    Description: Extract The early discovery of the Arbroath Field in 1969, and the Montrose and giant Forties fields in 1970, initiated intensive exploration of the Tertiary deep-marine play in the North Sea region. Numerous subsequent discoveries (Fig. 1), including Frigg (in 1971), Maureen (in 1973), Gannet (in 1973), Andrew (in 1974), Pierce (in 1976), Everest (in 1982), Alba (in 1984), Gryphon (in 1987), Nelson (in 1988), Harding (in 1988), Jotun (in 1994), Siri (in 1995) and Merganser (in 1995), demonstrate the success of this play and the geological diversity of Paleocene and Eocene systems present within the region. Although the North Sea Basin is now considered mature, with Cenozoic reservoirs well along their creaming curve (Vining et al. 2005), recent discoveries (e.g. the Catcher Field in 2010) highlight that potential still remains within intensively explored areas such as the Central North Sea, as well as in the less explored regions such as the Atlantic margin and the Norwegian Sea. The importance of these reservoirs is demonstrated by the large proportion of UK production to which they contribute, amounting to approximately 25% of all production from UK oil fields since 1975 on a barrel of oil equivalent (BOE) basis (Fig. 2).     Description: For many drugs, only racemic mixtures are available for clinical use. Because different stereoisomers of drugs often cause different physiological responses, the use of pure isomers could elicit more exact therapeutic effects. Differential complexation of a variety of drug stereoisomers by immobilized beta-cyclodextrin was investigated. Chiral recognition and racemic resolution were observed with a number of compounds from such clinically useful classes as beta-blockers, calcium-channel blockers, sedative hypnotics, antihistamines, anticonvulsants, diuretics, and synthetic opiates. Separation of the diastereomers of the cardioactive and antimalarial cinchona alkaloids and of two antiestrogens was demonstrated as well. Three dimensional projections of beta-cyclodextrin complexes of propanolol, which is resolved by this technique, and warfarin, which is not, are compared. These studies have improved the understanding and application of the chiral interactions of beta-cyclodextrin, and they have demonstrated a means to measure optical purity and to isolate or produce pure enantiomers of drugs.     Notes: Abstract The effectiveness of employing stationary phases composed of chemically bonded cyclodextrin molecules in the high performance liquid chromatographic separation of a variety of different types of compounds is summarized. Over one hundred compounds, including optical, geometrical, and structural isomers, diastereomers, and epimers were successfully separated from each other via use of beta- or gamma-cyclodextrin bonded phases and aqueous methanolic mobile phases. The mechanism of separation is based upon inclusion complex formation between the compounds being separated and the cyclodextrin molecules bonded to the stationary phase. The effects of temperature, mobile phase composition and flow rate upon the chromatographic selectivity and resolution are described. The results indicate that the cyclodextrin columns may be more versatile, flexible, and effective compared to the conventional normal or reversed phase columns.

    Notes: Summary High-performance liquid chromatographic and gas chromatographic methods were developed for the separation of unusual secondary aromatic amino acids. Amino acids containing 1,2,3,4-tetrahydroisoquinoline, 1,2,3,4-tetrahydronorharmane-1-carboxylic acid and 1,2,3,4-tetrahydro-3-carboxy-2-carboline moieties were synthetized in racemic or chiral forms. The high-performance liquid chromatography was carried out either on a teicoplanin-containing chiral stationary phase or on an achiral C18 column. In the latter case the diastereomers of the amino acids formed by precolumn derivatization with the chiral reagents 2,3,4,6-tetra-O-acetyl-β-D-glucopyranosyl isothiocyanate or 1-fluoro-2,4-dinitrophenyl-5-L-alanine amide were separated. The gas chromatographic analyses were based on separation on a Chirasil-L-Val column.

    Notes: Abstract. The toxicities and oxidative stress-inducing actions of (−)-nicotine and smokeless tobacco extract (STE), containing equivalent amounts of nicotine, were studied. Toxicities were determined by colony formation assays using Chinese hamster ovary (CHO) cells. Results indicated that nicotine is less toxic than smokeless tobacco extract that contained the same amount of nicotine. The generation of reactive oxygen species, following treatment with smokeless tobacco extract and nicotine, was assessed by measurement of changes in glutathione (GSH) and malondialdehyde (MDA) levels. CHO cells (5 × 105 cells/5 ml media) were incubated with 4, 0.8, and 0.08 mg of nicotine and STE containing the same amounts of nicotine. All preparations of smokeless tobacco extract significantly decreased GSH levels and increased MDA generation. However, 0.08 mg of nicotine treatment did not result in a significant change in GSH level, and only 4 mg of nicotine were sufficient to increase MDA generation. Addition of free radical scavenging enzymes, superoxide dismutase (SOD) and catalase (CAT), and an intracellular GSH precursor, N-acetyl-L-cysteine (NAC), replenished the GSH levels in nicotine-treated cells. GSH levels in cells exposed to smokeless tobacco extract containing 4 and 0.8 mg nicotine remained significantly lower than the control with the addition of SOD and CAT. However, co-addition of NAC with smokeless tobacco extract preparations returned the GSH levels to the control level. Lactate dehydrogenase (LDH) activities were measured in the media to establish the membrane damage following exposure to smokeless tobacco extract and nicotine. Treatment of cells with 4 mg nicotine caused a significant increase in LDH activity, which was returned to control level in the presence of the antioxidant enzymes and NAC. Smokeless tobacco extract did not change the LDH activity.
