Untersuchungen an Ketosen, 2. - Spirocyclische Dihydropyranone aus D-FructoseEinfache Reaktionsfolgen wurden entwickelt zur Überführung von D-Fructose in eine Reihe spirocyclischer Dihydropyrandiole mit 2S,3S- (10, 11), 2R,5S- (17, 18) und 2S,3R-Konfiguration (19) und Dihydropyranone 4-7 mit 2S- und 2R-Chiralität. Aufgrund der rigiden Verknüpfung von pyranoidem und Dioxolan-Ring über einen Spirokohlenstoff, was in Additionsreaktionen an die olefinische und/oder Carbonyl-Doppelbindung zu vorhersagbaren Regio- und Stereoselektivitäten führt, sind diese neuen Bausteine vielseitig verwendbare Sechs-Kohlenstoff-Synthons. - Schlüsselintermediate ihrer Darstellung sind die bekannten 3-O-Benzoyl- (8) und 3-O-Tosyl-Derivate der 1,2-O-Isopropyliden-D-fructopyranose sowie die erstmals erhaltenen 4,5-Di-O-mesyl- (9), 3,4-Di-O-tosyl- (15) und 3,4-Di-O-benzoyl-Analoga 23. Einführung der olefinischen Doppelbindung erfolgte durch reduktive Eliminierung mit Iodid/Zink in DMF (Tipson-Cohen-Verfahren) oder mittels Triphenylphosphan/Iod/Imidazol-Reagenzien, die der Carbonylgruppen durch PCC-Oxidation der entsprechenden Alkohole.

Expedient reaction sequences have been developed to convert D-fructose into a diverse collection of spirocyclic dihydropyrandiols with 2S,3S (10, 11), 2R, 5S (17, 18), and 2S,3R configuration (19), and of dihydropyranones 4-7 with 2S and 2R chirality. Due to the rigid attachment of pyranoid and dioxolane rings around a chiral spiro carbon which is to result in foreseeable regio-and stereo-selectivities on additions to the olefinic and/or carbonyl double bonds, they represent highly versatile six-carbon synthons. - Key intermediates for their obtention are the long-known 3-O-benzoyl (8) and 3-O-tosyl derivatives (12) of 1,2-O-isopropylidene-D-fructopyranose, as well as the novel 4,5-di-O-mesyl (9), the 3,4-di-O-tosyl (15), and the 3,4-di-O-benzoyl analogs 23. Introduction of olefinic unsaturation was effected with iodide/zinc in refluxing DMF (Tipson-Cohen procedure) or with triphenylphosphane/iodine/imidazole reagents, carbonyl groups were generated via PCC oxidation of the respective alcohols.
