts at their (physically) basal part. These cells also display structural peculiarities including plasmodesmatal disposition, little development of the endoplasmic reticulum and an absence of vacuolar tannins; moreover, the sedimentation of the amyloplasts, induced by gravistimulation, is accompanied by the variation of localization of the cytoplasm, vacuole and mitochondria and by structural modifications of the nucleus and endoplasmic reticulum.

    Notes: Summary Immunocytochemical techniques were employed to study the spatial distribution of the plasma membrane H+-ATPase within various cell types of the young reactive primary pulvinus ofMimosa pudica L. These cells were interconnected by large numbers of plasmodesmata, being concentrated within pit-fields. Although we could routinely detect evidence of the H+-ATPase along the plasma membrane, immunolabelling was rarely, if ever, observed along the plasma membranes of the plasmodesmata. This finding is discussed with respect to the likely specialized supramolecular structure of the plasmodesma.

    Notes: Abstract As part of a program to develop biological wastewatertreatment systems for cold climate areas four strainsof filamentous, mat-forming cyanobacteria isolatedfrom Arctic and Antarctic environments were evaluatedfor their nutrient stripping and growth capabilities. A tropical strain, Phormidium bohneri, known forits excellent performance in wastewater treatment, wasused as a comparison. Experiments were done inartificial media under controlled batch cultureconditions to avoid interactions with indigenousmicroorganisms such as bacteria and protozoa. Theculture medium simulated real effluents containinghigh concentrations of nitrate and phosphate.Temperatures (5, 15 and 25°C) and irradiances(80, 210, 350, 640 and 1470 µmol photon m-2s-1) wereselected according to situations encountered in avariety of field conditions. For all irradiancelevels, growth was satisfactory at 15 and 25 °C,but limited at 5 °C. At 25 °C a satisfactory nitrogen removal rate (3.5and 4.0 mg N L-1d-1) was obtained forone polar strain (Phormidium tenue) and thecontrol P. bohneri. At 15 °C, the bestnitrogen removal rate (3.5 mg N L-1d-1)was measured with P. bohneri while the best ratefor the polar strains was around 2.3 mg NL-1d-1. At 15 °C, a phosphorusremoval rate of 0.6 mg P L-1d-1 wasobtained with P. bohneri and polar strains P. tenue and Oscillatoria O-210. Nitrogen(NO3 -) and phosphorus (PO4 3-)uptake rates increased as a function of irradianceover the range 80 to 350 μmolphoton m-2s-1. Our results indicate thattertiary biological wastewater treatment at lowtemperatures (5 °C) cannot be anticipated withthe polar strains tested, because they arepsychrotrophic rather than psychrophilic and thus growtoo slowly under conditions of extreme cold. However, it appears that these cyanobacteria would beuseful for wastewater treatment at moderately cooltemperatures (c. 15 °C), which are commonduring spring and fall in northern climates.

    Notes: Polymer-bonded smoke compositions offers numerous advantages over conventional pressed smoke mixtures such as improved mechanical properties and safety in handling. The Defense Research Establishment Valcartier (DREV) has developed pour-castable coloured smoke compositions using a well known hydroxy-terminated polybutadiene prepolymer cured with a commercially available diisocyanate. These smoke formulations consist of ingredients typically found in pressed powder mix, but dispersed in a polymeric binder. A granulated dye is utilized to provide the low viscosities required for pour-casting. The necessary ingredients and processing techniques as well as the properties and inherent advantages of such compositions are described.
