Description: The growth factor progranulin (PGRN) has been implicated in embryonic development, tissue repair, tumorigenesis, and inflammation, but its receptors remain unidentified. We report that PGRN bound directly to tumor necrosis factor receptors (TNFRs) and disturbed the TNFalpha-TNFR interaction. PGRN-deficient mice were susceptible to collagen-induced arthritis, and administration of PGRN reversed inflammatory arthritis. Atsttrin, an engineered protein composed of three PGRN fragments, exhibited selective TNFR binding. PGRN and Atsttrin prevented inflammation in multiple arthritis mouse models and inhibited TNFalpha-activated intracellular signaling. Collectively, these findings demonstrate that PGRN is a ligand of TNFR, an antagonist of TNFalpha signaling, and plays a critical role in the pathogenesis of inflammatory arthritis in mice. They also suggest new potential therapeutic interventions for various TNFalpha-mediated pathologies and conditions, including rheumatoid arthritis.〈br /〉〈br /〉〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3104397/" target="_blank"〉〈img src="https://static.pubmed.gov/portal/portal3rc.fcgi/4089621/img/3977009" border="0"〉〈/a〉   〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC3104397/" target="_blank"〉This paper as free author manuscript - peer-reviewed and accepted for publication〈/a〉〈br /〉〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Tang, Wei -- Lu, Yi -- Tian, Qing-Yun -- Zhang, Yan -- Guo, Feng-Jin -- Liu, Guang-Yi -- Syed, Nabeel Muzaffar -- Lai, Yongjie -- Lin, Edward Alan -- Kong, Li -- Su, Jeffrey -- Yin, Fangfang -- Ding, Ai-Hao -- Zanin-Zhorov, Alexandra -- Dustin, Michael L -- Tao, Jian -- Craft, Joseph -- Yin, Zhinan -- Feng, Jian Q -- Abramson, Steven B -- Yu, Xiu-Ping -- Liu, Chuan-ju -- AI43542/AI/NIAID NIH HHS/ -- AR040072/AR/NIAMS NIH HHS/ -- AR050620/AR/NIAMS NIH HHS/ -- AR053210/AR/NIAMS NIH HHS/ -- GM061710/GM/NIGMS NIH HHS/ -- R01 AI030165/AI/NIAID NIH HHS/ -- R01 AI030165-20/AI/NIAID NIH HHS/ -- R01 GM061710/GM/NIGMS NIH HHS/ -- R01 GM061710-08/GM/NIGMS NIH HHS/ -- New York, N.Y. -- Science. 2011 Apr 22;332(6028):478-84. doi: 10.1126/science.1199214. Description: A long-term investigation, which covered 6 sampling campaigns over one year, was carried out to evaluate the occurrence, removal and risk of nonylphenol (NP), bisphenol A (BPA) and triclosan (TCS) in two full-scale sewage treatment plants (STPs) in Guangdong, South China. The target compounds were detected in every sample. The influent concentrations of NP were in the ppb level, while TCS in the ppt level. The seasonal variation in the influent was observed due to human consumption and the rainfall. Domestic sewage discharge was an important source of TCS to the STPs. Three kinds of endocrine disrupting compounds (EDCs) could not be completely removed in the STPs, and the activated sludge treatment process contributed to most of the removal of compounds. In addition, the oxic tank and the anaerobic tank both showed good removal performances for target EDCs. According to the results produced from the calculation of the risk quotient, in the STP1 effluents TCS could pose a high ri...

Description: Staphylococcus aureus produces hospital- and community-acquired infections, with methicillin-resistant S. aureus posing a serious public health threat. The golden carotenoid pigment of S. aureus, staphyloxanthin, promotes resistance to reactive oxygen species and host neutrophil-based killing, and early enzymatic steps in staphyloxanthin production resemble those for cholesterol biosynthesis. We determined the crystal structures of S. aureus dehydrosqualene synthase (CrtM) at 1.58 angstrom resolution, finding structural similarity to human squalene synthase (SQS). We screened nine SQS inhibitors and determined the structures of three, bound to CrtM. One, previously tested for cholesterol-lowering activity in humans, blocked staphyloxanthin biosynthesis in vitro (median inhibitory concentration approximately 100 nM), resulting in colorless bacteria with increased susceptibility to killing by human blood and to innate immune clearance in a mouse infection model. This finding represents proof of principle for a virulence factor-based therapy against S. aureus.〈br /〉〈br /〉〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2747771/" target="_blank"〉〈img src="https://static.pubmed.gov/portal/portal3rc.fcgi/4089621/img/3977009" border="0"〉〈/a〉   〈a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC2747771/" target="_blank"〉This paper as free author manuscript - peer-reviewed and accepted for publication〈/a〉〈br /〉〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Liu, Chia-I -- Liu, George Y -- Song, Yongcheng -- Yin, Fenglin -- Hensler, Mary E -- Jeng, Wen-Yih -- Nizet, Victor -- Wang, Andrew H-J -- Oldfield, Eric -- AI07482/AI/NIAID NIH HHS/ -- GM073216/GM/NIGMS NIH HHS/ -- GM65307/GM/NIGMS NIH HHS/ -- HD051796/HD/NICHD NIH HHS/ -- R01 GM065307/GM/NIGMS NIH HHS/ -- R01 GM065307-07/GM/NIGMS NIH HHS/ -- R01 GM073216/GM/NIGMS NIH HHS/ -- R01 GM073216-29/GM/NIGMS NIH HHS/ -- New York, N.Y. -- Science. 2008 Mar 7;319(5868):1391-4. doi: 10.1126/science.1153018. Description: Numerical simulation was performed to investigate convective heat transfer at the impinging zone of non-immersed pipe jet with Fluent in the ultra fast cooling process, and some influencing factors on heat transfer such as jet direction-to-plate angle, jet pressure (velocity), nozzle-to-impinging plate spacing (height), nozzle interval, nozzle diameter, and surface temperature of the plate were analyzed and designed. The results show that, in the ultra fast cooling process, the reasonable jet direction-to-plate angle is between 30 ° and 45 °; the economic water pressure is generally about 0.6MPa no more than 1.0MPa; since nozzle-to-impinging plate spacing (height) has little influence on the heat transfer coefficient due to the large water speed, nozzle-to-impinging plate spacing (height) is generally adjusted between 300 and 400mm; the nozzle interval should be set properly according to the dynamic relationship of the nozzle diameter and the jet acting zone, besides the maximum...

Description: We report on the validation of profiles from the MIPAS-Envisat on-line processor of ESA version 4.61 with correlative measurements derived from MIPAS-STR onboard the high-altitude aircraft M55-Geophysica. The validation is made for the July 22, 2002, orbit 2051 in the region of the campaign base in Forli, Italy and for several orbits in February / March 2003 around Kiruna, northern Sweden. This paper includes a careful comparison of the MIPAS-STR data with O3 sondes and with in-situ measurements from the Geophysica obtained during ascent, descent and occasional dives. To obtain accurate N2O and CH4 profiles close to the MIPAS-Envisat tangent points we used the MIPAS-STR measurements of CFC-11 and CFC-12 and correlations obtained by the HAGAR instrument.

Notes: We present the results of a combined He atom and x-ray diffraction study of CH3(CH2)n−1SH monolayers self assembled on Au(111) surfaces. By combining these two complementary probes, we have characterized both the surface and the interior structure of the monolayers. In both cases, we find the same structure containing four molecules per unit mesh. However, we demonstrate that there are significant differences in both the diffraction linewidths and the dependence of the linewidth upon chain length for these two techniques.
