    Description: Two endoplasmic reticulum (ER) molecular chaperones [glucose-regulated protein 78 ( grp78 ) and calreticulin ( crt )] and three ER stress sensors [PKR-like ER kinase ( perk ), inositol requiring enzyme ( ire )-1α, and activating transcription factor ( atf )-6α] cDNAs were first characterized from yellow catfish, Pelteobagrus fulvidraco . The predicted amino acid sequences for the yellow catfish grp78 , crt , perk , ire -1α, and atf -6α revealed that the proteins contained all of the structural features that were characteristic of the five genes in other species, including the KDEL motif, signal peptide, sensor domain, and effector domain. mRNAs of the five genes mentioned above were expressed in various tissues, but their mRNA levels varied among tissues. Dietary Cu excess, but not Cu deficiency, activated the chaperones ( grp78 and crt ) and folding sensors in ER, and the UPR signaling pathways ( i.e. , perk – eif 2α and the ire 1– xbp1 ) in a tissue-specific manner. For the first time, our study cloned grp78 , crt , perk , ire -1α, and atf -6α genes in yellow catfish and demonstrated their differential expression among tissues. Moreover, the present study also indicated differential regulation of these ER stress–related genes by dietary Cu deficiency and excess, which will be beneficial for us to evaluate effects of dietary Cu levels in fish at the molecular level, based on the upstream pathway of lipid metabolism (the ER) and thus provide novel insights regarding the nutrition of Cu in fish.

    Description: : DNA replication, a central event for cell proliferation, is the basis of biological inheritance. The identification of replication origins helps to reveal the mechanism of the regulation of DNA replication. However, only few eukaryotic replication origins were characterized not long ago; nevertheless, recent genome-wide approaches have boosted the number of mapped replication origins. To gain a comprehensive understanding of the nature of eukaryotic replication origins, we have constructed a Database of Eukaryotic ORIs (DeOri), which contains all the eukaryotic ones identified by genome-wide analyses currently available. A total of 16 145 eukaryotic replication origins have been collected from 6 eukaryotic organisms in which genome-wide studies have been performed, the replication-origin numbers being 433, 7489, 1543, 148, 348 and 6184 for humans, mice, Arabidopsis thaliana , Kluyveromyces lactis , Schizosaccharomyces pombe and Drosophila melanogaster , respectively.     Description: This interdisciplinary study covers bionics, optimization and vehicle engineering. Semi-track air-cushion vehicle (STACV) provides a solution to transportation on soft terrain, whereas it also brings a new problem of excessive fuel consumption. By mimicking the foraging behaviour of honeybees, the bioinspired adaptive bees algorithm (ABA) is proposed to calculate its running parameters for fuel economy optimization. Inherited from the basic algorithm prototype, it involves parallel-operated global search and local search, which undertake exploration and exploitation, respectively. The innovation of this improved algorithm lies in the adaptive adjustment mechanism of the range of local search (called ‘patch size’) according to the source and the rate of change of the current optimum. Three gradually in-depth experiments are implemented for 143 kinds of soils. First, the two optimal STACV running parameters present the same increasing or decreasing trend with soil parameters. This result is consistent with the terramechanics-based theoretical analysis. Second, the comparisons with four alternative algorithms exhibit the ABA's effectiveness and efficiency, and accordingly highlight the advantage of the novel adaptive patch size adjustment mechanism. Third, the impacts of two selected optimizer parameters to optimization accuracy and efficiency are investigated and their recommended values are thus proposed.

    Description: : Recent advances in high-throughput sequencing technologies have enabled us to sequence large number of cancer samples to reveal novel insights into oncogenetic mechanisms. However, the presence of intratumoral heterogeneity, normal cell contamination and insufficient sequencing depth, together pose a challenge for detecting somatic mutations. Here we propose a fast and an accurate somatic single-nucleotide variations (SNVs) detection program, FaSD-somatic. The performance of FaSD-somatic is extensively assessed on various types of cancer against several state-of-the-art somatic SNV detection programs. Benchmarked by somatic SNVs from either existing databases or de novo higher-depth sequencing data, FaSD-somatic has the best overall performance. Furthermore, FaSD-somatic is efficient, it finishes somatic SNV calling within 14 h on 50X whole genome sequencing data in paired samples.     Description: Cell-cell fusion is critical for the conception, development, and physiology of multicellular organisms. Although cellular fusogenic proteins and the actin cytoskeleton are implicated in cell-cell fusion, it remains unclear whether and how they coordinate to promote plasma membrane fusion. We reconstituted a high-efficiency, inducible cell fusion culture system in the normally nonfusing Drosophila S2R+ cells. Both fusogenic proteins and actin cytoskeletal rearrangements were necessary for cell fusion, and in combination they were sufficient to impart fusion competence. Localized actin polymerization triggered by specific cell-cell or cell-matrix adhesion molecules propelled invasive cell membrane protrusions, which in turn promoted fusogenic protein engagement and plasma membrane fusion.     Description: As the world’s leading aquaculture, aquatic production and trading country, China’s development of aquatic products trade with ASEAN is facing a historic opportunity in the favourable circumstances of construction of the 21st century Maritime Silk Road. In order to make guidance of the product selection and transformation for corresponding export enterprises, this article makes a quantitative analysis the Revealed Comparative Advantage of aquatic products trade from China and ASEAN respectively based on the HS classification and thoroughly compares the RCA indices. The comparison results show that the international competitiveness of aquatic products structures of China and ASEAN are quite different with few overlaps of strong competitive products, and there is a great gap between the two areas in many kinds of products.
