    Description: We seek to understand how biophysical factors such as soil temperature (Ts), soil moisture (θ), and gross primary production (GPP) influence CO2 fluxes across terrestrial ecosystems. Recent advancements in automated measurements and remote-sensing approaches have provided time series in which lags and relationships among variables can be explored. The purpose of this study is to present new applications of continuous measurements of soil CO2 efflux (F0) and soil CO2 concentrations measurements. Here we explore how variation in Ts, θ, and GPP (derived from NASA's moderate-resolution imaging spectroradiometer [MODIS]) influence F0 and soil CO2 production (Ps). We focused on seasonal variation and used continuous measurements at a daily timescale across four vegetation types at 13 study sites to quantify: (1) differences in seasonal lags between soil CO2 fluxes and Ts, θ, and GPP and (2) interactions and relationships between CO2 fluxes with Ts, θ, and GPP. Mean annual Ts did not explain annual F0 and Ps among vegetation types, but GPP explained 73% and 30% of the variation, respectively. We found evidence that lags between soil CO2 fluxes and Ts or GPP provide insights into the role of plant phenology and information relevant about possible timing of controls of autotrophic and heterotrophic processes. The influences of biophysical factors that regulate daily F0 and Ps are different among vegetation types, but GPP is a dominant variable for explaining soil CO2 fluxes. The emergence of long-term automated soil CO2 flux measurement networks provides a unique opportunity for extended investigations into F0 and Ps processes in the near future.

    Description: We had continuously measured soil CO2 efflux (Rs) in a larch forest in northern Japan at hourly intervals for the snow-free period in 2003 with an automated chamber system and partitioned Rs into heterotrophic respiration (Rh) and autotrophic respiration (Rr) by using the trench method. In addition, we applied the soil CO2 concentration gradients method to continuously measure soil CO2 profiles under snowpack in the snowy period and to partition Rs into topsoil (Oa and A horizons) CO2 efflux (Ft) with a depth of 0.13 m and sub-soil (C horizon) CO2 efflux (Fc). We found that soil CO2 effluxes were strongly affected by the seasonal variation of soil temperature but weakly correlated with soil moisture, probably because the volumetric soil moisture (30–40% at 95% confidence interval) was within a plateau region for root and microbial activities. The soil CO2 effluxes changed seasonally in parallel with soil temperature in topsoil with the peak in late summer. On the other hand, the contribution of Rr to Rs was the largest at about 50% in early summer, when canopy photosynthesis and plant growth were more active. The temperature sensitivity (Q10) of Rr peaked in June. Under snowpack, Rs was stable until mid-March and then gradually increased with snow melting. Rs summed up to 79 gC m−2 during the snowy season for 4 months. The annual Rs was determined at 934 gC m−2 y−1 in 2003, which accounted for 63% of ecosystem respiration. The annual contributions of Rh and Rs to Rs were 57% and 43%, respectively. Based on the gradient approach, Rs was partitioned vertically into litter (Oi and Oe horizons) with a depth of 0.01–0.02 m, topsoil and sub-soil respirations with proportions of 6, 72 and 22%, respectively, on an annual basis. The vertical distribution of CO2 efflux was consistent with those of soil carbon and root biomass.

    Description: Ocean Drilling Program (ODP) Leg 164 recovered a number of large solid gas hydrate from Sites 994, 996, and 997 on the Blake Ridge. Sites 994 and 997 samples, either nodular or thick massive pieces, were subjected to laboratory analysis and measurements to determine the structure, molecular and isotopic composition, thermal conductivity, and equilibrium dissociation conditions. X-ray computed tomography (CT) imagery, X-ray diffraction, nuclear magnetic resonance (NMR), and Raman spectroscopy have revealed that the gas hydrates recovered from the Blake Ridge are nearly 100% methane gas hydrate of Structure I, cubic with a lattice constant of a = 11.95 ± 0.05 angström, and a molar ratio of water to gas (hydration number) of 6.2. The d18O of water is 2.67 per mil to 3.51 per mil SMOW, which is 3.5-4.0 heavier than the ambient interstitial waters. The d13C and dD of methane are -66 per mil to -70 per mil and -201 per mil to -206 per mil, respectively, suggesting that the methane was generated through bacterial CO2 reduction. Thermal conductivity values of the Blake Ridge hydrates range from 0.3 to 0.5 W/(m K). Equilibrium dissociation experiments indicate that the three-phase equilibrium for the specimen is 3.27 MPa at 274.7 K. This is almost identical to that of synthetic pure methane hydrate in freshwater.

    Notes: Abstract. The nucleotide sequence of the 18S rDNA coding gene in the ascomycetes parasitic fungus Isaria japonica contains a group I intron with a length of 379 nucleotides. The identification of the DNA sequence as a group I intron is based on its sequence homology to other fungal group I introns. Its group I intron contained the highly conserved sequence elements P, Q, R, and S found in other group I introns. Surprisingly, the intron sequence of I. japonica is more similar to that of Ustilago maydis than to the one found in Sclerotinia sclerotiorum. This is in contrast to the sequence identity found on the neighboring rDNA. This is an interesting finding and suggests a horizontal transfer of group I intron sequences.

    Notes: CaTiO3 thin films were prepared by a 193 nm ArF excimer laser deposition method. Epitaxial CaTiO3 thin films were obtained on SrTiO3 (100) substrate and YBa2Cu3Oy (001) deposited on MgO (100) single-crystal substrate. The grown CaTiO3 thin films orientated to (010) direction and the unit cell of CaTiO3 rotated 45° around the [100] axis of the SrTiO3 substrate. It also happened in CaTiO3/YBa2Cu3Oy/MgO system. The static dielectric constant was 120 at room temperature and reached 175 at lower temperatures.

    Notes: Nontypeable Haemophilus influenzae (NTHi) is a major cause of otitis media in children. We investigated whether intranasal immunization with a detoxified lipooligosaccharide-tetanus toxoid (dLOS-TT) conjugate vaccine would generate protective immunity against NTHi in a mouse model of nasopharyngeal clearance. The results demonstrated that intranasal immunization with dLOS-TT plus adjuvant cholera toxin (CT) significantly induced LOS-specific IgA antibodies in mouse external secretions, especially in nasal wash (90-fold), bronchoalveolar lavage fluid (25-fold), saliva (13-fold) and fecal extract (three-fold). LOS-specific IgA antibody-forming cells were also found in mucosal and lymphoid tissues with their highest numbers in the nasal passage (528 per 106 cells). In addition, the intranasal immunization elicited a significant rise in LOS-specific IgG (32-fold) and IgA (13-fold) in serum. For the immunized mice which had been challenged through the nose with 107 live NTHi strain 9274 cells, the vaccine group showed a significant reduction (74–77%) of NTHi, compared to that of control groups with CT alone or dLOS plus CT (P〈0.05). Negative correlations were found between bacterial counts and the levels of nasal wash IgA or IgG, saliva IgA and serum IgG. The clearance of five heterologous strains was investigated and revealed a significant clearance of strains 3198, 5657 and 7502 but not of strains 1479 and 2019. These data suggest that intranasal immunization with dLOS-TT vaccine elicits both mucosal and systemic immunity against NTHi and enhances bacterial clearance from nasopharynx in mice. Such a vaccine and vaccination regime may be applicable to humans with an appropriate formulation.
