    Notes: Summary Measurements of air temperature, wind speed and the temperature depth profile at Llyn Tegid, North Wales, from April to November 1969, are used to establish equations which are then used for the observed meteorological data to predict the temperature profile of the lake during this period and in particular the conditions under which the thermocline rises or sinks. A fair measure of agreement is found between observed and predicted profiles.

    Notes: Summary A new variant of human erythrocyte carbonic anhydrase II (CA II) was discovered in a single Caucasian family during routine screening of blood samples from Melbourne, Australia. The normal and variant enzymes in the heterozygous CA II mixture, as well as a minor component of the normal enzyme, were resolved by isoelectric focusing following purification by a specific affinity matrix. Specific esterase activities of all three were very similar, but quite different Michaelis-Menten constants were noted for the minor component. No differences were noted with respect to inhibition by acetazolamide, but the minor component was more sensitive to chloride inhibition. Double diffusion analysis showed the immunological identity of the normal, variant, and minor components. Both the variant CA II and the minor component were less heat stable than the normal enzyme, but all forms showed identical rates of inactivation upon dialysis against the zinc chelator pyridine dicarboxylic acid. Amino acid analyses of the whole protein and the single difference peptide were consistent with a proline to histidine substitution in the variant. This was identified as 237 Pro→His by a process of elimination involving direct sequencing of tryptic and cyanogen bromide peptides. The numbering is by homology with the human CA I sequence.

    Notes: Summary A study of the distribution of α-thalassemia in Papua New Guinea (PNG) was carried out by DNA analysis. A total of 664 DNA samples were screened for α-thalassemia 2 and α-thalassemia 1 caused respectively by either deletion of one or both of the duplicated α-globin genes. α-Thalassemia 2 was detected in high frequencies in coastal and lowland regions where malaria has been holo- to hyperendemic but in low frequencies in non-malarious highland regions. The highest frequency was observed in the north coast of PNG. The distribution of α-thalassemia 2 seems to be in accordance with other conditions such as ovalocytosis and G6PD deficiency which are also prevalent in this population, suggesting that they may interact in protection against malaria. However, it appears to be negatively correlated with β-thalassemia and α-thalassemia 1, the latter being extremely rare in this population. Analysis of the types and subtypes of the single α-globin gene deletion revealed a predominance of the −α4.2 type in general, except in some regions in the south where the −α3.7 type is prevalent. The −α3.7 I subtype is the common form of the −α3.7 deletion in the PNG mainland. The −α3.7 III subtype, previously reported to be unique in Melanesians and Polynesians, was detected in an offshore island of PNG. However, this subtype is very rare in Melanesians from the PNG mainland.

    Notes: Summary The ultrastructure and composition of the extracellular matrices (ECMs) associated with germ tubes and appressoria ofColletotrichum lindemuthianum have been examined. Flexuous fibres (fimbriae), up to 6 μm long and 4–30 nm in diameter, protruded from the surface of germ tubes and appressoria. Anionic colloidal gold and lectin cytochemistry showed that ECMs of germ tubes and appressoria contain basic proteins, α-D-mannose and α-D-galactose residues. A monoclonal antibody, UB26, was raised to infection structures isolated from leaves ofPhaseolus vulgaris infected withC. lindemuthianum. UB26 recognised a protein epitope on two glycoproteins (Mr 133,000 and 146,000). Reductions in the Mr of these proteins after treatment with peptide-N-glycosidase and trifluoromethane sulphonic acid suggest that they carry N- and O-linked side-chains. Immunofluorescence and EM-immunogold labelling showed that glycoproteins recognised by UB26 were restricted to the ECMs around germ tubes and appressoria but fimbriae were not labelled. Unlike appressorial germ tubes formed in vitro, intracellular infection hyphae were not labelled, suggesting that the glycoproteins recognised by UB26 are not present on fungal structures formed within host cells. In liquid culture, these glycoproteins were not released into the medium, suggesting they are physically linked to the cell wall. Also, the glycoproteins were not removed from glass surfaces by ultrasonication. These results suggest that glycoproteins recognised by UB26 may be involved in the adhesion of germ tubes and appressoria to substrata. Our results show that the ECMs of germ tubes and appressoria differ markedly in structure and composition from those of conidia and intracellular hyphae, and that extracellular glycoproteins are associated with specific regions of the fungal cell surface.

    Notes: Abstract This paper describes a test fixture and alignment jig used for testing small concrete or rock cylinders in uniaxial tension. The fixture may be easily modified to test cube specimens and is adaptable for use in any standard universal testing machine. A testing procedure is described and results presented for 3 in. × 6 in. (76 mm × 152 mm) concrete cylinders. The main virtues of the fixture are its ease of construction, simple alignment procedure for the test specimen, and accuracy comparable to that obtained from the standard compression test of concrete cylinders.

    Notes: Abstract Acoustic-emission (AE) count rates are presented for tensile loading of unidirectional boron-epoxy and for aluminum sheets reinforced with unidirectional boron-epoxy. It is shown that different prepreg materials have different characteristic AE patterns. Results from composite-reinforced metal specimens show that early failures are accompanied by a sharp increase in AE count rate at the knee of the bilinear stress-strain diagram. It is further shown that the count rates are a function of specimen fabrication and that higher total counts do not necessarily correspond to early failures.
