    Description: : Transcription activator-like effector nucleases (TALENs) have become an accepted tool for targeted mutagenesis, but undesired off-targets remain an important issue. We present TALENoffer, a novel tool for the genome-wide prediction of TALEN off-targets. We show that TALENoffer successfully predicts known off-targets of engineered TALENs and yields a competitive runtime, scanning complete mammalian genomes within a few minutes.     Description: Aims and Methods Disturbance is supposed to play an important role for biodiversity and ecosystem stability as described by the intermediate disturbance hypothesis (IDH), which predicts highest species richness at intermediate levels of disturbances. In this study, we tested the effects of artificial soil disturbances on diversity of annual and perennial vascular plants and bryophytes in a field experiment in 86 agricultural grasslands differing in land use in two regions of Germany. On each grassland, we implemented four treatments: three treatments differing in application time of soil disturbances and one control. One year after experimental disturbance, we recorded vegetation and measured biomass productivity and bare ground. We analysed the disturbance response taking effects of region and land-use-accompanied disturbance regimes into account. Important Findings Region and land-use type strongly determined plant species richness. Experimental disturbances had small positive effects on the species richness of annuals, but none on perennials or bryophytes. Bare ground was positively related to species richness of bryophytes. However, exceeding the creation of 12% bare ground further disturbance had a detrimental effect on bryophyte species richness, which corresponds to the IDH. As biomass productivity was unaffected by disturbance our results indicate that the disturbance effect on species richness of annuals was not due to decreased overall productivity, but rather due to short-term lowered inter- and intraspecific competition at the newly created microsites. Generally, our results highlight the importance of soil disturbances for species richness of annual plants and bryophytes in agricultural grasslands. However, most grasslands were disturbed naturally or by land-use practices and our additional experimental soil disturbances only had a small short-term effect. Overall, total plant diversity in grasslands seemed to be more limited by the availability of propagules rather than by suitable microsites for germination. Thus, nature conservation efforts to increase grassland diversity should focus on overcoming propagule limitation, for instance by additional sowing of seeds, while the creation of additional open patches by disturbance might only be appropriate where natural disturbances are scarce.

    Description: The pathogenicity of many bacteria depends on the injection of effector proteins via type III secretion into eukaryotic cells in order to manipulate cellular processes. TAL (transcription activator-like) effectors from plant pathogenic Xanthomonas are important virulence factors that act as transcriptional activators in the plant cell nucleus, where they directly bind to DNA via a central domain of tandem repeats. Here, we show how target DNA specificity of TAL effectors is encoded. Two hypervariable amino acid residues in each repeat recognize one base pair in the target DNA. Recognition sequences of TAL effectors were predicted and experimentally confirmed. The modular protein architecture enabled the construction of artificial effectors with new specificities.     Notes: Abstract Production of the compatible solute glycine betaine from its precursors choline or glycine betaine aldehyde confers a considerable level of tolerance against high osmolarity stress to the soil bacterium Bacillus subtilis. The glycine betaine aldehyde dehydrogenase GbsA is an integral part of the osmoregulatory glycine betaine synthesis pathway. We strongly overproduced this enzyme in an Escherichia coli strain that expressed a plasmid-encoded gbsA gene under T7φ10 control. The recombinant GbsA protein was purified 23-fold to apparent homogeneity by fractionated ammonium sulfate precipitation, ion-exchange chromatography on Q-Sepharose, and subsequent hydrophobic interaction chromatography on phenyl-Sepharose. Molecular sieving through Superose 12 and sedimentation centrifugation through a glycerol gradient suggested that the native enzyme is a homodimer with 53.7-kDa subunits. The enzyme was specific for glycine betaine aldehyde and could use both NAD+ and NADP+ as cofactors, but NAD+ was strongly preferred. A kinetic analysis of the GbsA-mediated oxidation of glycine betaine aldehyde to glycine betaine revealed K m values of 125 μM and 143 μM for its substrates glycine betaine aldehyde and NAD+, respectively. Low concentrations of salts stimulated the GbsA activity, and the enzyme was highly tolerant of high ionic conditions. Even in the presence of 2.4 M KCl, 88% of the initial enzymatic activity was maintained. B. subtilis synthesizes high levels of proline when grown at high osmolarity, and the presence of this amino acid strongly stimulated the GbsA activity in vitro. The enzyme was stimulated by moderate concentrations of glycine betaine, and its activity was highly tolerant against molar concentrations of this osmolyte. The high salt tolerance and its resistance to its own reaction product are essential features of the GbsA enzyme and ensure that B. subtilis can produce high levels of the compatible solute glycine betaine under conditions of high osmolarity stress.

    Description / Table of Contents: Summary It is reported on the experiences hitherto made of the employment of “Nicotinsalicylat” for the purpose of catching chamois, ibex and red deer. By virtue of the Cap-Chur gun, which was altered in course of the experiments (carried trough to reduce the scab of the chamois), a reliable intramuscular injection was finally guaranteed within the range of 70 m. Nicotinsalicylat proved in doses of 9–10 mg/kg applied to chamois, Nicotinsalicylat proved in doses of 5–6 mg/kg applied to ibex, Nicotinsalicylat proved in doses of 15 mg/kg applied to red deer. Further experiments with other races of game are prepared.
    Abstract: Résumé Il est rapporté les expériences qui ont été exécutés jusqu' à présent de l'emploi de Nicotinsalicylat pour prendre le chamois, le bouquetin et les bêtes fauves. Grâce au Cap-Chur fusil, qui fut modifié au cours des expériences faites pour réduire la gale de chamois, une injection sûrement intra-musculaire était garantie enfin jusq' à une portée de 70 m. Nicotinsalicylat a fait ses preuves par l'emploi au chamois en doses de 9–10 mg/kg, au bouquetin en doses de 5–6 mg/kg, aux bêtes fauves en doses de 15 mg/kg. D'autres expériments aux autres éspeces de bêtes fauves sont préparés.
    Notes: Zusammenfassung Es wird über bisherige Erfahrungen über die Verwendung von Nikotinsalicylat zum Einfangen von Gemsen, Steinwild und Rotwild berichtet. Mit Hilfe des im Laufe der Versuche zur Bekämpfung von Gamsräude umgeänderten Cap-Chur-Gewehres war eine sichere intramuskuläre Injektion schließlich bei Schußentfernungen bis 70 m gewährleistet. Nikotinsalicylat hat sich bei Gemsen in einer Dosierung von 9–10 mg/kg, bei Steinwild von 5–6 mg/kg und bei Rotwild von 15 mg/kg Körpergewicht bewährt. Weitere Versuche an anderen Wildarten sind in Vorbereitung.

An 12 künstlich mit Psoroptes cuniculi infizierten Kaninchen im Alter von 6 Wochen bis 1 1/2 Jahren wurden die bei der Ohrräude auftretenden pathologisch-histologischen Veränderungen und Abwehrreaktionen untersucht. 2. Die Jungtiere erkrankten nach Einbringen derselben Milbenmenge in die äußeren Gehörgänge schneller und schwerer als erwachsene Kaninchen. 3. Je nach Lebensalter und allgemeiner Abwehrkraft der Versuchstiere sowie nach dem Befallsgrad waren die entzündlichen Veränderungen an Epidermis und Corium des äußeren Gehörganges verschieden. 4. In fortgeschrittenen Fällen der Erkrankung wurden große Epidermisteile völlig zerstört und abgestoßen. Sekundär durch Eitererreger verursachte Entzündungen der Subkutis führten zum Tod der Tiere.

    Notes: In Barrett's esophagus, metaplastic columnar epithelium replaces the normal squamous epithelium. The importance of this lesion lies in the increased incidence of adenocarcinoma of the esophagus occuring in patients with Barrett's esophagus. We characterized the surface epithelial cells of Barrett's esophagus using quantitative scanning electron microscopy. Three distinct surface cell types, in addition to the goblet cell, were recognized in Barrett's epithelium: the gastric-like cell and the intestinal-like cell, both of which were similar to normal gastric and small intestinal surface cells, respectively, by quantitative scanning electron microscopy, and the variant cell which had a range of surface features. In four biopsy specimens from the squamo-Barrett's junction in three patients, we found the distinctive cell that had features intermediate between those of squamous and columnar epithelium. On the distinctive cell's surface there are two disparate structures not normally present on the same cell in the gastrointestinal tract: microvilli (a scanning electron microscopy feature of glandular epithelium) and intercellular ridges (a scanning electron microscopy feature of squamous epithelium). The surface characteristics of this cell were almost identical to those of cells found in the transformation zone of the uterine cervix, an area in which squamous epithelium physiologically replaces columnar epithelium. Scanning electron microscopy of Barrett's esophagus has increased our understanding of this precancerous lesion by showing striking cellular heterogeneity. It has also identified the distinctive cell which may represent an intermediate step in the development of Barrett's epithelium during which the surface characteristics of two different cell types, columnar and squamous, coexist in the same cell. © 1995 Wiley-Liss, Inc.
