Notes: The influence of pH and temperature on the substrate yield coefficient for growth of Saccharomyces cerevisiae in a chemostat under limited organic substrate conditions was studied.Mathematical analysis of the substrate yield coefficient as a function of pH and temperature in the near-optimal area was made. It was shown that the location of pH and temperature optima were independent of each other. The maximum substrate yield coefficient had the following coordinates: pH = 4.1, temperature = 28.5°C.

Notes: We consider the problem of making allowance for superhelicity in the statistical-mechanical calculations of fluctuational violations of the DNA double helix. A simple model is discussed, making it possible in the calculations to use an approach based on the theory of helix-coil transition in DNA. The proposed algorithms allow calculating the effect of superhelicity on the base-pair fluctuational opening for any given sequence of nucleotides. An algorithm is also proposed allowing for the hairpin and cruciform structures in the palindromic regions of a sequence, as well as the open and helical states. The theory is used to calculate the melting curve for superhelical DNA at temperatures well below the melting point of the linear or nicked forms. The maps of opening probability are calculated for SV40 and φX174 DNA using their recently published complete nucleotide sequences. The data explain well the experimental results of probing the secondary structure of these DNA by single strand-specific endonucleases.

Notes: Oligotripeptides Z-(Gly-Pro-Pro)n-OMe (n = 1,2,…,8), Z-Gly-Pro_Pro-Gly-Pro-Gly-OMe, Z-Gly-Pro-Pro-Gly-Pro-Gly-Gly-Pro-Pro-OMe, Z-Gly-Pro-Pro-(Gly-Pro-Gly)2-Gly-Pro-Pro-OMe, and Z-(Gly-Pro-Ala)n-OMe (n = 1,2,…,4) were synthesized step-by-step and then studied by means of x-ray diffraction, ir spectroscopy, the kinetics of hydrogen-deuterium exchange of peptide groups, and circular dichroism,. Different stages in the formation of a triple helix in Z-(Gly-Pro-Pro)n-OMe were revealed during the chain elongation. In the solid state, at the first stage a conformation of the polyproline II-type is formed in the tripeptide and in the second stage a triple helical complex appears in the hexapeptide. Interpeptide hydrogen bonds in this complex are still of low order. At further stages an ordered set of interpeptide hydrogen bonds is gradually formed. It is shown that the degree of order of interpeptide H bonds depends on the length of the molecular chain, the amino acid composition, and residue sequence in the triplets.

Description / Table of Contents: Studies on the Kinetics and the Reaction Heat of the Cyclotrimerization of Aryl CyanatesThe kinetics and the reaction enthalpy of the polycyclotrimerization of 2,2-bis(4-cyanatophenyl)propane in ditolylmethane solution were investigated by means of direct calorimetry, varying the concentration of the catalyst (chromium(III) acetyl acetonate) and of the monomer as well as the reaction temperature, the water content of the solvent, and the amount of added acetyl acetone. The maximum reaction rate is proportional to the monomer concentration, to the square root of the concentration of the catalyst, and to the water content. It is inversely proportional to the amount of water added.Presumably, water participates in the formation of the active species of the catalyst, and the autocatalytic nature of the polycyclotrimerization of 2,2-bis(4-cyanatophenyl)propane in presence of chromium acetylacetonate is due to accumulation of the latter during reaction.

Notes: The Mandel'shtam-Brillouin spectra of single crystals of ZrO2-Y2O3 and HfO2—Y2O3 solid solutions have been measured and the results explained qualitatively. Using the methods of Mandel'shtam-Brillouin spectra and ultrasound propagation the elastic and photoelastic constants of single crystals of ZrO2—Y2O3 and HfO2—Y2O3 solid solutions have been found and their dependence on the composition obtained. The elastic and photoelastic properties of single crystals of these solid solutions are discussed.

Notes: The DNA helix-coil transition has been studied in the presence of high concentrations of manganese ions (about 10-3M), which corresponds to the conditions close to equal stability of the A+T and G+C pairs, at the ionic strengths of 10-1, 10-2, and 1.6 × 10-3M Na+. With the Mn2+ ion effect, the transition range is significantly reduced to not more than 0.2°C at 1.2 × 10-3M Mn2+ and 1.6 × 10-3M Na+. The melting curves display a sharp kink at the end of the helix-coil transition, which is interpreted as an indication of the second-order phase transition. It is shown that the melting curves obtained can be approximated by a simple analytical expression 1 - θ = exp[-a(tc - t)], where θ is the DNA helix fraction, tc is the phase transition temperature, and a is an empirical parameter characterizing the breadth of the melting range and responsible for the magnitude of a jump of the helicity derivative with respect to the temperature at the phase transition point.

Notes: The rheological properties of molten mixtures of polyoxymethylene and copolyamide have been investigated by the methods of capillary viscometry over a wide range of shear stresses. It has been found that at relatively close values of the melt viscosity polyoxymethylene (POM) and copolyamide (CPA) form mixtures which differ markedly in viscosity and elastic properties from the original polymers. Depending on the magnitude of the shear stress, the viscosity of mixtures of definite composition may be either higher or lower than the viscosity of the components. Three- to tenfold swelling of the stream emerging from the capillary is observed for such mixtures, whereas at the same temperatures and stresses no swelling of the POM and CPA extrudates is practically observed. The data obtained are accounted for by the fact that the flow of mixtures through the capillary involves the formation of hundreds of thousands of ultrathin fibres of POM in the matrix of CPA, which are oriented along the flow axis.

Notes: We describe the preparation and some of the properties of heparin-bound α-chymotrypsin that were obtained via activation of heparin with water-soluble carbodiimide. Immobilized enzyme has unchanged kinetic characteristics toward low-molecular-weight and macromolecular substrates. The heparin-bound enzymes could have a wide range of medical applications.
