Structure and Synthesis of 5-Methoxy-5,6-dehydromethysticin, a Minor Component of Piper sanctumThe woody roots of the Mexican shrub Piper sanctum contain small amounts of a novel Kawa-type lactone with an extra oxygen function in the acetogenine part of the molecule. Its structure was established mainly by a study of the spectral data of the isolated substance itself as well as of those of its tetrahydroderivative cis-4. 4,5-Dimethoxy-6-(3,4-methylen-dioxystyryl)-2-pyronc (5b) was synthesized by hydroxylation [SeO2] of methysticin in position C—5, methylation [CH3J/AgO], and dehydrogenation [chloranil] and shown to be identical with the natural product.

Aus dem Wurzelholz von Piper sanctum wurde erstmalig ein Lacton vom Typus des Yangonins mit einer zweiten Methoxygruppe im Pyronteil des Moleküls isoliert. Die Konstitution 5b [4,5-Dimethoxy-6-(3,4-methylendioxystyryl)-2-pyron] wurde auf spektroskopischem Wege ermittelt und durch Synthese sowohl des Naturstoffes 5b selbst als auch seines Tetrahydro-derivates [5,6-cis-Dihydro-4,5-dimethoxy-6-(3,4-methylendioxyphenäthyl)-2-pyron (cis-4)] bestätigt.
