    Notes: Summary Cuphea lanceolata Ait. has had a significant role in the domestication of Cuphea and is a useful experimental organism for investigating how medium-chain lipids are synthesized in developing seeds. To expand the genetics of this species, a linkage map of the C. lanceolata genome was constructed using five allozyme and 32 restriction-fragment-length-polymorphism (RFLP) marker loci. These loci were assigned to six linkage groups that correspond to the six chromosomes of this species. Map length is 288 cM. Levels of polymorphism were estimated for three inbred lines of C. lanceolata and an inbred line of C. viscosissima using 84 random genomic clones and two restriction enzymes, EcoRI and HindIII. Of the probes 29% detected RFLPs between C. lanceolata and C. viscosissima lines. Crosses between these species can be exploited to expand the map.

    Notes: Abstract  Soybean (Glycine max) is one of the world’s most important crop plants due to extensive genetic improvements using traditional breeding approaches. Recently, marker-assisted selection has enhanced the ability of traditional breeding programs to improve soybeans. Most methods of assessing molecular markers involve electrophoretic techniques that constrain the ability to perform high-throughput analyses on breeding populations and germplasm. In order to develop a high-capacity system, we have developed allele-specific hybridization (ASH) markers for soybean. As one example, restriction fragment length polymorphism (RFLP) locus A519-1 (linkage group B) was converted into an ASH marker by (1) sequencing the pA519 cloned insert, (2) designing locus-specific PCR amplification primers, (3) comparative sequencing of A519-1 amplicons from important soybean ancestors, and (4) designing allele-specific oligonucleotide probes around single nucleotide polymorphisms (SNPs) among soybean genotypes. Two SNPs were identified within approximately 400 bp of the sequence. Allele-specific probes generated a 100-fold greater signal to target amplicons than to targets that differed by only a single nucleotide. The A519-1 ASH marker is shown to cosegregate with the A519-1 RFLP locus. In order to determine ASH usefulness, we genotyped 570 soybean lines from the Pioneer Hi-Bred soybean improvement using both A519-1 SNPs. Combined haplotype diversity (D) was 0.43 in this adapted germplasm set. These results demonstrate that ASH markers can allow for high-throughput screening of germplasm and breeding populations, greatly enhancing breeders’ capabilities to do marker-assisted selection.

    Description: The battery is designed with a minimum battery to cell weight ratio consistent with adequate containment for operating conditions and dynamic environments and minimized weight. The battery is fully qualified and the environments to which it was successfully subjected were selected by NASA Goddard to cover a wide range of probable uses. The battery is suitable for either near-Earth geosynchronous missions, is compatible with passive or active thermal control systems and may be electrically controlled by a variety of changing routines. The initial application of the 50 A.H. Battery is a near-Earth mission aboard the LANDSAT D Satellite.

    Description: Three batteries were operated in parallel from a common bus during charge and discharge. SMM utilized NASA Standard 20AH cells and batteries, and LANDSAT-D NASA 50AH cells and batteries of a similar design. Each battery consisted of 22 series connected cells providing the nominal 28V bus. The three batteries were charged in parallel using the voltage limit/current taper mode wherein the voltage limit was temperature compensated. Discharge occurred on the demand of the spacecraft instruments and electronics. Both flights were planned for three to five year missions. The series/parallel configuration of cells and batteries for the 3-5 yr mission required a well controlled product with built-in reliability and uniformity. Examples of how component, cell and battery selection methods affect the uniformity of the series/parallel operation of the batteries both in testing and in flight are given.
