    Notes: Thc crystal structure of an α-chymotrypsin inhibitor (P6122; a = 61.4, c = 210.9 Å) isolated from winged bean (Psophocarpus. tetragonolobus) seeds has been determined at 2.95 Å resolution by the molecular-replacement method using the 2.6 Å coordinates of Erythrina trypsin inhibitor (ETI) as the starting model (57% sequence homology). This protease inhibitor, WCI, belongs to the Kunitz (STI) family and is a single polypeptide chain with 183 amino-acid residues having a molecular weight of 20 244 Da. Structure refinement with RESTRAIN and X-PLOR has led to a crystallographic R factor of 19.1% for 3469 observed reflections (I 〉 2σ) in the resolution range 8–2.95 Å. A total of 56 water molecules have been incorporated in the refined model containing 181 amino-acid residues. In the refined structure the deviations of bond lengths and bond angles from ideal values are 0.015 Å and 2.2°, respectively. The inhibitor molecule is spherical and consists of 12 antiparallel β-strands with connecting loops arranged in a characteristic folding (a six-stranded β-barrel and a six-stranded lid on one hollow end of the barrel) common to other homologous serine protease inhibitors in the Kunitz (STI) family as well as to some non-homologous proteins like interleukin-lα and interleukin-lβ. In the structure the conformation of the protruding reactive-site loop is stabilized through hydrogen bonds mainly formed by the side chain of Asnl4, which intrudes inside the cavity of the reactive-site loop, with the side-chain and main-chain atoms of some residues in the loop region. A pseudo threefold axis exists parallel to the barrel axis of the structure. Each of the three subdomains comprises of four β-strands with connecting loops.

    Notes: We describe quantitative imaging of the sheet resistance of metallic thin films by monitoring frequency shift and quality factor in a resonant scanning near-field microwave microscope. This technique allows fast acquisition of images at approximately 10 ms per pixel over a frequency range from 0.1 to 50 GHz. In its current configuration, the system can resolve changes in sheet resistance as small as 0.6 Ω/(square, open) for 100 Ω/(square, open) films. We demonstrate its use at 7.5 GHz by generating a quantitative sheet resistance image of a YBa2Cu3O7−δ thin film on a 5 cm diam sapphire wafer. © 1998 American Institute of Physics.

    Notes: We describe the use of a cryogenic near-field scanning microwave microscope to image microwave electric fields from superconducting and normal-metal microstrip resonators. The microscope employs an open-ended coaxial probe and operates from 77 to 300 K in the 0.01–20 GHz frequency range with a spatial resolution of about 200 μm. We describe the operation of the system and present microwave images of Cu and Tl2Ba2CaCu2O8 microstrip resonators, showing standing wave patterns at the fundamental and second harmonic frequencies. © 1998 American Institute of Physics.

    Notes: A number of isolates of the cellular slime mold, Acrasis rosea, differed from each other in the range of microorganisms on which they could grow and fruit. In a test for the possible occurrence of sexual reproduction and genetic recombination, two of the most widely divergent of these physiological races were studied further. Mixed colonies were obtained in agar plate cultures of Rhodotorula mucilaginosa, a yeast utilized by both. When spores from mature fruiting bodies or amoebae migrating out from the mixed colonies were isolated singly, regrown on Rhodotorula, and tested on a critical spectrum of food microorganisms, about 3-4% proved to be distinct from the original two strains. Although some of these new types remained stable for several transfers, they eventually reverted to one or both of the parental types. The possibilities of explaining the neotypes on the basis of diploidy, heterokaryosis, or mixed cytoplasmic effects are discussed. The occurrence of cell anastomoses and occasional binucleate cells favors the latter two possibilities.

    Notes: SYNOPSIS. Deoxyribonucleic acids (DNAs) isolated from 12 representatives of cellular slime molds grown in monoxenic cultures were analyzed by density gradient centrifugation in CsC1. The unique contribution of the DNA of each food organism was subtracted and the DNA base composition of each cellular slime mold determined. The guanine plus cytosine contents range 22–37 moles % within the representatives of the order. Minor bands (satellite bands) of DNA have been observed in preparations from Dictyostelium spp. and from Polysphondylium pallidum.

    Notes: Application of picosecond and femtosecond laser pulses to the controlled ablation of materials represents a relatively unexplored yet important topic in laser processing. Such ultrashort pulses are of potential value in areas of thin-film deposition, micromachining, and surgical procedures. We report here some early results of systematic studies being done from the femtosecond to the nanosecond regime, as an assessment of the problems and benefits associated with various laser pulse durations and their use in processing optically absorbing media. Experimental data and theoretical results of computer simulations are presented and compared for the threshold energies of ablation in gold as a function of pulse width from 10 ns to 100 fs. This work is then extended to include further numerically computed results for gold and silicon on ablation rates, threshold surface temperatures, liquid thicknesses, and vaporization rates as a function of pulse duration throughout the ultrafast regime from tens of femtoseconds to a few hundred picoseconds. © 1995 American Institute of Physics.

    Notes: By scanning a fine open-ended coaxial probe above an operating microwave device, we image local electric fields generated by the device at microwave frequencies. The probe is sensitive to the electric flux normal to the face of its center conductor, allowing different components of the field to be imaged by orienting the probe appropriately. Using a simple model of the microscope, we are able to interpret the system's output and determine the magnitude of the electric field at the probe tip. We show images of electric field components above a copper microstrip transmission line driven at 8 GHz, with a spatial resolution of approximately 200 μm. © 1999 American Institute of Physics.

    Notes: We describe a technique for extracting topographic information using a scanning near-field microwave microscope. By monitoring the shift of the system's resonant frequency, we obtain quantitative topographic images of uniformly conducting metal surfaces. At a frequency of 9.572 GHz, our technique allows a height discrimination of about 55 nm at a separation of 30 μm. We present topographic images of uneven, conducting samples and compare the height response and sensitivity of the system with theoretical expectations. © 1998 American Institute of Physics.

    Notes: We describe near-field imaging of sample sheet resistance via frequency shifts in a resonant coaxial scanning microwave microscope. The frequency shifts are related to local sample properties, such as surface resistance and dielectric constant. We use a feedback circuit to track a given resonant frequency, allowing measurements with a sensitivity to frequency shifts as small as two parts in 106 for a 30 ms sampling time. The frequency shifts can be converted to sheet resistance based on a simple model of the system. © 1997 American Institute of Physics.

    Notes: Unhydrogenated diamondlike carbon (DLC) thin films have been deposited by laser ablation of graphite, using a high power Ti: sapphire solid state laser system. DLC films were deposited onto silicon substrates at room temperature with subpicosecond laser pulses, at peak intensities in the 4×1014–5×1015 W/cm2 range. A variety of techniques, including scanning and transmission electron microscopy (SEM and TEM), Raman spectroscopy, spectroscopic ellipsometry (SE), and electron energy loss spectroscopy (EELS) have been used to analyze the film quality. Smooth, partially transparent films were produced, distinct from the graphite target. Sp3 volume fractions were found to be in the 50%–60% range, with Tauc band gaps ranging from 0.6 to 1.2 eV, depending on laser intensity. Kinetic energies carried by the carbon ions in the laser induced plasma were measured through time-of-flight (TOF) spectroscopy. Their most probable kinetic energies were found to be in the 700–1000 eV range, increasing with laser intensity. © 1995 American Institute of Physics.
