    Notes: Summary Two excitatory photoreceptor sites (P1, P2) have been demonstrated on the genitalia in adults of both sexes of the Chinese yellow swallowtail butterflyPapilio xuthus. The light stimulus reaches sensory units through transparent windows in the overlying cuticle. Photoreceptor cells in the scaphium (♂P1) or papilla analis (♀P1) and both P2's respond similarly to light flashes with spike trains produced in afferent axons. All sites have spectral response curves which peak near 380 nm with a lower maximum at 450 nm. Afferent axons from P1 and P2 have ipsilateral terminal arborizations in the last abdominal ganglion; their somata are peripheral. Well developed in adults, this synthesis is absent in the mature larva ofPapilio xuthus. Comparative study of 22 other lepidopteran species in 10 families proves the presence of similar genital extraocular photoreception in all butterflies tested. It also occurs in skippers, but not in three other families of moths.

    Notes: Summary 1. A butterfly Papilio has 5 types of spectral receptors in the compound eye. The spectral sensitivity of each type peaks in the UV, violet, blue, green, and red wavelengths, respectively. The green type contains two subtypes with and without a UV secondary peak. Here we studied the localization of these spectral receptors within the ommatidium. 2. An ommatidium contains 9 photoreceptors (R1–9), each of which is one of the 5 spectral receptor types. The photoreceptors bear parallel microvilli to form a nontwisted rhabdom, and thereby the photoreceptors are polarization sensitive. 3. We first examined the microvillar orientation by electron microscopy. The microvilli of R1, 2, and 9 are oriented dorso-ventrally (0°), whereas those of R3 and 4 are parallel to the antero-posterior axis (90°). The R5–8 bear microvilli diagonally: 45° for R6 and R8, 135° for R5 and R7. 4. We then recorded spectral and polarization sensitivities from single photoreceptors. The peak angle of the polarization sensitivity (θmax) of the UV, violet, and blue receptors were around 0°, whereas that of the green receptors was around 90°. In the double-peaked green receptors, the θmax at UV was also around 90°. The red receptors showed a θmax at around 35°. The polarization sensitivity ratio (PSmax/PSmin) of the double-peaked green receptors measured at UV was around 4, whereas the ratio of other receptors was around 2. 5. We conclude that R1 and R2 are either UV, violet, or blue receptors whereas R3 and R4 are green receptors. Some R6 and R8 are red receptors. We also conclude that the UV secondary peak in the double-peaked green receptor is not simply attributable to the coupling with UV receptors.

    Notes: Summary The compound eye of the crabHemigrapsus sanguineus undergoes daily changes in morphology as determined by light and electron microscopy, both in the quantity of chromophore substances studied by HPLC and in visual sensitivity as shown by electrophysiological techniques. 1. At a temperature of 20 °C, the rhabdom occupation ratio (ROR) of an ommatidial retinula was 11.6% (maximum) at midnight, 8.0 times larger than the minimum value at midday (1.4%) (Figs. 2, 6). 2. Observations by freeze-fracture revealed that the densities of intra-membranous particles (9–11 nm in diameter) of rhabdomeric membrane were ca. 2000/μm2 and ca. 3000/μm2 for night and daytime compound eyes, respectively (Fig. 3). 3. Screening pigment granules migrated longitudinally and aggregated at night, but dispersed during the day. Reflecting pigment granules migrate transversally in the proximal half of the retinula layer i.e. cytoplasmic extensions containing reflecting pigment granules squeeze between neighbouring retinula cells causing optical isolation (Fig. 4). Thus the screening pigment granules within the retinula cells show longitudinal migration and radial movement so that the daytime rhabdoms are closely surrounded by the pigment granules (Fig. 2). 4. At 20 °C, the total amount of chromophore of the visual pigment (11-cis and all-trans-retinal) was 1.4 times larger at night than during the day i.e. 46.6 pmol/eye at midnight and 33.2 pmol/eye at midday (Fig. 9). Calculations of the total surface area of rhabdomeric membrane, total number of intra-membranous particles in rhabdomeric membrane and the total number of chromophore molecules in a compound eye, indicate that a considerable amount of chromophore-protein complex exists outside the rhabdom during the day. 5. The change in rhabdom size and quantity of chromophore were highly dependent on temperature. At 10 °C both rhabdom size and amount of chromophore stayed close to daytime levels throughout the 24 hours (Figs. 6, 9). 6. The intracellularly determined relative sensitivity of the dark adapted night eye to a point source of light was about twice as high as the darkadapted day eye (Fig. 10). Most of the increase in the sensitivity is attributed primarily to the effect of reflecting pigment migration around the basement membrane (Fig. 4) and, secondarily, to the changes in the amount and properties of the photoreceptive membrane. The results form the basis of a detailed discussion as to how an apposition eye can function possibly as a night-eye.

    Notes: Abstract This paper describes the primary structure of two visual pigment opsins (DfRh1 and DfRh2) in the regionalized compound eye of a dragonfly,Sympetrum frequens. The amino acid sequences were deduced from the nucleotide sequences of cDNAs isolated from a cDNA library of the dragonfly retina. The two opsins both consist of 379 amino acids with 81.3% identity. Analysis of hydropathy indicated that the sequences have seven transmembrane domains like those of previously described opsins. Expression analysis using RT-PCR revealed that DfRh1 was present only in the dorsal region whereas DfRh2 was detected in both the dorsal and the ventral regions of the eye.

    Notes: Actin has many diverse functions in the outer retina. To help elucidate its organization in this area, we have investigated the extent of its association with the actin cross-linking protein alpha-actinin. Ultrathin sections of chicken retina were double-immunolabelled with monospecific antibodies against actin and alpha-actinin. The highest relative amount of alpha-actinin to actin label was measured in the adherens junctions between the individual retinal pigmented epithelial (RPE) cells and between the photoreceptor and Mueller cells; in the photoreceptor myoid; and in the RPE basal microvilli. The lowest amount was in the Mueller cell microvilli, the RPE apical processes, and in the photoreceptor ellipsoid. It is likely that the areas containing the highest ratio of alpha-actinin to actin labelling are where the actin filaments are most highly cross-linked into bundles and linked to the plasma membrane by alpha-actinin. Actin filaments terminate in these areas, and, except for the myoid region, they are involved in cell-cell or cell-substrate adherens junctions.

This paper demonstrates that the actin filaments in the photoreceptor cell body of the crab Hemigrapsus sanguineus are involved in the transport of the smooth endoplasmic reticulum in the process of rhab- dom synthesis. In isolated eyes, the transverse sectional area of a rhabdom increased at dusk. The increase was inhibited by disrupting the actin filaments with cytochalasin D. In the photoreceptor cell body of cytochalasin D-treated eyes, rhodamine-conjugated phalloidin labeled some spots, which appeared to be the aggregates of short filaments as revealed by electron microscopy. The photoreceptor cell bodies in cytochalasin D-treated eyes contained more vesicular smooth endoplasmic reticulum than those in the control eye. These results indicate that the disruption of actin organization with cytochalasin D resulted in the accumulation of smooth endoplasmic reticulum. Microtubules do not play a direct role in the rhabdom synthesis because colchicine failed to stop the rhabdom increase.

    Description: Dragonflies are colorful and large-eyed animals strongly dependent on color vision. Here we report an extraordinary large number of opsin genes in dragonflies and their characteristic spatiotemporal expression patterns. Exhaustive transcriptomic and genomic surveys of three dragonflies of the family Libellulidae consistently identified 20 opsin genes, consisting of 4 nonvisual opsin genes and 16 visual opsin genes of 1 UV, 5 short-wavelength (SW), and 10 long-wavelength (LW) type. Comprehensive transcriptomic survey of the other dragonflies representing an additional 10 families also identified as many as 15–33 opsin genes. Molecular phylogenetic analysis revealed dynamic multiplications and losses of the opsin genes in the course of evolution. In contrast to many SW and LW genes expressed in adults, only one SW gene and several LW genes were expressed in larvae, reflecting less visual dependence and LW-skewed light conditions for their lifestyle under water. In this context, notably, the sand-burrowing or pit-dwelling species tended to lack SW gene expression in larvae. In adult visual organs: (i) many SW genes and a few LW genes were expressed in the dorsal region of compound eyes, presumably for processing SW-skewed light from the sky; (ii) a few SW genes and many LW genes were expressed in the ventral region of compound eyes, probably for perceiving terrestrial objects; and (iii) expression of a specific LW gene was associated with ocelli. Our findings suggest that the stage- and region-specific expressions of the diverse opsin genes underlie the behavior, ecology, and adaptation of dragonflies.
