    Notes: Summary Plasmodial cells of the slime moldPhysarum polycephalum become competent for sporulation following a prolonged period of starvation in darkness. Then sporulation can be induced by illumination. Microinjections of the stable (Sp)- and (Rp)-diastereoisomers of adenosine cyclic 3′,5′ monothionophosphate before and during a sensitive period from the start of illumination until 5 h after lead to a significant delay in the sporulation process. Both of the diastereoisomers of cyclic AMP prolong the time for sporangia to form in darkness. However, the (Sp)-diastereoisomer is more effective and causes morphological changes in plasmodia. The experimental data suggest that cyclic AMP is decisively involved in light-induced differentiation in the lower eukaryotoPhysarum polycephalum.

    Notes: Abstract: Dictyostelium cells exhibit four types of kinetically distinct surface cAMP binding sites, the AH, AL, BS, and BSS sites, which are down-regulated during persistent stimulation with cAMP. Although most cAMP-induced responses are subject to desensitization during constant stimulation, some responses, notably the induction of post-aggregative gene expression, require persistent cAMP stimulation. The kinetics and specificity of residual cAMP-binding activity in cells treated for 4 h with micromolar cAMP were investigated. It was found that around 4000 rapidly dissociating binding sites per cell with an affinity of about 300 nM are retained after down-regulation. The nucleotide specificity of the remaining sites was very similar, but not completely identical to the AH, AL and B sites, suggesting that these sites belong to the same class of cell surface cAMP receptors and may be utilized to mediate responses requiring continuous cAMP stimulation.

    Description / Table of Contents: The Stereoselectivity of the Synthesis of 5′-N-Substituted 5′-Amido Analogs of Adenosine-3′,5′-cyclophosphate p-Nitrophenyl EsterThe base-catalysed cyclization of bis(p-nitrophenyl) N-(5′-deoxy-5′-adenosyl)-N-octylphosphor-amidate (1, R = n-octyl) gives stereoselectively 5′-deoxy-5′-(octylamino)adenosin-3′-phosphoric acid p-nitrophenyl ester 3′,5′-cycloainide (2a). It can be converted into its diastcreomer 2 b by treatment with p-nitrophenolate ion. The configuration of the chiral phosphorus atom was derived from the reaction mechanism and was confirmed through n.m.r. spectra.
    Notes: Die basenkatalysierte Ringschlußreaktion von N-(5′-Desoxy-5′-adenosyl)-N-octylphosphor-säure-bis(p-nitrophenylester)-amid (1, R = n-Octyl) führt stereoselektiv zu 5′-Desoxy-5′-(octylamino)adenosin-3′-phosphorsäure-p-nitrophenylester-3′,5′-cycloamid (2a). Es kann durch Behandlung mit p-Nitrophenolat-Ion in das Diastereomere 2b übergeführt werden. Die Konfiguration am chiralen Phosphoratom wurde aufgrund des Reaktionsmechanismus abgeleitet und durch NMR-Spektren bestätigt.

    Description / Table of Contents: Systematic Investigation of the Reaction Conditions for the Phosphorylation 5′-Amino-5′-deoxyguanosine CompoundsReaction conditions for the phosphorylation of 5′-amino-, 5′-Amino-2′,3′-isopropylidene-, and 5′-butylamino- 5′-deoxyguanosine (1a,b,c respectively) with diesters of phosphorochloridic acid have been systematically investigated and the dependence of the reaction on reagent, solvent, tertiary base and guanosine derivative has been determined. A new, generally applicable phosphorylating system employing a diester of phosphorochloridic acid, N,N- diiso-propylethylamine and triethylphosphate (or dimethylformamide) is described.
    Notes: Die Phosphorylierungsbedingungen für 5′-Amino-, 5′-Amino-2′,3′-isopropyliden- bzw. 5′- Butylamino-5′-desoxyguanosin (1a, b bzw. 1c) mit Phosphorsäurediesterchloriden wurden systematisch untersucht und eine Abhängigkeit von allen Reaktionsparametern wie Reagenz, Lösungsmittel, tertiärer Hilfsbase und Guanosinderivat festgestellt. Ein neues generell geeignetes Phosphorylierungssystem, bestehend aus Phosphorsäurediesterchlorid, N,N-Diiso-propyläthyiamin und Triäthylphosphat bzw. Dimethylformamid, wurde gefunden.

Removal of the O-protecting groups and one phenyl group from the diphenyl ester phosphoramidates 4a and 6a yields substrates for snake venom phosphodiesterase.
    Notes: Eine neue, mit hohen Ausbeuten verlaufende Darstellung von Diestern der 5′-Amino-5′-desoxy-nucleosid-N-phosphate 4 bzw. 6 durch Phosphorylierung verschiedener 5′-Azido-5′-desoxy-nucleoside 3 bzw. 5 mit Triphenyl-bzw. Trimethylphosphit (1 bzw. 2) wird beschrieben. Durch Abspaltung der O-Schutzgruppen und einer Phenylgruppe aus den Diphenylesteramiden 4a und 6a erhält man Substrate für die Schlangengift-Phosphodiesterase.

    Description / Table of Contents: 5′-Amino-5′-deoxyguanosine and its 5′-N-Substituted DerivativesA modified synthesis and previously unknown properties of 2′,3′O-isopropylidene-5′-O-tosylguanosine (2) are described. Ten derivatives 7a-k of 5′-amino-5′-deoxyguanosine with different residues on the 5′-nitrogen were synthesized by direct aminolysis of the 5′-O-tosylguanosine (4) with primary amines. The chemical properties of the guanosine analogues can be modified in a predictable manner by the  -  NHR residue in the 5′-position.
    Notes: Eine modifizierte Synthese und bislang nicht bekannte Eigenschaften des 2′,3′-O-Isopropyliden-5′-O-tosylguanosins (2) werden beschrieben. Zehn Derivate 7a-k des 5′-Amino-5′-desoxyguanosins mit verschiedenen Resten am 5′-Stickstoff wurden durch direkte Aminolyse von 5′-O-Tosylguanosin (4) mit primären Aminen synthetisiert. Die chemischen Eigenschaften der Guanosin-Analogen lassen sich durch den - NHR-Rest in gewünschter Weise verändern.
