    Notes: Proteins have been immobilized in porous support particles held in a fixed-bed reactor through which protein solution is continuously circulated. Changing the recirculation flow rate alters the observed immobilization kinetics and the maximum enzyme loading which can be achieved for glucose oxidase and glucoamylase on carbodiimide-treated activated carbon and for glucoamylase immobilized on CNBr-Sepharose 4B. Direct microscopic examination of FITC-labelled protein in sectioned Sepharose particles and indirect activity-loading studies with activated carbon-enzyme conjugates all indicate that immobilized enzyme is increasingly localized near the outer surface of the support particles at larger recirculation flow rates. Restricted diffusion of enzymes may be implicated in this phenomenon. These contacting effects may be significant considerations in the scaleup of processes for protein impregnation in porous supports, since apparent activity and stability of the final preparation depend on internal protein distribution.

    Notes: A carrier-supported mycelial growth of Penicillium chrysogenum was applied to penicillin fermentation system using celite as a support material. Hyphal growth through the pore matrices of the material showed strong anchorages and provided highly stable biofilm growth. With bioparticles developed in such a manner, both cell growth and penicillin production were observed to increase significantly compared to the conventional dispersed filamentous cultures. Maximum values of specific penicillin production rate were found to be constant regardless of the growth form. A three-phase fluidized-bed fermentor was designed and tested for penicillin production using the bioparticles. Two modes of operation, semicontinuous and repeated fed batch, of the fermentor were tried. It was noted that the overgrowth of free mycelia and the development of fluffy loose bioparticles caused poor mixing and made the fermentor operation quite difficult. Control of the bioparticle size and the extension of production phase were therefore considered important to maintain the reactor productivity at a desired level. From the results of repeated fed-batch operation it was found that the control of bioparticle size could be successfully achieved by phosphate-limiting culture condition. Penicillin production under this condition was also observed to be maintained at a high level (about 80% of the maximum) for at least 1 month.

    Notes: A soluble poly(amic acid) precursor solution of fully rod-like poly(p-phenylene pyromellitimide) (PMDA-PDA) was spin cast on silicon substrates, followed by soft bake at 80-185°C and subsequent thermal imidization at various conditions over 185-400°C in nitrogen atmosphere to be converted to the polyimide in films. Residual stress generated at the interface was measured in situ during imidization. In addition, the imidized films were characterized in the aspect of polymer chain orientation and ordering by prism coupling and X-ray diffraction. The soft-baked precursor film revealed a residual stress of 16-28 MPa at room temperature, depending on the soft bake condition: higher temperature and longer time in the soft bake gave higher residual stress. The stress variation in the soft-baked precursor film was not significantly reflected in the final stress in the resultant polyimide film. However, the residual stress in the polyimide film varied sensitively with variations in imidization process parameters, such as imidization temperature, imidization steps, heating rate, and film thickness. The polyimide film exhibited a wide range of residual stress, -7 MPa to 8 MPa at room temperature, depending on the imidization condition. Both rapid imidization and low-temperature imidization generated high stress in the tension mode in the polyimide film, whereas slow imidization as well as high temperature imidization gave high stress in the compression mode. Thus, a moderate imidization condition, a single- or two-step imidization at 300°C for 2 h with a heating rate of 〈 10 K/min was proposed to give a relatively low stress in the polyimide film of 〈 10 μm thickness. However, once a precursor film was thermally imidized at a chosen process condition, the residual stress-temperature profile was insensitive to variations in the cooling process. All the films imidized were optically anisotropic, regardless of the imidization history, indicating that rod-like PMDA-PDA polyimide chains were preferentially aligned in the film plane. However, its degree of in-plane chain orientation varied on the imidization history. It is directly correlated to the residual stress in the film, which is an in-plane characteristic. For films with residual stress in the tension mode, higher stress films exhibited lower out-of-plane birefringence, that is, lower in-plane chain orienta-tion.     Notes: New finite element procedures based on the streamline-upwind/Petrov-Galerkin formulations are developed for time-dependent convection-diffusion-reaction equations. These procedures minimize spurious oscillations for convection-dominated and reaction-dominated problems. The results obtained for representative numerical examples are accurate with minimal oscillations.As a special application problem, the single-well chemical tracer test (a procedure for measuring oil remaining in a depleted field) is simulated numerically. The results show the importance of temperature effects on the interpreted value of residual oil saturation from such tests.

    Notes: Heterogeneous equation systems in a pair of coupled co-ordinate systems are solved by a finite element method. The specific physical application studied is the effect of temperature on single-well chemical tracer (SWCT) tests to measure residual oil saturation (volume fraction of immobile oil phase) remaining after waterflooding of an oil reservoir. Since temperature effects are caused by injecting cooler surface fluid down a well into a warm reservoir, the vertical temperature profile in the wellbore as well as the temperature distribution in the porous oil-bearing layer must be considered.The entire system is modelled to account for the different transport mechanisms. However, it is expedient to divide the connected geometrical region into two model domains. The equations for each submodel are expressed in an appropriate set of co-ordinates. The variational formulation of each model is then discussed.A significant temperature effect on the estimation of residual oil saturation occurs when the radial temperature and concentration wave propagation speeds in the porous formation are about the same. In this case the temperature gradient is located across the chemical tracer bank, causing the chemical reaction rate to vary radially. The temperature effects are demonstrated for two actual field tests in complex reservoirs.

    Notes: Raman spectra of polycrystalline KVO3 have been recorded at various temperatures. Splittings have been observed for most fundamentals and the external mode region contains at least eight bands. The spectra are interpreted in terms of a group theoretical analysis based on the molecular symmetry and the known crystal structure. The frequency versus temperature plots for external modes of KVO3 indicate that a single crystal structure exists between 10 and 300 K.

    Notes: 15NH4ReO4 was synthesized and the Raman spectrum recorded at various temperatures between 10 and 300 K. 15N isotopic shifts of both internal and external modes of the ammonium ion relative to 14NH4ReO4 were observed. The temperature dependence of the external modes was studied. The librational lattice mode associated with the ammonium ion disappeared above about 180 K. At this temperature anomalies were observed in the temperature dependence of the perrhenate ion lattice modes.
