    Notes: Abstract In this study a technique is used which allows observation of chromatin fibers in sections of isolated liver nuclei of the rat. The fibers, which exhibit a rather uniform diameter (diameter = 125 ± 20 Å) seem to be constructed of coiled or folded sub-filaments. — After application of a preferential ribonucleoproteinstain (EDTA-method) only parts of the fibers retain their stain whereas others are completely bleached. Finer filaments (diameter = 45 Å) seem to be continuous with the stained parts of the chromatin fibers. The RNP-stain is lost when the nuclei have been treated with RNase. From these observations it is concluded that RNA-rich and RNA-free segments exist on chromatin fibers which may be related to RNA transcription processes. — Granules and thin filaments are the only structures stained with the EDTA-method in the nucleolus.

    Notes: Summary In freeze-etched vertebrate smooth muscles cells of the mouse intestine thick filaments were observed, showing a main orientation in the long fiber axis and a random arrangement within the cell. The filaments strongly resemble that obtained by thin-sectioning technique of the same tissue.

    Description / Table of Contents: Summary When fibers of a smooth invertebrate muscle (Anterior byssus retractor muscle ofMytilus edulis) were perfused with a solution of calcium, oxalate and ATP, electron opaque calcium oxalate was accumulated in vesicles subjacent to the sarcolemn. Salyrgan, an inhibitor of the extra ATPase, which is related to calcium uptake, blocks the calcium oxalate storage. This suggests a function of these vesicles similar to that of sarcoplasmic reticulum in vertebrate striated muscle, which consists in regulating the contraction relaxation cycle by varying the calcium concentration in the sarcoplasm.
    Notes: Zusammenfassung Muskelfasern des vorderen Byssusretraktors vonMytilus edulis wurden mit einem Medium aus Calcium, Oxalat und ATP behandelt und anschließend elektronenmikroskopisch untersucht. Dabei konnten Calciumoxalatkristalle, die eine lokale Anreicherung von Calcium anzeigen, ausschließlich im Inneren von randständigen Vesikeln festgestellt werden. Salyrgan, ein Hemmstoff für die mit dem Calciumtransport verbundene sogenannte Extra-ATPase, verhinderte die Calciumakkumulation völlig. Die Befunde deuten darauf hin, daß die Vesikel durch eine Veränderung der Calciumkonzentration im Sarcoplasma die Muskelaktivität zu regulieren vermögen, und damit funktionell dem sarcoplasmatischen Retikulum der quergestreiften Muskeln entsprechen.
