    Notes: Stable isotopes, 15N and 18O, have been used as tracers to differentiate a contaminant nitrate plume emanating from a single domestic septic system, in a ground-water system characterized by high and similar nitrate content outside and inside of the contaminant plume. A good delineation of the nitrate plume of septic origin was obtained using 15 N analysis in nitrate. The 15N content ofthe nonplunie nitrate is in agreement with the sources of nitrate: solid cattle manure, synthetic fertilizer (NH4-NO3), and soil organic nitrogen, at the study site. 18O analysis in nitrate did not provide enough isotopic contrast to permit separation of nitrate derived from the septic system and that in the surrounding ground water, derived from agricultural fertilizer sources. 18O data indicated that nitrification of ammonium is the main process responsible for formation of nitrate at the study site. 18O in ground water clearly delineated the ground-water plume associated with the septic system and suggest that this tracer should be considered in studies related with contaminant plumes of different origin.

    Notes: Although exogenous electric fields have been reported to influence the orientation of plant root growth, reports of the ultimate direction of differential growth have been contradictory. Using a high-resolution image analysis approach, the kinetics of electrotropic curvature in Vigna mungo L. roots were investigated. It was found that curvature occurred in the same root toward both the anode and cathode. However, these two responses occurred in two different regions of the root, the central elongation zone (CEZ) and distal elongation zone (DEZ), respectively. These oppositely directed responses could be reproduced individually by a localized electric field application to the region of response. This indicates that both are true responses to the electric field, rather than one being a secondary response to an induced gravitropic stimulation. The individual responses differed in the type of differential growth giving rise to curvature. In the CEZ, curvature was driven by inhibition of elongation, whereas curvature in the DEZ was primarily due to stimulation of elongation. This stimulation of elongation is consistent with the growth response of the DEZ to other environmental stimuli.

    Notes: In an earlier study (Evans, Ishikawa & Estelle 1994, Planta 194, 215–222) we used a video digitizer system to compare the kinetics of auxin action on root elongation in wild-type seedlings and seedlings of auxin response mutants of Arabidopsis thaliana (L.) Heynh. We have since modified the system software to allow determination of elongation on opposite sides of vertical or gravistimulated roots and to allow continuous measurement of the angle of orientation of sequential subsections of the root during the response. We used this technology to compare the patterns of differential growth that generate curvature in roots of the Columbia ecotype and in the mutants axr1-3, axr1-12 and axr2, which show reduced gravitropic responsiveness and reduced sensitivity to inhibition by auxin. The pattern of differential growth during gravitropism differed in roots of wild-type and axr1 seedlings. In wild-type roots, initial curvature resulted from differential inhibition of elongation in the distal elongation zone (DEZ). This was followed by an acceleration of elongation along the top side of the DEZ. In roots of axr1-3, curvature resulted from differential stimulation of elongation whereas in roots of axr1-12 the response was variable. Roots of axr2 did not exhibit gravitropic curvature. The observation that the pattern of differential growth causing curvature is dramatically altered by a change in sensitivity to auxin is consistent with the classical Cholodny-Went theory of gravitropism which maintains that differential growth patterns induced by gravistimulation are mediated primarily by gravi-induced shifts in auxin distribution. The new technology introduced with this report allows automated determination of stimulus response patterns in the small but experimentally popular roots of Arabidopsis.

    Notes: Although the effects of gravity on root growth are well known and interactions between light and gravity have been reported, details of root phototropic responses are less documented. We used high-resolution image analysis to study phototropism in primary roots of Zea mays L. Similar to the location of perception in gravitropism, the perception of light was localized in the root cap. Phototropic curvature away from the light, on the other hand, developed in the central elongation zone, more basal than the site of initiation of gravitropic curvature. The phototropic curvature saturated at approximately 10 µmol m−2 s−1 blue light with a peak curvature of 29 ± 4°, in part due to induction of positive gravitropism following displacement of the root tip from vertical during negative phototropism. However, at higher fluence rates, development of phototropic curvature is arrested even if gravitropism is avoided by maintaining the root cap vertically using a rotating feedback system. Thus continuous illumination can cause adaptation in the signalling pathway of the phototropic response in roots.

    Notes: Summary In an effort to examine the specificity of the auxin transport system, the movement of a variety of growth substances and of auxin analogues through corn coleoptile sections was measured in both the basipetal and acropetal directions. In contrast to the basipetal, polar transport of the auxins indoleacetic acid (IAA) and 2,4-dichlorophenoxyacetic acid, no such movement was found for benzoic acid or for gibberellin A1. A comparison of the α- and β-isomers of naphthaleneacetic acid showed that the growth-active α-form is transported, but not the inactive β-analogue. Both the dextro (+) and leavo (-) isomer of 3-indole-2-methylacetic acid showed the basipetal movement characteristic of IAA, the dextro isomer being more readily transported than the (-)-form. In this instance, too, the transport was roughtly proportional to the growth promoting activity. The antiauxin p-chlorophenoxyisobutyric acid inhibited auxin transport as it inhibited auxin-induced growth. These results agree with the hypothesis that processes involved in auxin transport are closely linked to or even identical with the primary auxin action.

    Notes: Summary Calcium has been implicated as a key component in the signal transduction process of root gravitropism. We measured cytoplasmic free calcium in protoplasts isolated from the elongation zone and cap of primary roots of light-grown, vertically oriented seedlings ofZea mays L. Protoplasts were loaded with the pentapotassium salts of fura-2 and indo-1 by incubation in acidic solutions of these calcium indicators. Loading increased with decreasing pH but the pH dependence was stronger for indo-1 than for fura-2. In the case of fura-2, loading was enhanced only at the lowest pH (4.5) tested. Dyes loaded in this manner were distributed predominantly in the cytoplasm as indicated by fluorescence patterns. As an alternative method of loading, protoplasts were incubated with the acetoxymethylesters of fura-2 and indo-1. Protoplasts loaded by this method exhibited fluorescence both in the cytoplasm and in association with various organelles. Cytoplasmic calcium levels measured using spectrofluorometry, were found to be 160±40 nM and 257±27 nM, respectively, in populations of protoplasts from the root cap and elongation zone. Cytoplasmic free calcium did not increase upon addition of calcium to the incubation medium, indicating that the passive permeability to calcium was low.

    Notes: Research in improved materials and methods for internal fixation has centered on internal fixators made of bioabsorbable materials such as polylactic acid, polyglycolic acid, and polyparadioxanone. These materials have two problems: the first is a postoperative complication related to a delayed inflammatory response; and the second is low strength characteristics. An alternative material developed to alleviate these problems is a composite of phosphate glass fibers embedded in the polymer polycaprolactone, referred to as PCL. In this study, intramedullary pins made of PCL were compared to stainless steel pins in a rabbit humerus osteotomy model. Specimens were harvested at 0, 6, and 12 weeks postoperatively, radiographs and mechanical testing to failure were performed at each time interval, and tissue was examined microscopically at 6 and 12 weeks. Histologic results showed PCL pins to be well tolerated with minimal inflammation around the pin. Mechanical testing revealed the PCL fixation to be weaker initially than the stainless steel fixation. There was significant stress shielding of stainless-steel-healed rabbit humeri when compared to the PCL/bone humeri. All osteotomies immobilized with PCL healed with abundant periosteal callus production. © 1997 John Wiley & Sons, Inc. J Biomed Mater Res, 36, 536-541, 1997.
