    Description: Somatic chromosomal deletions in cancer are thought to indicate the location of tumour suppressor genes, by which a complete loss of gene function occurs through biallelic deletion, point mutation or epigenetic silencing, thus fulfilling Knudson's two-hit hypothesis. In many recurrent deletions, however, such biallelic inactivation has not been found. One prominent example is the 5q- syndrome, a subtype of myelodysplastic syndrome characterized by a defect in erythroid differentiation. Here we describe an RNA-mediated interference (RNAi)-based approach to discovery of the 5q- disease gene. We found that partial loss of function of the ribosomal subunit protein RPS14 phenocopies the disease in normal haematopoietic progenitor cells, and also that forced expression of RPS14 rescues the disease phenotype in patient-derived bone marrow cells. In addition, we identified a block in the processing of pre-ribosomal RNA in RPS14-deficient cells that is functionally equivalent to the defect in Diamond-Blackfan anaemia, linking the molecular pathophysiology of the 5q- syndrome to a congenital syndrome causing bone marrow failure.     Description: Cells of the osteoblast lineage affect the homing and the number of long-term repopulating haematopoietic stem cells, haematopoietic stem cell mobilization and lineage determination and B cell lymphopoiesis. Osteoblasts were recently implicated in pre-leukaemic conditions in mice. However, a single genetic change in osteoblasts that can induce leukaemogenesis has not been shown. Here we show that an activating mutation of beta-catenin in mouse osteoblasts alters the differentiation potential of myeloid and lymphoid progenitors leading to development of acute myeloid leukaemia with common chromosomal aberrations and cell autonomous progression. Activated beta-catenin stimulates expression of the Notch ligand jagged 1 in osteoblasts. Subsequent activation of Notch signalling in haematopoietic stem cell progenitors induces the malignant changes. Genetic or pharmacological inhibition of Notch signalling ameliorates acute myeloid leukaemia and demonstrates the pathogenic role of the Notch pathway. In 38% of patients with myelodysplastic syndromes or acute myeloid leukaemia, increased beta-catenin signalling and nuclear accumulation was identified in osteoblasts and these patients showed increased Notch signalling in haematopoietic cells. Das Problem wird dimensionsanalytisch untersucht, und die Grundgleichungen des verallgemeinertennewtonschen Fließens durch ein Rohr werden abgeleitet. Für den Spezialfall dernewtonschen Flüssigkeit mit druck- und temperaturabhängiger Viskosität werden durch Anwendung eines Störungsverfahrens sowohl für isotherme als auch adiabatische Bedingungen an der Wand Ausdrücke für die Geschwindigkeitskomponenten, die Druckverteilung und den Volumenstrom angegeben. Man findet, daß die Druck- und Aufheizungs-Effekte nicht-newtonsches Verhalten vortäuschen können.
    Notes: Summary The basic relations governing viscous flow of liquids with variable transport properties is considered. A dimensional analysis of the problem is given, and the basic equations of generalizedNewtonian flow through a pipe are derived. A perturbation solution is given for the special case of aNewtonian liquid with pressure and temperature dependent viscosity and expressions for the velocity components, pressure distribution and flow rate are obtained for both isothermal and adiabatic wall conditions. It is shown that pressure and viscous heating effects may result in apparently nonNewtonian behaviour.

    Description / Table of Contents: Zusammenfassung Die Grundgleichungen für das verallgemeinerteNewtonsche Rohrfließen wird mit Hilfe einer Störungsmethode für eine Potenzflüssigkeit mit temperaturund druckabhängiger Viskosität gelöst. In der ersten Näherung werden die Geschwindigkeitskomponenten, der Durchsatz und das Druckprofil erhalten. Es werden Tabellen gegeben, um die Temperatur-Effekte zu berechnen. Die Lösung scheint in guter Übereinstimmung mit den numerischen und experimentellen Ergebnissen zu sein und enthalten als Grenzfälle Ergebnisse anderer Autoren. Die Analyse führt zu einem einfachen Kriterium, das die Temperaturund Druckeffekte vorhersagen kann.
    Notes: Summary The basic equations of generalizedNewtonian pipe flow are solved by a perturbation procedure, for the case of power-law liquids with pressure and temperature variable viscosity. A first-approximation to the solution is obtained for the velocity components, the flow rate, and the pressure distribution, and tables are given for the calculation of viscous heating effects. The solution is shown to be in agreement with numerical and experimental results, and includes, as limiting cases, some other cases studied. The analysis leads to simple criteria for the prediction of the mutual effects of heat and pressure in viscous flow.
