    Description: Cultured skin fibroblasts from subjects with cystic fibrosis exhibited normal population doubling times in early passages. After about 13 cumulative population doublings, cystic fibrosis lines doubled more slowly than controls and ceased doubling after about 19 weekly passages. Control lines continued doubling for 27 passages. The premature senescence noted in cells from subjects with cystic fibrosis reconciles controversial observations of cell doubling reported in the literature.     Notes: Records of electrical activity in the tracheal muscles of domestic chickens were obtained for a variety of ad libitum vocalizations. Primary attention was given to an analysis of events during the most complex call, crowing.Three pairs of muscles, Mm. tracheohyoideus, tracheolateralis, and sternotrachealis, can affect the configuration of a chicken's syrinx. The firing patterns of the three muscle pairs are related to their different abilities to affect the tension of the syringeal membranes. The influence of M. tracheohyoideus is most indirect and imprecise, and its role the least clearly defined. It appears to adjust the position of the trachea so that the syrinx is isolated from unpredictable and/or undesirable consequences of nuchal position and tracheal elasticity, and also helps draw the glottis caudad, thereby deepening the pharyngeal chamber. The other two muscles interact to control the tension of the vocal membranes. M. sternotrachealis relaxes the membranes by drawing the drum of the trachea caudad, or, via the syringeal ligament, by rotating the pessulus cranioventrad, or both. M. tracheolateralis tenses the membranes and/or prevents caudal movement of the origin of M. sternotrachealis, a necessity if the syringeal ligament is to rotate the pessulus.Vocalization depends on both syringeal configuration and appropriate air flow. Hence, tracheal muscles, syrinx, air sacs, and ventilatory muscles cooperate to form a vocal system. Cooperation elicits a surprising degree of redundancy. At least one call, a high pitched wail, may be produced by two very different techniques.

    Notes: Anaerobic digestion offers a potential means of converting organic solid waste into fuel gas and thereby provide a supplemental and readily utilizable source of energy. We are particularly interested in the use of thermophilic digestion over a mesophilic operation for it can achieve higher rates of digestion, greater conversion of waste organics to gas, faster solid-liquid separation, and minimization of bacterial and viral pathogen accumulation. Our results comparing mesophilic (37°C) and thermophilic (65°C) anaerobic digestion of domestic solid waste confirm the increased rate and conversion of waste to methane. In addition, utilizing radioactive labeling of glucose and acetic acid, we have measured the volumetric rates of volatile acid production and disappearance under both mesophilic and thermophilic conditions.

    Notes: Studies to examine the microbial fermentation of coal gasification products (CO2, H2 and CO) to methane have been done with a mixed culture of anaerobic bacteria selected from an anaerobic sewage digestor. The specific rate of methane production at 37°C reached 25 mmol/g cell hr. The stoichiometry for methane production was 4 mmol H2/mol CO2. Cell recycle was used to increase the cell concentration from 2.5 to 8.3 g/liter; the volumetric rate of methane production ran from 1.3 to 4 liter/liter hr. The biogasification was also examined at elevated pressure (450 psi) and temperature to facilitate interfacing with a coal gasifier. At 60°C, the specific rate of methane production reached 50 mmol/g cell hr. Carbon monoxide utilization by the mixed culture of anaerobes and by a Rhodopseudomonas species was examined. Both cultures are able to carry out the shift conversion of CO and water to CO2 and hydrogen.

    Notes: A natural microbial population of sewage origin was grown at four temperatures (10, 20, 30, 40°C) and five mean residence times (3, 6, 12, 18, 24 hr) in a continuous stirred tank reactor. Samples of cells were removed during steady-state operation and analyzed for chemical oxygen demand (COD), ash content, protein, carbohydrate, and nucleic acids. The oxygen equivalence (OE) of the cells was measured directly as g COD/g ash-free cells and was determined indirectly by calculating the calorific content from the macromolecular composition. The results indicate that OE is a function of growth conditions. Furthermore the COD test is a poor measureof OE because of a high level of variation associated with the test. Additional studies should be performed using bomb calorimetry to establish a quantitative relationship between growth conditions and OE.
