    Description: 〈p〉Recent worldwide outbreaks of enterovirus 71 (EV71) have caused major epidemics of hand, foot, and mouth disease with severe neurological complications, including acute flaccid paralysis. EV71 is transmitted by the enteral route, but little is known about the mechanisms it uses to cross the human gastrointestinal tract. Using primary human intestinal epithelial monolayers, we show that EV71 infects the epithelium from the apical surface, where it preferentially infects goblet cells. We found that EV71 infection did not alter epithelial barrier function but did reduce the expression of goblet cell–derived mucins, suggesting that it alters goblet cell function. We also show that the intestinal epithelium responds to EV71 infection through the selective induction of type III interferons (IFNs), which restrict EV71 replication. Collectively, these findings define the early events associated with EV71 infections of the human intestinal epithelium and show that host IFN signaling controls replication in an IFN-specific manner.〈/p〉

    Description: Nitrous oxide (N 2 O) emissions within the US Corn Belt have been previously estimated to be 200-900% larger than predictions from emission inventories, implying that one or more source categories in bottom-up approaches are underestimated. Here we interpret hourly N 2 O concentrations measured during 2010 and 2011 at a tall tower using a time-inverted transport model and a scale factor Bayesian inverse method to simultaneously constrain direct and indirect agricultural emissions. The optimization revealed that both agricultural source categories were underestimated by the Intergovernmental Panel on Climate Change (IPCC) inventory approach. However, the magnitude of the discrepancies differed substantially, ranging from 42–58% and 200–525% for direct and indirect components, respectively. Optimized agricultural N 2 O budgets for the Corn Belt were 319 ± 184 (total), 188 ± 66 (direct), and 131 ± 118 Gg-N yr -1 (indirect) in 2010, versus 471 ± 326, 198 ± 80, and 273 ± 246 Gg-N yr -1 in 2011. We attribute the inter-annual differences to varying moisture conditions, with increased precipitation in 2011 amplifying emissions. We found that indirect emissions represented 41–58% of the total agricultural budget, a considerably larger portion than the 25–30% predicted in bottom-up inventories, further highlighting the need for improved constraints on this source category. These findings further support the hypothesis that indirect emissions are presently underestimated in bottom-up inventories. Based on our results, we suggest an indirect emission factor for runoff and leaching ranging from 0.014–0.035 for the Corn Belt, which represents an upward adjustment of 1.9–4.6 times relative to the IPCC and is in agreement with recent bottom-up field studies.

    Description: Tides exert a major control on the coastal zone by influencing high sea levels and coastal flooding, navigation, sediment dynamics and ecology. Therefore, any changes to tides have wide ranging and important implications. In this paper, we uniquely assess secular changes in 15 regularly used tidal levels (five high water, five low water and five tidal ranges), which have direct practical applications. Using sea level data from 220 tide gauge sites, we found changes have occured in all analysed tidal levels in many parts of the world. For the tidal levels assessed, between 36% and 63% of sites had trends significantly different (at 95% confidence level) from zero. At certain locations, the magnitude of the trends in tidal levels were similar to trends in mean sea level over the last century, with observed changes in tidal range and high water levels of over 5mm/yr and 2mm/yr respectively. More positive than negative trends were observed in tidal ranges and high water levels, and vice versa for low water levels. However we found no significant correlation between trends in mean sea level and any tidal levels. Spatially coherent trends were observed in some regions, including the north-east Pacific, German Bight and Australasia, and we also found that differences in trends occur between different tidal levels. This implies that analysing different tidal levels is important. Because changes in the tide are widespread and of similar magnitude to mean sea level rise at a number sites, changes in tides should be considered in coastal risk assessments.

    Description: Northern Europe was influenced by consecutive episodes of extreme winter weather at the start and end of the 2010 calendar year. A tripole pattern in North Atlantic sea surface temperature anomalies (SSTAs), associated with an exceptionally negative phase of the North Atlantic Oscillation (NAO), characterized both winter periods. This pattern was largely absent at the surface during the 2010 summer season; however equivalent sub-surface temperature anomalies were preserved within the seasonal thermocline throughout the year. Here, we present evidence for the re-emergence of late-winter 2009/10 SSTAs during the following early winter season of 2010/11. The observed re-emergence contributes toward the winter-to-winter persistence of the anomalous tripole pattern. Considering the active influence of the oceans upon leading modes of atmospheric circulation over seasonal timescales, associated with the memory of large-scale sea surface temperature anomaly patterns, the re-emergence of remnant temperature anomalies may have also contributed toward the persistence of a negative winter NAO, and the recurrence of extreme wintry conditions over the initial 2010/11 winter season.

    Description: Abstract Surface water flooding (SWF) is a recurrent hazard that affects lives and livelihoods. Climate change is projected to change the frequency of extreme rainfall events that can lead to SWF. Increasingly, data from Regional Climate Models (RCMs) are being used to investigate the potential water‐related impacts of climate change; such assessments often focus on broad‐scale fluvial flooding and the use of coarse resolution (〉12km) RCMs. However, high‐resolution (〈4km) convection‐permitting RCMs are now becoming available that allow impact assessments of more localised SWF to be made. At the same time, there has been an increasing demand for more robust and timely real‐time forecast and alert information on SWF. In the UK, a real‐time SWF Hazard Impact Model framework has been developed. The system uses 1km gridded surface runoff estimates from a hydrological model to simulate the SWF hazard. These are linked to detailed inundation model outputs through an Impact Library to assess impacts on property, people, transport and infrastructure for four severity levels. Here, a set of high‐resolution (1.5km and 12km) RCM data has been used as input to a grid‐based hydrological model over southern Britain to simulate Current (1996‐2009) and Future (~2100s; RCP8.5) surface runoff. Counts of threshold‐exceedance for surface runoff and precipitation (at 1‐, 3‐ and 6‐hour durations) are analysed. Results show that the percentage increases in surface runoff extremes, are less than those of precipitation extremes. The higher‐resolution RCM simulates the largest percentage increases, which occur in winter, and the winter exceedance counts are greater than summer exceedance counts. For property impacts the largest percentage increases are also in winter however, it is the 12km RCM output that leads to the largest percentage increase in impacts. The added‐value of high‐resolution climate model data for hydrological modelling is from capturing the more intense convective storms in surface runoff estimates.

    Description: The determination of aerosol direct radiative forcing over desert regions requires accurate information about the aerosol single-scattering albedo (SSA); however, the brightness of desert surfaces in the visible and near-IR range complicates the retrieval of aerosol optical properties using passive space-based measurements. Here we use the critical reflectance method to retrieve spectral aerosol absorption from space over North Africa, a desert region that is predominantly impacted by absorbing dust and biomass burning aerosol. We examine the sensitivity of the critical reflectance parameter to aerosol physical and optical properties that are representative of the region, and we find that the critical reflectance has low sensitivity to assumptions of aerosol size and refractive index for dust-like particles, except at scattering angles near 180°, which should be avoided with this method. We use our findings to retrieve spectral SSA from critical reflectance derived from Moderate Resolution Imaging Spectroradiometer (MODIS) reflectances in the vicinity of two Aerosol Robotic Network (AERONET) stations: Tamanrasset, in the Algerian Sahara, and Banizoumbou, in the Sahel. We retrieve lower aerosol SSAs at Banizoumbou, which is often impacted by dust-smoke mixtures, and higher SSAs at Tamanrasset, where pure desert dust is the dominant aerosol. Our results generally fall within the AERONET uncertainty envelopes, although at Banizoumbou we retrieve a spectral dependence different from that of AERONET. On the basis of our analysis, we expect to be able to retrieve SSA from critical reflectance for pure dust with an uncertainty of 0.02 and to provide spatial and spectral SSA information that will help reduce current uncertainties in the aerosol radiative forcing over desert regions.

    Description: Studies of predator?prey demographic responses and the physical drivers of such relationships are rare, yet essential for predicting future changes in the structure and dynamics of marine ecosystems. Here, we hypothesize that predator?prey relationships vary spatially in association with underlying physical ocean conditions, leading to observable changes in demographic rates, such as reproduction. To test this hypothesis, we quantified spatio-temporal variability in hydrographic conditions, krill, and forage fish to model predator (seabird) demographic responses over 18 years (1990?2007). We used principal component analysis and spatial correlation maps to assess coherence among ocean conditions, krill, and forage fish, and generalized additive models to quantify interannual variability in seabird breeding success relative to prey abundance. The first principal component of four hydrographic measurements yielded an index that partitioned ?warm/weak upwelling? and ?cool/strong upwelling? years. Partitioning of krill and forage fish time series among shelf and oceanic regions yielded spatially explicit indicators of prey availability. Krill abundance within the oceanic region was remarkably consistent between years, whereas krill over the shelf showed marked interannual fluctuations in relation to ocean conditions. Anchovy abundance varied on the shelf, and was greater in years of strong stratification, weak upwelling and warmer temperatures. Spatio-temporal variability of juvenile forage fish co-varied strongly with each other and with krill, but was weakly correlated with hydrographic conditions. Demographic responses between seabirds and prey availability revealed spatially variable associations indicative of the dynamic nature of ?predator?habitat? relationships. Quantification of spatially explicit demographic responses, and their variability through time, demonstrate the possibility of delineating specific critical areas where the implementation of protective measures could maintain functions and productivity of central place foraging predators. # doi:10.1890/13-1605.1

    Description: This study describes comprehensive polling of transcription start and termination sites and analysis of previously unidentified full-length complementary DNAs derived from the mouse genome. We identify the 5' and 3' boundaries of 181,047 transcripts with extensive variation in transcripts arising from alternative promoter usage, splicing, and polyadenylation. There are 16,247 new mouse protein-coding transcripts, including 5154 encoding previously unidentified proteins. Genomic mapping of the transcriptome reveals transcriptional forests, with overlapping transcription on both strands, separated by deserts in which few transcripts are observed.
