    Notes: Zygotic embryos of leek (Allium ampeloprasum L.) were isolated from mature seeds of different cultivars, selfings and full-sib families. The embryos were cultured on callus induction and shoot regeneration medium and employed to study several parameters: percentage of embryos forming calluses, percentage of embryos forming compact calluses, callus weight, percentage of regenerating calluses, numbers of shoot primordia and numbers of regenerated shoots. Differences between cultivars and selfings were found for most parameters studied. For all cultivars all parameters, except callus weight, decreased after one generation of selfing. Compact callus types enhanced primordia formation and shoot regeneration. Genetic characteristics of callus development and plant regeneration were studied in a 4 × 4 diallel cross. Analysis of variance revealed significant differences between full-sib families. The diallel analysis showed that additive gene effects were significant for all parameters. The predominance of additive gene effects indicated high narrow-sense heritability. Breeding for an increased number of regenerated shoots was successful.

    Notes: Summary A high frequency plant regeneration system via organogenesis and somatic embryogenesis was established with callus cultures derived from mature zygotic embryos of different leek genotypes (Allium ampeloprasum L.). Four different callus types with varying morphogenetic potential were obtained. Relatively high concentrations of the auxin 2,4-dichlorophenoxy-acetic acid reduced callus weight and subsequent shoot regeneration and primordia formation of the callus. Shoot regeneration and primordia formation of the callus decreased after prolonged subculture on media containing 2,4-dichlorophenoxy acetic acid. A callus growth period of six weeks on Murashige and Skoog medium with 0.25–0.5 mg l-1 2,4-dichlorophenoxy acetic acid showed the highest rate of shoot regeneration after transfer of callus to regeneration medium with 1 mg l-1 kinetin. Differences between leek genotypes in callus type, callus weight, shoot regeneration and primordia formation were observed. Histological observations showed that plant regeneration took place, both via the pathway of somatic embryogenesis and organogenesis.

    Notes: Summary A fast neutron irradiation experiment using two different dose rates has been performed on the potato variety Desiree in order to obtain chimera-free mutants. Both tubers and tuber parts (cross sections) were de-eyed at different moments before and after irradiation. Adventitious sprouts successively developing on the callus in the holes of excised eyes were separated from the treated tubers and planted individually. So called ‘grouped’ sprouts were separated. Results indicated that (practically) chimera-free mutants can be obtained by de-eying tuber parts shortly before irradiation, by applying relatively high dose-rates and by taking adventitious sprouts which develop not earlier than three months after irradiation. Separation of the grouped sprouts demonstrated that each part originated from a single original cell.

    Notes: Summary Dihaploids of the potato variety Desirée were used to obtain preliminary information about the doses and dose rates most suitable for mutation breeding purposes. The sprouts that developed after irradiation were removed in succession and then planted separately in pots. A broad distinction of 3 successive groups of sprout production can be made. The low dose rate object (50 rad/min) demonstrated an increase in overall as well as in uniform mutation frequency in the successive groups, while the high dose rate group (1000 rad/min) showed a converse behaviour with regard to overall mutation frequency.

    Notes: Summary In the period 1980–1984 about 70000 selfed seeds of the petunia cultivar Blue Bedder were treated with the chemical mutagens ethidium bromide (EB) and ethylnitroso urea (ENU) with the intention to induce (new sources of) cytoplasmic male sterility (CMS). This work resulted in 27 stable male sterile MS-plants, all derived from the ENU-treatments. Crosses with the normal fertile type and with material carrying restorer genes for the common type of CMS in petunia were made. Results obtained from some MS-plants raise a presumption of genic male sterility, probably of the dominant type, having been induced. More detailed investigations are underway.

    Notes: Summary It was investigated what potentialities for mutation breeding of potato are offered by using adventitious sprouts that arise in vitro from leaf explants (rachis, petiole, leaflet-disc) after X-irradiation. Mutation frequency and chimerism were studied in subterranean and aerial parts in three vegetative generations (vM1, vM2, vM3). Plants obtained from irradiated series produced a very high mutation frequency, a wide mutation spectrum and a very low rate of chimerism. Mutations were observed also in control series, especially in plants derived from the rachis and petiole explant group.

    Notes: Summary While investigating the formation of adventitious sprouts from L1 tissue in potato as part of our work on potato mutation, we tested the ‘Jørgensen’ method. This unpublished method of raising sprouts at the base of plantlets that have been severed from tubers was tried with our red-splashed, stable monectochimeric clones B165 and EM52. The changes of tuber colour from fully-grown (adventitious?) sprouts allowed us to draw the following conclusions: 1. The new sprouts cannot be of epidermal origin, since no yellow-tubered progeny were obtained. 2. The many red-splashed tubers that were obtained must be of axillary origin, i.e. from axillary buds at the edges of the fracture. 3. The red tubers may be of adventitious origin, but uncovering via layer perforation is not excluded the more so as a regeneration time of three weeks is rather short for adventitious origin.
