    Notes: Comparative analysis was carried out for hydrogen peroxide (H2O2) accumulation in response to powdery mildew (Blumeria graminis[Erysiphe graminis] f.sp. tritici) primary germ tube (PGT) and appressorial germ tube (AGT) contact, and in papilla (cell wall apposition) and hypersensitive responses (HR) in wheat (Triticum aestivum). Using primary leaves of three susceptible wheat lines (Bainong 3217; Beijing 837; Jingshuang 16) and five resistant lines [Mardler (Pm2 + Pm6); Ulka/8 × Cc (Pm2); Mardler/7 × Bainong 3217 (Pm2); Pm16; Pm16/7 × Beijing 837 (Pm16)], early H2O2 accumulation between 10 and 48 h after inoculation was studied. Strong H2O2 accumulation was found in effective papillae and associated cytosolic vesicles in both susceptible and resistant wheat lines, suggesting the important role of H2O2 in effective papillae formed as a general plant defence against powdery mildew. High frequency of effective papilla formation was observed in all five resistant lines. Among resistant lines, hypersensitive cell death was detected earlier in Mardler and Ulka/8 × Cc than in Pm16 and Pm16/7 × Beijing 837. In all cases this was associated with H2O2 accumulation in attacked epidermal cells. Interestingly, penetration resistance but not HR appeared to be mediated by the Pm2 gene in the Mardler/7 × Bainong 3217 line, suggesting that Pm2 may govern an HR-independent defence pathway in this genetic background. As effective papillae and HR did not occur in the same cells, papilla deposition may be independent of the HR response, despite the fact that both defence mechanisms were associated with high H2O2 accumulation. In wheat powdery mildew-incompatible interactions, HR acts as a second line of defence to contain infection when the papilla defence fails.

    Notes: Abstract An experimental study was performed to modify the conventional impact-echo technique to detect small diameter spherical voids in concrete. High velocity impact of steel balls was used to generate the high frequency waves in concrete necessary for detection of small voids. Elastodynamic wave propagation in concrete, type of impact source, and the size of the flaw that can be detected were studied. These laboratory studies were carried out on a 4 in. (10.2 cm) thick 4 ft. × 4 ft. (1.22 m × 1.22 m) concrete slab that contained a variety of artificial spherical flaws embedded at known locations. Analysis of the frequency spectrum of the response of the slab was used to determine the slab's thickness and presence of voids. Signal processing techniques were applied for a better interpretation of the slab's response. Autocorrelation and cross-correlation algorithms were used to determine the characteristics of the slab at different locations. The problems associated with determination of flaws and voids in concrete have been delineated. Some practical and easy to implement solutions have been proposed and ways of using the impact-echo technique more efficiently for field applications have been discussed.

    Notes: Summary A map of chromosome 1R of rye was constructed using 16 molecular and biochemical loci. From long arm to short arm, known-function loci were placed in the order: XAdh — XLee — Glu-R1[Sec-3] — XPpdk-1R — XEm-1R-1 — XEm-1R-2 — Centromere — XNor-R1 —Gpi-R1 — XGli-R1 [Sec-1a] along with six anonymous genomic and cDNA clones from wheat. The map, which spans 106 cM with 12 loci clustered in a 15-cM region around the centromere, shows reasonably good agreement with previously published maps for the centromeric region, whereas the XNor-R1 — Gpi-R1 region gives a much larger distance than previously reported.

    Notes: Abstract A rapidly growing, long-term suspension culture derived from Triticum aestivum L. (wheat) was synchronized using hydroxyurea and colchicine, and a chromosome suspension with chromosomes was made. After staining with the DNA-specific fluorochromes Hoechst 33258 and Chromomycin univariate and bivariate flow-cytometry histograms showed 15 clearly resolved peaks corresponding to individual chromosome types or groups of chromosomes with similar DNA contents. The flow karyotype was closely similar to a histogram of DNA content measurements of Feulgen-stained chromosomes made by microdensitometry. We were able to show the stability of the flow karyotype of the cell line over a year, while a parallel subculture had a slightly different, stable, karyotype following different growth conditions. The data indicate that flow cytometric analysis of plant karyotypes enables accurate, statistically precise chromosome classification and karyotyping of cereals. There was little overlap between individual flow-histogram peaks, so the method is useful for flow sorting and the construction of chromosome specific-recombinant DNA libraries. Using bivariate analysis, the AT:GC ratio of all the chromosomes was remarkably similar, in striking contrast to mammalian flow karyotypes. We speculate about a fundamental difference in organization and homogenization of DNA sequences between chromosomes within mammalian and plant genomes.

    Notes: Summary A genetic map of barley chromosome 5 (1H) was constructed using DNA markers. Seventeen loci were mapped to 15 locations, and these included the known-function loci (in order from the most distal on the long arm) XAdh (alcohol dehydrogenase), XLec (homologous to wheat germ agglutinin), XHor3 (D-hordein), XPpdk (pyruvate orthophosphate dikinase), centromere, XIcal (chymotrypsin inhibitor), and 6 loci in the B- and C-hordein cluster towards the end of the short arm. The gene order on the barley map agreed closely with that of chromosome 1 of rye. Intervarietal comparisons showed that single-copy cDNA and genomic DNA probes revealed about twice the level of RFLPs found in wheat.

    Notes: Abstract Shifted Legendre polynomial functions are employed to solve the linear-quadratic optimal control problem for lumped parameter system. Using the characteristics of the shifted Legendre polynomials, the system equations and the adjoint equations of the optimal control problem are reduced to functional ordinary differential equations. The solution of the functional differential equations are obtained in a series of the shifted Legendre functions. The operational matrix for the integration of the shifted Legendre polynomial functions is also introduced in the simulation step in order to simplify the computational procedure. An illustrative example of an optimal control problem is given, and the computational results are compared with those of the exact solution. The proposed method is effective and accurate.
