    Description: The 5'-AMP-activated protein kinase (AMPK) functions as a cellular energy sensor. 5-Aminoimidazole-4-carboxyamide-1-β-D-ribofranoside (AICAR) is a chemical activator of AMPK. In the liver, AICAR suppresses expression of the phosphoenolpyruvate carboxykinase ( PEPCK ) gene. The rat enhancer of split- and hairy-related protein-2 (SHARP-2) is an insulin-inducible transcriptional repressor and its target is the PEPCK gene. In this study, we examined an issue of whether the SHARP-2 gene expression is regulated by AICAR via the AMPK. AICAR increased the level of SHARP-2 mRNA in H4IIE cells. Whereas an AMPK inhibitor, compound-C, had no effects on the AICAR-induction, inhibitors for both phosphoinositide 3-kinase (PI 3-K) and protein kinase C (PKC) completely diminished the effects of AICAR. Western blot analyses showed that AICAR rapidly activated atypical PKC lambda (aPKC). In addition, when a dominant negative form of aPKC was expressed, the induction of SHARP-2 mRNA level by AICAR was inhibited. Calcium ion is not required for the activation of aPKC. A calcium ion-chelating reagent had no effects on the AICAR-induction. Furthermore, the AICAR-induction was inhibited by treatment with an RNA polymerase inhibitor or a protein synthesis inhibitor. Thus, we conclude that the AICAR-induction of the SHARP-2 gene is mediated at transcription level by a PI 3-K/aPKC pathway.

    Description: We examine the applicability of the stochastic electron acceleration to two high synchrotron peaked blazars, Mrk 421 and Mrk 501, assuming synchrotron self-Compton emission of gamma-rays. Our model considers an emitting region moving at relativistic speed, where non-thermal electrons are accelerated and attain a steady-state energy spectrum together with the photons they emit. The kinetic equations of the electrons and photons are solved numerically, given a stationary wavenumber spectrum of the magnetohydrodynamic (MHD) disturbances, which are responsible for the electron acceleration and escape. Our simple formulation appears to reproduce the two well-sampled, long-term averaged photon spectra. In order to fit the model to the emission component from the radio to the X-ray bands, we need both a steeper wave spectral index than the Kolmogorov spectrum and efficient particle escape. Although the model provides a natural explanation for the high-energy cutoff of the electron energy distribution, the derived physical parameters raise a problem with an energy budget if the MHD waves with the Alfvén velocity are assumed to be the acceleration agent.

    Description: To achieve the high sealing capability required for the bentonite engineered barriers used in geological disposal facilities for radioactive waste, the bentonite should be highly compacted. Previous investigation of methods to enable such compaction has revealed that the construction methods used have an impact on the density distribution. However, these investigations focused on achieving the required average dry density in the bentonite barriers and little attention has been paid to the distribution of dry density. As a result, there has been a lack of understanding of how the construction method used affects this distribution, which is highly pertinent to the evaluation of the long-term performance of bentonite engineered barriers. This paper first describes the residual density distribution after resaturation and then highlights how swelling behaviour can be studied and evaluated based on elasto-plastic theory. The conceptual understanding is that the buffer does not swell to achieve an entirely homogeneous density, but will reach an equilibrium state with a remaining gradient.

    Description: A cornerstone of Einstein's special relativity is Lorentz invariance-the postulate that all observers measure exactly the same speed of light in vacuum, independent of photon-energy. While special relativity assumes that there is no fundamental length-scale associated with such invariance, there is a fundamental scale (the Planck scale, l(Planck) approximately 1.62 x 10(-33) cm or E(Planck) = M(Planck)c(2) approximately 1.22 x 10(19) GeV), at which quantum effects are expected to strongly affect the nature of space-time. There is great interest in the (not yet validated) idea that Lorentz invariance might break near the Planck scale. A key test of such violation of Lorentz invariance is a possible variation of photon speed with energy. Even a tiny variation in photon speed, when accumulated over cosmological light-travel times, may be revealed by observing sharp features in gamma-ray burst (GRB) light-curves. Here we report the detection of emission up to approximately 31 GeV from the distant and short GRB 090510. We find no evidence for the violation of Lorentz invariance, and place a lower limit of 1.2E(Planck) on the scale of a linear energy dependence (or an inverse wavelength dependence), subject to reasonable assumptions about the emission (equivalently we have an upper limit of l(Planck)/1.2 on the length scale of the effect).     Description: Gamma-ray bursts (GRBs) are highly energetic explosions signaling the death of massive stars in distant galaxies. The Gamma-ray Burst Monitor and Large Area Telescope onboard the Fermi Observatory together record GRBs over a broad energy range spanning about 7 decades of gammaray energy. In September 2008, Fermi observed the exceptionally luminous GRB 080916C, with the largest apparent energy release yet measured. The high-energy gamma rays are observed to start later and persist longer than the lower energy photons. A simple spectral form fits the entire GRB spectrum, providing strong constraints on emission models.     Description: We sequenced the 29,751-base genome of the severe acute respiratory syndrome (SARS)-associated coronavirus known as the Tor2 isolate. The genome sequence reveals that this coronavirus is only moderately related to other known coronaviruses, including two human coronaviruses, HCoV-OC43 and HCoV-229E. Phylogenetic analysis of the predicted viral proteins indicates that the virus does not closely resemble any of the three previously known groups of coronaviruses.     Description: The observations of the exceptionally bright gamma-ray burst (GRB) 130427A by the Large Area Telescope aboard the Fermi Gamma-ray Space Telescope provide constraints on the nature of these unique astrophysical sources. GRB 130427A had the largest fluence, highest-energy photon (95 GeV), longest gamma-ray duration (20 hours), and one of the largest isotropic energy releases ever observed from a GRB.     Description: The Radioactive Waste Management Funding and Research Center (RWMC) is leading a research programme to evaluate the gas transport mechanisms through a TRU (TRans-Uranium) waste disposal facility in Japan and acquire information on gas migration properties. In this paper we describe a series of laboratory gas injection tests using the bentonite adopted for use in Japanese TRU disposal, as well as an attempted visualization of the gas migration path inside the bentonite used as a buffer. By building a conceptual model from the results of these tests, the characteristics of gas migration through to breakthrough for bentonite can now be better understood in the context of Japanese TRU disposal. Thanks to the outcome of this research project, advanced knowledge may be applied to the conceptualization of TRU waste disposal.

    Description: We have determined the x-ray crystal structure of l -lysine -oxidase from Marinomonas mediterranea in its native and l -lysine-complex forms at 1.94- and 1.99-Å resolution, respectively. In the native enzyme, electron densities clearly indicate the presence of cysteine tryptophylquinone (CTQ) previously identified in quinohemoprotein amine dehydrogenase. In the l -lysine-complex, an electron density corresponding to the bound l -lysine shows that its -amino group is attached to the C6 carbonyl group of CTQ, suggesting the formation of a Schiff-base intermediate. Collectively, the present crystal structure provides the first example of an enzyme employing a tryptophylquinone cofactor in an amine oxidase.
