Notes: Summary Energy dispersive X-ray microanalysis was applied to freeze-dried blocks of enamel organ tissue to determine levels of calcium in various celular regions. The tissue blocks were dissected free from adjacent forming enamel following injection of cobalt or fluoride ions, both of which temporarily inhibit enamel mineralization. In all control and experimental specimens there was an increasing gradient of calcium from the stratum intermedium cells to the distal ends of the ameloblasts. Calcium levels were significantly reduced near the distal ends of the ameloblasts following cobalt or fluoride injection as compared with controls. It is suggested that evidence of an intercellular buildup of calcium near the distal ends of the ameloblast supports a controlling function of these cells. The changes in calcium levels are correlated with alterations in mineralization known to occur in the adjacent enamel of the model systems employed.

Notes: Abstract Adult male pigeons were fed 500 ppm of nontoxic chlordene per day for 6 weeks. The ultrastructure of hepatic cells of treated pigeons was examined and compared with that of the control birds. Chlordene feeding induced cytological changes in the structure of liver cells. Thinning of the microvilli of bile canaliculi and clear marking of fibrous long spacing and non-fibrous segment long spacing on collagen fibers were noticed. Increase in the number as well as the vacuolation of lipid bodies in certain portions of the cytoplasm, presence of myeloid bodies in mitochondria, and an increase in the smooth-surfaced endoplasmic reticulum were observed in the liver cells of pigeons given chlordene.

Notes: Summary Some of the parameters affecting the staining of keratinized oral epithelium with the zinc iodide-osmium reaction were examined using light and electron microscopy and electron probe microanalysis. Factors examined were block size, incubation temperature and the effect of aldehyde prefixation. Large blocks (4 mm cube) were subdivided after incubation and the staining of the centre and edge compared. Generally the reaction was more variable at the edge than in the centre. Small blocks (1 mm cube) showed a more intense reaction when incubated at 24°C than at 4°C. In all these preparations, final reaction product was seen over Golgi systems, lysosome-like bodies, membrane-coating granules and, in the more intensely stained regions, over endoplasmic reticulum and nuclear membranes as well. In prefixed material, mitochondria were frequently stained in addition to the other organelles. Energy dispersive analysis showed the reaction product to be similar in all preparations and to contain high levels of zinc and osmium but not iodine.
