    Description: In the mammalian CNS, N-methyl-D-aspartate (NMDA) receptors serve prominent roles in many physiological and pathophysiological processes including pain transmission. For full activation, NMDA receptors require the binding of glycine. It is not known whether the brain uses changes in extracellular glycine to modulate synaptic NMDA responses. Here, we show that synaptically released glycine facilitates NMDA receptor currents in the superficial dorsal horn, an area critically involved in pain processing. During high presynaptic activity, glycine released from inhibitory interneurons escapes the synaptic cleft and reaches nearby NMDA receptors by so-called spillover.     Description: We present video-rate (28 frames per second) far-field optical imaging with a focal spot size of 62 nanometers in living cells. Fluorescently labeled synaptic vesicles inside the axons of cultured neurons were recorded with stimulated emission depletion (STED) microscopy in a 2.5-micrometer by 1.8-micrometer field of view. By reducing the cross-sectional area of the focal spot by about a factor of 18 below the diffraction limit (260 nanometers), STED allowed us to map and describe the vesicle mobility within the highly confined space of synaptic boutons. Although restricted within boutons, the vesicle movement was substantially faster in nonbouton areas, consistent with the observation that a sizable vesicle pool continuously transits through the axons.     Description: We present a record of extreme spring–summer runoff events for the past 1600 yr preserved in the varved sediments of Lake Mondsee (Austrian Pre-Alps). Combined sediment microfacies analyses and high-resolution micro-X-ray fluorescence element scanning allow us to identify 157 detrital event layers deposited in spring–summer and to discriminate between regional flood and local debris flow deposits. Higher spring–summer flood activity with a mean event recurrence of 3–5 yr occurred in several well-confined multidecadal episodes during the Dark Ages Cold Period and Medieval time (A.D. 450–480, 590–640, 700–750, and 1140–1170) as well as during the early Little Ice Age (LIA; A.D. 1300–1330 and 1480–1520). In contrast, lowest spring–summer flood activity with an event recurrence of only 30–100 yr is observed during the Medieval Climate Anomaly (A.D. 1180–1300) and the coldest interval of the LIA (A.D. 1600–1700). These findings indicate a complex relationship between temperature conditions and extreme hydro-meteorological events and suggest that enhanced summer Mediterranean cyclogenesis triggers large-scale floods in the northeast Alps during climatic transitions. The Lake Mondsee data demonstrate the climatic sensitivity of spring–summer floods and prove the potential of varved sediment records to investigate the impact of changing climate boundary conditions on seasonal flood activity for pre-instrumental time.

    Description: Stimulated Emission Depletion (STED) microscopy enables superresolution imaging of fluorescently marked nano-structures in vivo. Biological investigations are often hindered by the difficulty of relating super-resolved structures to other non-labeled features. Here we demonstrate that the similarity in optical design of Spiral Phase Contrast (SPC) and STED microscopes allows straightforward implementation of a phase contrast channel into a STED microscope in widefield and scanning modes. This method allows dual imaging and overlay in two contrast modes in fixed and in living specimens, in which double labeling is especially challenging. Living GFP- and YPF-stained neurons are imaged in one label-free phase contrast and one high-resolution STED channel. Furthermore, we implement SPC in widefield and scanning modes demonstrating that scanning confocal SPC yields the highest optical contrast. The latter configuration can provide contour detection or highlights and shadows reminiscent of differential interference contrast. Scientific Reports 3 doi: 10.1038/srep02050

    Description: A theoretical model based on heat and mass transfer equations was developed for determining a reliable comparison between different solar kiln designs. It involves dividing the drying process into two sequential steps, one in which only heat transfer occurs and a second one in which the actual drying process takes place. The model gives an equation for determining the evaporated water in function of the normal drying variables, together with a specially defined parameter which gives account for the air recirculation inside the kiln. The model was validated experimentally for a box-type collector solar kiln operating under constant and falling rate drying periods. It was found that although the model could be applied satisfactorily, the tested design proved to be a very inefficient one, giving low values for the recirculating parameter.
