    Notes: Abstract. We have developed a simple dye transfer method, which allows the gap junction permeability of lens fiber cells to be quantified. Two fixable fluorescent dyes (Lucifer yellow and rhodamine-dextran) were introduced into peripheral lens fiber cells via mechanical damage induced by removing the lens capsule. After a defined incubation period, lenses were fixed, sectioned, and the distribution of the dye recorded using confocal microscopy. Rhodamine-dextran and Lucifer yellow both labeled the extracellular space between fiber cells and the cytoplasm of fiber cells that had been damaged by capsule removal. For the gap junctional permeable dye Lucifer yellow, however, labeling was not confined to the damaged cells and exhibited intercellular diffusion away from the damaged cells. The extent of dye diffusion was quantified by collecting radial dye intensity profiles from the confocal images. Effective diffusion coefficients (D eff ) for Lucifer yellow were then calculated by fitting the profiles to a series of model equations, which describe radial diffusion in a sphere. D eff is the combination of dye diffusion through the cytoplasm and through gap junction channels. To calculate the gap junctional permeability (P j ) an estimate of the cytoplasmic diffusion coefficient (D cyt = 0.7 × 10−6 cm2/sec) was obtained by observing the time course of dye diffusion in isolated elongated fiber cells loaded with Lucifer yellow via a patch pipette. Using this approach, we have obtained a value for P j of 31 × 10−5 cm/sec for fiber-fiber gap junctions. This value is significantly larger than the value of P j of 4.4 × 10−6 cm/sec reported by Rae and coworkers for epithelial-fiber junctions (Rae et al., 1996. J. Membrane Biol. 150:89–103), and most likely reflects the high abundance of gap junctions between lens fiber cells.

    Notes: A new asymmetric domain for intercrystalline misorientation is defined in the space of Euler angles for materials exhibiting cubic (Oh point-group) lattice symmetry. The invariant measure for this new domain is nearly constant; this is in significant contrast to the previous domain defined by MacKenzie [Biometrika (1958), 45, 229-240]. Distribution functions in the misorientation can now be represented with greater clarity and convenience in the new domain. A detailed theoretical analysis of special misorientations exhibiting multiplicities m 〉 1 is described. It is demonstrated that all such special misorientations fall upon the surfaces separating distinct asymmetric domains. This result convincingly proves that the derived asymmetric domain is correct. The location of all possible coincidence site lattice boundaries for Σ ≤ 49 are identified in the asymmetric domain, and their characteristic multiplicities are given.

    Notes: A procedure for land-surface zoning, related to the protection of groundwater against both point and diffuse pollution, is described. The procedure is based upon two independent elements:(i) Division of the entire land surface on the basis of the aquifer pollution vulnerability, which relates to protection of groundwater resources; and(ii) A series of special protection areas for individual sources, in which various potentially polluting activities are either prohibited or strictly controlled.The procedure is hydrogeologically based, but not so complex as to be unworkable in practice. For resource protection three classes of formation type are defined, with a further subdivision on the basis of depth to saturated aquifer. In the case of diffuse pollution, it will also be necessary to consider the nature of the soil cover in the area where the polluting activity occurs. For source protection, up to four special protection areas are specified, and the criteria used to define them are discussed.

    Notes: . Two factors affecting the centralization of wastewater treatment facilities were investigated; the cost of collection and treatment systems and the performance of treatment plants. Based on computer-generated minimum cost designs, wastewater collection networks were found to be characterized by diseconomies of scale of magnitude similar to the reported economies of scale for wastewater treatment works. The combined costs of collection and treatment are U-shaped functions from which the least cost size of collection and treatment systems were found for particular values of population density. Examination of the day-to-day performance of five metropoitan-area waste-water treatment plants revealed that, for time series shorter than one month, the day-to-day variation in effluent quality was random, although the variation in quantity discharged was distinctly non-random. The performances of all five plants on any given day showed little correlation. This suggests that the decentralization of treatment facilities can produce benefits both through the reduction in quantities of waste discharged at a given point and through in-stream averaging of the varying performances of several treatment plants. Since the cost function of collection and treatment combined is generally flat in the region of the minimum-cost size, little penalty is invoked to gain the potential benefits of treatment plant decentralization.
