    Description: The hepatitis C virus (HCV) has developed a small membrane protein, p7, which remarkably can self-assemble into a large channel complex that selectively conducts cations. We wanted to examine the structural solution that the viroporin adopts in order to achieve selective cation conduction, because p7 has no homology with any of the known prokaryotic or eukaryotic channel proteins. The activity of p7 can be inhibited by amantadine and rimantadine, which are potent blockers of the influenza M2 channel and licensed drugs against influenza infections. The adamantane derivatives have been used in HCV clinical trials, but large variation in drug efficacy among the various HCV genotypes has been difficult to explain without detailed molecular structures. Here we determine the structures of this HCV viroporin as well as its drug-binding site using the latest nuclear magnetic resonance (NMR) technologies. The structure exhibits an unusual mode of hexameric assembly, where the individual p7 monomers, i, not only interact with their immediate neighbours, but also reach farther to associate with the i+2 and i+3 monomers, forming a sophisticated, funnel-like architecture. The structure also points to a mechanism of cation selection: an asparagine/histidine ring that constricts the narrow end of the funnel serves as a broad cation selectivity filter, whereas an arginine/lysine ring that defines the wide end of the funnel may selectively allow cation diffusion into the channel. Our functional investigation using whole-cell channel recording shows that these residues are critical for channel activity. NMR measurements of the channel-drug complex revealed six equivalent hydrophobic pockets between the peripheral and pore-forming helices to which amantadine or rimantadine binds, and compound binding specifically to this position may allosterically inhibit cation conduction by preventing the channel from opening.     Description: Mitochondria from many eukaryotic clades take up large amounts of calcium (Ca(2+)) via an inner membrane transporter called the uniporter. Transport by the uniporter is membrane potential dependent and sensitive to ruthenium red or its derivative Ru360 (ref. 1). Electrophysiological studies have shown that the uniporter is an ion channel with remarkably high conductance and selectivity. Ca(2+) entry into mitochondria is also known to activate the tricarboxylic acid cycle and seems to be crucial for matching the production of ATP in mitochondria with its cytosolic demand. Mitochondrial calcium uniporter (MCU) is the pore-forming and Ca(2+)-conducting subunit of the uniporter holocomplex, but its primary sequence does not resemble any calcium channel studied to date. Here we report the structure of the pore domain of MCU from Caenorhabditis elegans, determined using nuclear magnetic resonance (NMR) and electron microscopy (EM). MCU is a homo-oligomer in which the second transmembrane helix forms a hydrophilic pore across the membrane. The channel assembly represents a new solution of ion channel architecture, and is stabilized by a coiled-coil motif protruding into the mitochondrial matrix. The critical DXXE motif forms the pore entrance, which features two carboxylate rings; based on the ring dimensions and functional mutagenesis, these rings appear to form the selectivity filter.     Notes: Abstract An X-ray preionized, discharge-pumped XeCl laser with a variable beam cross-section of up to 6×6 cm2 is described. It uses flat electrodes and the beam width is determined by X-ray collimation. Its operation characteristics concerning reduced electric field strength (E/p) and X-ray dose are discussed in detail. The inductance of the discharge loop is minimized using a water capacitor arrangement. A very high specific optical power (90 MW/l) is achieved in an active volume of 1.2 l. The pulse energy exceeds 5 J in a 45 ns pulse (FWHM).

    Notes: Abstract In this paper, a high power Nd:YAG laser system is described, which consists of an actively-passively mode-locked Nd:YAG laser oscillator with variable subnanosecond pulsewidths (200 ps to 1 ns), a Pockels-cell shutter and a coaxial four-pass Nd:YAG amplifier. With the oscillator operating in the colliding pulse regime, and a BDN dye film as the saturable absorber, and an acousto-optic modulator as the active modelocker, we have achieved a shot-to-shot stability of the pulse-train amplitude 〈 ±4%. For a 0.1 mJ, 200 ps laser pulse, we obtained four-pass amplification of 7 × 102 with a pump energy of 106 J.

    Notes: Abstract 57Fe Mössbauer effect study has been performed on intercalated compound 2H−Fe0.5TaSe2 over a temperature range between 14K and 500K. The highly concentrated intercalation samples 2H−FexTaSe2 with Fe atoms (x up to 2) are prepared by the electrochemical method for the first time. The Mössbauer measurement shows that iron is in Fe3+ high spin state and with the same probability occupies both octahedral and tetrahedral interstices of the Van der Waals gap. The fact that effective masses of iron ions in both sites are close to 57 amu and characteristic temperatures (ΘM =130K, 107K respectively) are found to be relatively small is attributed to the weak forces on Fe3+.

    Description: In order to investigate the heavy metal pollution conditions of soil of smelting area in Zijiang of Chenzhou, Hunan province, 42 samples were studied. The concentrations of heavy metals As, Pb, Cd, Zn and Cu in the soil were determined by using atomic absorption spectrometry(AAS) and atomic fluorescence spectrometry(AFS). Then the potential pollution risks of heavy metal in the soil were evaluated by method of geological acumination index (I geo ). The results indicated that the average concentrations of As, Pb, Cd, Zn and Cu were 187.79, 2074.52, 15.72, 2178.89, 39.69 mg/kg respectively. The geological evaluation of the cumulative index results showed that the contamination degree of 5 heavy metals follow the sequence of Cd〉 Zn 〉Pb 〉 As 〉Cu.
