    Abstract: This book explains the fundamental characteristics and biofunctionality of graphene-based nanomaterials and provides up-to-date information on the full range of their biomedical applications. An introductory section gives an overview of the chemical composition and physical properties of graphene and its derivatives as well as their potential toxicity and biosafety. Detailed attention is then devoted to the potential of multifunctional graphene-based nanomaterials (MFGNs) to direct the differentiation of stem cells into specific lineages and induce tissue regeneration. Here, individual chapters address the application of MFGNs for the purposes of neurogenesis, osteo- and chrondrogenesis, myogenesis, and wound healing. Subsequent sections focus on the capability of MFGNs as agents for drug delivery, bioimaging, theranostics, and therapeutics as well as their effectiveness as biomimetic platforms for nanobiosensors, biochips, medical devices, and dental applications. The book will be essential reading for graduate students, scientists, and engineers in any of the biomedical research fields in which efforts are being made to utilize novel MFGN-incorporated composite materials and develop functional devices based on them.

    Notes: Stabilization of the fibrous protein collagen is important in biomedical applications. Thisstudy investigated the efficacy of degradation control of collagen using (-)-epigallocatechin-3-Ogallate(EGCG). EGCG treatment of collagen in solid state was carried out and collagen spongesproduced were characterized by measuring the physicochemical properties such as gel fraction, theenzymatic degradability and cytocompatibility. According to gel fraction, EGCG-treated spongesshowed the increase of insolubility compared to intact sponges. It showed that EGCG played a rolein a crosslinker of collagen. Through in vitro enzymatic degradation test, EGCG-treated collagensponges showed significant enhancement of resistance to collagenase in comparison with 25 mMEDC-treated collagen sponges. Also, cell proliferation assays showed that 40 mM EGCG-treatedcollagen sponges exhibited similar cytocompatibility properties compared with tissue culture plate.In summary, EGCG treatment of collagen sponges increased the stability of collagen. Therefore,crosslinking of collagen based scaffold with EGCG imparted more desirable properties, making itmore applicable for use as a scaffold in tissue engineering applications

    Notes: In this study, such behaviors of vascular smooth muscle cells (VSMCs), as proliferationand migration, with serum stimulation were investigated onto (−)-epigallocatechin-3-O-gallate(EGCG)-blended poly(L-lactide-co-ε-caprolactone, PLCL) copolymers (EGCG-b PLCL). VSMCswere primarily cultured from rat aorta, and EGCG-b PLCL films were fabricated by mixing PLCLwith EGCG. The proliferation of VSMCs cultured onto EGCG-b PLCL film was significantlysuppressed in spite of serum induction. Moreover, recovery of denuded area by VSMCs receivingconditioned media obtained from EGCG-b films was completely inhibited, whereas VSMCs ontointact films migrated into denuded area in response to serum showing essentially complete recovery.These results suggest that inhibition in the behaviors of serum-stimulated VSMCs may be mediatedthrough the anti-proliferative effects of EGCG released from polymer films, and EGCG-b polymerscan be applied for fabricating an EGCG-eluting vascular stent

    Notes: In this study, human dermal fibroblast behaviors onto non-porous PLGA (75:25) films immobilized with 1, 10 and 100 µg/ml collagen (CN) or fibronectin (FN) were investigated according to different cell-seeding densities (1,000, 10,000 and 100,000 cells/ml). Cell attachment and proliferation were assessed using water soluble tetrazolium salt. The results indicated that 1 µg/ml of FN-immobilized PLGA film demonstrated significantly (p 〈 0.05) superior cellular attachment to the intact PLGA film after 4 hr of incubation. Moreover, the number of attached cells was shown to be directly proportional to that of initially seeded cells. After 48 hr, the cells showed significantly (p 〈 0.05) higher proliferation onto 1 or 10 µg/ml of FN-immobilized PLGA films than onto other PLGA films, regardless of the initial cell-seeding density. In terms of CN-immobilization, cell proliferation was appreciably increased but it was relatively lower than FN-immobilization. These results suggested that ECM-immobilization can enhance the cell affinity of hydrophobic scaffolds and be used to potential applications for tissue engineering by supportingcell growth

    Notes: b-glucan, an immunomodulator, can selectively enhance the immunobiologicalactivities of neutrophils and macrophages without stimulating proinflammatory cytokine production. Biologic response modifiers, like beta-glucan, will modulate immunity, modify neoplastic disease and increase resistance to microbial challenge. Therefore, beta-glucan polymers can be applied in bone induction and regeneration model and have a possibility of association with bone morphogenetic protein (BMP) because of tissue-regenerative and antimicrobial effects of those polymers. In this report, we studied an E. coli expression system for BMP-7 production and the biological activities of b-glucan associated with BMP-7. The proliferation of MC3T3-E1 osteoblastic cells was enhanced by treatment with Aureobasidium b-glucan, while neither mushroom b-glucan nor barley b-glucan increased the cell proliferation. Mushroom b-glucan alone or associated with BMP-7 increased alkaline phosphatase (ALP) activity of MC3T3-E1 cells, one of the osteoblast phenotype markers, but the other b-glucans did not affect ALP activityof the cells. In mineralization assay, a highly significant increase in nodular staining was observed in cultures treated with both mushroom and Aureobasidium b-glucans in the presence of BMP-7 compared with nontreated controls, while barley b-glucan showed a significant decrease in nodule number compared with cultures treated only with BMP-7

    Notes: The potential protective roles played by green tea compounds (GTPCs) against reactive oxygen species-induced oxidative stress in cultured fetal human dermal fibroblasts (fHDFs) were investigated according to cell viability measurement methods, such as fluorescence double staining followed by flow cytometry (FCM), MTT assay and crystal violet uptake. Oxidative stress was induced in the fHDFs, either by adding 50 mM H2O2 or by the action of 40 U/L xanthine oxidase(XO) in the presence of xanthine (250 µM). FCM analysis was the most suitable to show that both treatments produced a significant (p 〈 0.05) reduction in the fHDF viability, attributed to its high sensitivity. On the microscopic observations, the cell death with necrotic morphology was appreciably induced by both treatments. These oxidative stress-induced damages were significantly (p 〈 0.05) prevented by pre-incubating the fHDFs with 200 µg/ml GTPC for 1 h. These results suggest that GTPC can act as a biological antioxidant in a cell culture experimental model and prevent oxidative stress-induced cytotoxicity in cells

    Description: Purpose Biological age (BA) has recently emerged as a substitute for chronological age (CA), and many subjects seek to optimally control their BA. However, in South Korea, no study has adequately explored factors that affect BA, although individual health management is essential to preventing chronic diseases. In the present study, we focus on the use of health information, in particular nutrition facts, to control BA. Methods We used data from the Korea National Health and Nutrition Examination Surveys (2010–2015; 26,914 eligible participants) using BA and age differences as outcome variables. We used multiple linear regression to explore the relationship between the use of nutrition data and differences in BA after adjusting for covariates. In addition, we used multiple linear regression to examine subgroup differences in such relationships. Results 12.8% of males and 27.5% of females used nutrition facts when deciding which foods to purchase. The more attention paid to such facts, the lower the BA and BA differences in both males and females (males: β = −2.646, females: β = −2.787, p 〈 0.05, for BA; males: β = −1.181, females, β = −2.161, p 〈 0.05, for BA differences). However, BA differences were more significant in subjects with chronic disease, obesity, and/or a family history of chronic disease. Conclusion High-level awareness of and active use of nutrition facts permitted effective self-management in preventing chronic disease and improving BA, particularly in subjects at higher risk for chronic disease. Thus, considering nutrition facts when deciding what to purchase is important.

Studies are needed to suggest evidence and arouse public awareness of the negative effects of abnormal sleep duration. In this study, we investigated the relationship between biological age (BA) and sleep duration. (2) Methods: We used data from the Korea National Health and Nutrition Examination Surveys (KNHANES V-VI; 2010–2015, which is an annually cross-sectional study including 29,309 participants). We performed multiple linear regression to investigate the associations between sleep duration and differences in BA and chronological age (CA). (3) Results: A total of 14.22% of respondents had short sleep duration (less than 6 h per day) and 7.10% of respondents had long sleep duration (more than 8 h per day). People with long sleep duration had a positive correlation with difference between BA and CA (>8 h per day, β = 1.308, p-value = 0.0001; ref = 6~8 h per day, normal). Short sleep duration had an inverse trend with the difference, although the result was not statically significant. Associations were greater in vulnerable populations, such as low income, obese, or people with chronic diseases. (4) Conclusions: Excess sleep duration that is greater than the normal range was associated with increased BA. In particular, such relationships that are related to worsening BA were greater in patients with low income, obesity, and chronic diseases. Based on our findings, healthcare professionals should also consider the negative effects of excess sleep, not only insufficient sleep. Alternatives for controlling optimal sleep duration should be reviewed, especially with vulnerable populations.
