    Notes: Summary An RFLP-based genetic map of Secale Cereale has provided evidence for multiple evolutionary translocations in the rye genome relative to that of hexaploid wheat. DNA clones which have previously been mapped in wheat indicated that chromosome arms 2RS, 3RL, 4RL, 5RL, 6RS, 6RL, 7RS and 7RL have all been involved in at least one translocation. A possible evolutionary pathway, which accounts for the present day R genome relative to the A, B and D genomes of wheat, is presented. The relevance of these results for strategies designed to transfer useful genes from rye, and probably other related species, to wheat is discussed.

    Notes: Summary The ultrastructure of the epidermal and sub-epidermal cells of the appendix of the Sauromatum guttatum inflorescence reveals developmental changes during anthesis. These changes precede, and probably make possible, heat and odor production. Two days before D-day (the day of heat production and inflorescence-opening) the mitochondria of the epidermis divide; apparent division of the amyloplasts was observed at the same time. The presence of lipid bodies and peroxisomes in the epidermis was clearly evident. On D-day, the epidermis becomes a continuous layer in which the cell walls separating two adjacent cells disappear. At the same time, in the sub-epidermal cells, the mitochondria and the amyloplasts undergo division. The mitochondria become electron-dense, and their DNA is clearly visible. On that day, lipids as well as starch are being depleted. The peroxisomes change in structure every day, from D-2 to D-day. It has also been demonstrated by histochemical techniques that during anthesis the activity of cytochrome c oxidase (3,3-diaminobenzidine as a substrate) decreases whereas the activity of NADH dehydrogenase [tetrazolium salts: nitro-blue tetrazolium chloride (NBT) or neotetrazolium chloride (NT) in the presence of NADH], increases. Oxygen consumption of isolated mitochondria from the D-day appendix was inhibited in the presence of the two tetrazolium salts to a different degree: oxidation of NADH in the presence of NBT was the most sensitive to inhibition, more so than the oxidation of malate and succinate. NT was less effective as an inhibitor in the presence of those three respiratory substrates.

    Notes: Abstract This paper presents the results of simultaneous DTA-TG-DTG and DSC studies on the thermal decomposition of Cu(NO3)2·3H2O, Co(NO3)2·6H2O and Ni(NO3)2·6H2O in an air atmosphere. The mechanism and enthalpies of the investigated processes were determined, as well as the kinetic parameters of the processes run under non-isothermal conditions by means of Kissinger's method. The dependence of the activation energy on the ionic radius of the cations building up the crystal lattices of the investigated compounds was also studied.

    Notes: Abstract Results obtained by the comparative investigations of the natural mineral marmatite (Zn,Fe)S (Stari Trg, Yugoslavia) oxidation process are presented in this paper. Determination of the oxidation process mechanism was done by thermal analysis methods: DTA-TG-DTG and DSC, while Mössbauer spectroscopy, X-ray and electronic microanalysis were used for phase composition determination of the products formed during the oxidation process. Kinetics of the process was defined for two temperature intervals 862–973 and 1023–1173 K, under isothermal conditions.

    Notes: Summary RFLP mapping of chromosome 5R in the F3 generation of a rye (Secale cereale L.) cross segregating for gibberellic acid (GA3)-insensitive dwarfness (Ct2/ct2) and spring growth habit (Sp1/sp1) identified RFLP loci close to each of these agronomically important genes. The level of RFLP in the segregating population was high, and thus allowed more than half of the RFLP loci to be mapped, despite partial homozygosity in the parental F2 plant. Eight further loci were mapped in an unrelated F2 rye population, and a further two were placed by inference from equivalent genetic maps of related wheat chromosomes, allowing a consensus map of rye chromosome 5R, consisting of 29 points and spanning 129 cM, to be constructed. The location of the ct2 dwarfing gene was shown to be separated from the segment of the primitive 4RL translocated to 5RL, and thus the gene is probably genetically unrelated to the major GA-insensitive Rht genes of wheat located on chromosome arms 4BS and 4DS. The map position of Sp1 is consistent both with those of wheat Vrn1 and Vrn3, present on chromosome arms 5AL and 5DL, respectively, and with barley Sh2 which is distally located on chromosome arm 7L (= 5HL).

    Notes: Abstract A map of the barley genome consisting of 295 loci was constructed. These loci include 152 cDNA restriction fragment length polymorphism (RFLP), 114 genomic DNA RFLP, 14 random amplified polymorphic DNA (RAPD), five isozyme, two morphological, one disease resistance and seven specific amplicon polymorphism (SAP) markers. The RFLP-identified loci include 63 that were detected using cloned known function genes as probes. The map covers 1,250 centiMorgans (cM) with a 4.2 cM average distance between markers. The genetic lengths of the chromosomes range from 124 to 223 cM and are in approximate agreement with their physical lengths. The centromeres were localized to within a few markers on all of the barley chromosomes except chromosome 5. Telomeric regions were mapped for the short (plus) arms of chromosomes 1, 2 and 3 and the long (minus) arm of chromosomes 7.

    Notes: Summary Male gametes ofPlumbago zeylanica were examined in pollen grains and tubes using light and electron microscopy of chemically and physically fixed tissues, and Nomarski interference microscopy of isolated, living sperm cells. Male gametes are elongate, spindleshaped cells containing a nucleus, mitochondria, ER, ribosomes, vesicles, dictyosomes, probable microfilaments, and a variable number of plastids. In mature pollen grains ofP. zeylanica, the two sperm cells are directly linked; they share a transverse cell wall with plasmodesmata and are enclosed together by the inner vegetative cell plasma membrane. One of these two sperms is also associated with the vegetative nucleus as a consistent feature of pollen grain organization. The basis of this association appears to be a long, narrow projection of the sperm cell (averaging 〈 1 μm wide and about 30 μm long) which wraps around the periphery of the vegetative nucleus and occupies embayments of that nucleus. This association is maintained throughout pollen tube growth but becomes less extensive near the completion of tube growth and is severed following tube discharge. The consistent occurrence of the sperm-vegetative nucleus association in pollen grains, tubes and isolated pollen cytoplasm suggests that the two structures may be directly connected, but attempts to visualize this type of connection were unsuccessful. Possibly, the entwining nature and extent of complementary interfaces between vegetative nucleus and sperm may have a role in stabilizing their association. Functionally, the two sperms and vegetative nucleus appear to travel as a linked unit within the pollen tube, possibly increasing the effectiveness of gamete delivery and helping to ensure nearly simultaneous transmission of sperms into the receptive megagametophyte.

    Notes: Abstract Infection of ryegrass (Lolium perenne) by an endophytic fungus (Acremonium loliae) confers resistance against the Argentine stem weevil (Listronotus bonariensis). Extracts from ryegrass clones, infected and uninfected withA. loliae, were compared in a feeding choice bioassay, and several fractions were identified which affected stem weevil feeding behavior. One stem weevil feeding deterrent, peramine C12H17N5O, has been isolated from infected ryegrass and partially characterized as a basic indole derivative. Extracts from culturedA. loliae had no effect on stem weevil feeding behavior nor was peramine detected in the fungal cultures examined. Peramine and the other active substances are hydrophilic in contrast to the lipophilic properties reported for the neurotoxic lolitrems also isolated from ryegrass infected withA. loliae and associated with causing ryegrass staggers disorder in livestock. It is suggested that ryegrass staggers and stem weevil feeding deterrency may arise by different biochemical mechanisms.

    Notes: Abstract We used artificial selection experiments to study genetic allocation costs and physiological mechanisms of resistance to herbivory and fungal disease. Genetic costs to resistance were present in some instances and absent in others. Genetic resistance to the fungal pathogen, Leptosphaeria maculans was cost-free, while resistance to Peronospora parasitica showed a negative genetic correlation between disease resistance and growth rate. Leptosphaeria resistant genotypes had 13% higher chitinase activity. Genetic increases in myrosinase activity were correlated with increased resistance to flea beetles (Phyllotreta cruciferae), but resulted in lower plant fecundity, presumably due to production costs of myrosinase. Genetic costs of resistance may maintain genetic variation in natural plant populations. These studies demonstrate the predictive and explanatory power of a functional approach to plant-herbivore and plant-pathogen interactions.
