    Notes: Summary On the basis that unbound concentration better correlates with response than total plasma or blood concentration, the inter- and intra-subject variability in the distribution of cyclosporin within blood and to plasma components was studied in renal transplant patients. Pharmacokinetic aspects were also studied. Blood samples were analysed from patients who received the drug both by a 72-h i.v. infusion and orally (7 mg·kg−1 twice daily). Steady-state was reached within 18 h of starting the i.v. infusion; the plasma data were best fitted by a biexponential equation with half-times of 0.13–1.02 h and 4.3–13.9 h, associated with the two phases. The mean plasma clearance was 700 ml/min. Concentrations during the infusions measured by RIA and HPLC were comparable. Oral profiles showed rapid and extensive absorption. The peak plasma concentrations were 1460–1880 µg·l−1 and occurred 2–4 h after dosing, with bioavailability estimates of 41–113%. Concentrations measured by RIA were higher than by HPLC. Blood-to-plasma concentration ratio measurements of cyclosporin at 37°C decreased with increasing plasma concentration and increased with haematocrit. Fraction unbound, measured by ultracentrifugation, was in the range 0.042–0.122 with an average of 0.068, and varied little in some patients but showed systematic changes with time in others. Cyclosporin binding was found to be related not only to the triglyceride but, more particularly, to the cholesterol-related lipoproteins in plasma. Monitoring cholesterol may be helpful in identifying patients with extremes in binding or with widely varying binding.

    Notes: Summary The excretion of radioactivity has been investigated in 3 healthy volunteers following rapid intravenous administration of 5 g of [35S]-ticarcillin. The radioactive dose was rapidly and completely excreted, since within 4 days 98.5% was recovered, 95% in the urine and 3.5% in faeces. All the urine radioactivity was accounted for as ticarcillin and its penicilloic acid. Plasma and urine samples collected from the volunteers at frequent intervals during the first 6 h of the experiment were assayed for penicillin by an automated chemical method and also for radioactivity. The results obtained by the chemical autoanalyser method were in excellent agreement with the plasma levels of radioactivity. From the data it was possible to calculate the renal clearance of the penicillin, a mean value of 104 ml/min was observed in the 3 volunteers. A further three volunteers were dosed intravenously with a 5 g bolus of non-radiolabelled ticarcillin in a cross-over study with and without predosing with probenecid. Serum samples were analysed by the chemical method for penicillin and the data subjected to pharmacokinetic analysis using a two compartment open model. The results indicate a shift of the distribution equilibrium of ticarcillin from the serum into the peripheral compartment after predosing with probenecid. Furthermore, the mean half-life of ticarcillin in the serum of the three volunteers was significantly increased from 1.3 h to 2.1 h by predosing with probenecid.

    Notes: Abstract Carbon dioxide is transported from the atmosphere to leaf surfaces very efficiently, and atmospheric diffusion resistances rarely affect photo-synthetic rates by more than 10%. The largest single resistance is in the boundary layer of individual leaves, and this is a well-known function of leaf size, wind speed, and leaf to air temperature difference. For uniform vegetative canopies a fully turbulent boundary layer exists above; this layer is also well understood and well-proven equations can be used to relate the carbon dioxide flux to the concentration gradient. Transport in the air within and just above vegetative canopies is less well understood. Turbulent diffusion theory has been shown to be inadequate, and consequently the diffusion resistance analogy cannot be used. Recent work, however, shows that Lagrangian theories of dispersion offer an alternative and more accurate description of transport within canopies.

    Notes: Abstract Recent papers by Wilson et al. (1981b) and Legg and Raupach (1982) give methods for the calculation of particle trajectories in turbulence with a gradient in vertical velocity variance Σ 2 w. However the two methods seem contradictory. This paper demonstrates that in systems in which l(dΣ w /dz) (where / is the length scale) varies only slowly with height z, the two methods give similar predictions, and indicates why this is the case. For a particular system in which the restriction on l(dΣ w /dz) is not satisfied, it is shown that neither method is correct but that a simple modification of the method of Wilson et al. (1981b) gives reasonable predictions.

Conclusions were: (i) Despite the very heterogeneous flow within the canopy, the observed dispersive heat flux (due to spatial correlation between time-averaged temperature and vertical velocity) was small. (iii) The eddy diffusivity for heat from the line source (K HL ) increased, downstream of the source, to a nearly constant ‘far-field’ vertical profile. Within the canopy, the far-field K HL was an order of magnitude larger than K HP , the equivalent diffusivity for a plane source; well above the canopy, the two were equal. The time scale defined by (far-field K HL )/(vertical velocity variance) was independent of height within the canopy. (iv) Budgets for temperature variance, vertical heat flux and streamwise heat flux are remarkably similar to the equivalent budgets for an elevated line source in the surface layer well above the canopy, except in the lower part of the canopy in the far field, where vertical transport is much more important than in the surface layer. (v) A random flight simulation of the mean height and depth of the temperature plume was generally in good agreement with experiment. However, details of the temperature and streamwise turbulent heat flux profiles were not correct, suggesting that the model formulation needs to be improved.

    Notes: Abstract This is the first of a series of three papers describing experiments on the dispersion of trace heat from elevated line and plane sources within a model plant canopy in a wind tunnel. Here we consider the wind field and turbulence structure. The model canopy consisted of bluff elements 60 mm high and 10 mm wide in a diamond array with frontal area index 0.23; streamwise and vertical velocity components were measured with a special three-hot-wire anemometer designed for optimum performance in flows of high turbulence intensity. We found that: (i) The momentum flux due to spatial correlations between time-averaged streamwise and vertical velocity components (the dispersive flux) was negligible, at heights near and above the top of the canopy. (ii) In the turbulent energy budget, turbulent transport was a major loss (of about one-third of local production) near the top of the canopy, and was the principal gain mechanism lower down. Wake production was greater than shear production throughout the canopy. Pressure transport just above the canopy, inferred by difference, appeared to be a gain in approximate balance with the turbulent transport loss. (iii) In the shear stress budget, wake production was negligible. The role of turbulent transport was equivalent to that in the turbulent energy budget, though smaller. (iv) Velocity spectra above and within the canopy showed the dominance of large eddies occupying much of the boundary layer and moving downstream with a height-independent convection velocity. Within the canopy, much of the vertical but relatively little of the streamwise variance occurred at frequencies characteristic of wake turbulence. (v) Quadrant analysis of the shear stress showed only a slight excess of sweeps over ejections near the top of the canopy, in contrast with previous studies. This is a result of improved measurement techniques; it suggests some reappraisal of inferences previously drawn from quadrant analysis.

    Notes: Abstract The Langevin equation is used to derive the Markov equation for the vertical velocity of a fluid particle moving in turbulent flow. It is shown that if the Eulerian velocity variance Σ wE is not constant with height, there is an associated vertical pressure gradient which appears as a force-like term in the Markov equation. The correct form of the Markov equation is: w(t + δt) = aw(t) + bΣ wEζ + (1 − a)T L ∂(Σ wE 2)/∂z, where w(t) is the vertical velocity at time t, ζ a random number from a Gaussian distribution with zero mean and unit variance, T L the Lagrangian integral time scale for vertical velocity, a = exp(−δt/T L), and b = (1 − a 2)1/2. This equation can be used for inhomogeneous turbulence in which the mean wind speed, Σ wE and T L vary with height. A two-dimensional numerical simulation shows that when this equation is used, an initially uniform distribution of tracer remains uniform.
