    Notizen: Summary Isolated resistance vessels in the rabbit ear preconstricted with histamine were relaxed by acetylcholine by a proportionately greater amount than the central ear artery. The relaxation was antagonized by atropine and also by endothelium removal. Our studies represent the first direct evidence that endothelium-dependent dilation can occur in resistance vessels.

    Notizen: Abstract Functional studies concerning the unique interaction between class I H-2 allodeterminants and cytolytic T lymphocyte (CTL) antigen receptors have benefitted from the development of H-2Kb mutant mouse strains and somatic H-2 variants selected with monoclonal antibody. Here, we describe the development of a novel approach to immunoselection of somatic H-2Kb variants employing a Kb-specific CTL clone as the negative selective agent. The rationale for this method is that the use of an alloreactive CTL clone as the selective agent should enable us to detect the emergence of structural Kb variants based on their loss of the relevant CTL-defined allodeterminant. Thus, these structural variants are well suited to an in-depth analysis of the functional relationship between H-2 antigens and receptor recognition by CTL. Using this approach, we successfully isolated two types of structural Kb variants, as well as numerous Kb-loss variants. The functional studies described in this paper indicate that these structural variants exhibit alterations in expression of both CTL-defined and serologically defined H-2Kb allodeterminants. The structural characterization of such variants should enable us to identify the precise amino acid residues responsible for the creation of the relevant CTL-defined Kb allodeterminants.

    Notizen: Abstract A tuber disc transformation and regeneration system was devised for potato (Solanum tuberosum). Tuber discs were found to be the most morphogenetic organ on a medium previously optimised for tomato regeneration. Shoot regeneration from tuber discs was rapid and transformed as shown by nopaline assays and Southern blot analysis. The ease and speed of the tuber disc method will allow for the increased use of this commercially important plant in transformation studies.

    Notizen: Abstract The disamed Ti-binary vector pBIN 6 in Agrobacterium tumefaciens has been used in leaf disc transfomations to produce transgenic apple (Malus pumila Mill.) plants with a nomal phenotype except for a somewhat reduced capacity to root. The presence of the genes for nopaline synthase and neomycin phosphotrans ferase (conferring kanamycin resistance), inserted into the host genome by the vector, was confirmed by Southern blot analysis, the detection of nopaline synthase activity and rooting in the presence of the antibiotic. The nopaline synthase gene continued to be expressed in glasshouse-grown plants several months after removal from in vitro growth conditions.

    Notizen: Abstract The binding of trimethoprim and [1,3,2-amino-15N3]-trimethoprim to Lactobacillus casei dihydrofolate reductase has been studied by 15N and 1H NMR spectroscopy. 15N NMR spectra of the bound drug were obtained by using polarisation transfer pulse sequences. The 15N chemical shifts and 1H-15N spin-coupling constants show unambiguously that the drug is protonated on N1 when bound to the enzyme. The N1-proton resonance in the complex has been assigned using the 15N-enriched molecule. The temperature-dependence of the linewidth of this resonance has been used to estimate the rate of exchange of this proton with the solvent: 160±10s-1 at 313 K, with an activation energy of 75 (±9) kJ·mole-1. This is considerably faster than the dissociation rate of the drug from this complex, demonstrating that there are local fluctuations in the structure of the complex.

    Notizen: Abstract Transmission electron microscopy has been used to investigate dislocation structures in deformed binary and ternary Zr3Al-based alloys. In the binary alloy deformed at temperatures between 293 and 673 K the dislocations in the Zr3Al phase consisted of a/3〈1 1 2〉-type partial dislocations bounding superlattice intrinsic stacking fault on {1 1 1} planes. The {111} a/3〈1 1 ¯2〉 stacking fault energy was approximately 2mJ m−2 at 673 K. In binary specimens deformed between 873 and 1073 K cube slip predominated. Dislocations consisted mainly of a/2 〈1 1 0〉 pairs separated by antiphase boundary. For this temperature range the {1 0 0} a/2〈01 1〉 antiphase boundary energy was between 30 and 45 mJ m−2. Alloying with niobium or titanium was found to increase the {111} a/3〈1 1¯2〉 stacking fault energy and thus increase the propensity for antiphase boundary-type dissociation.

    Notizen: Abstract Polygalacturonase (PG) is the major cell wall degrading enzyme of tomato fruit. It is developmentally regulated and is synthesised de novo in ripening fruit. Genomic clones encoding a PG gene of tomato (Lycopersicon esculentum Mill cv. Ailsa Craig) have been isolated, mapped and sequenced. The sequence of the protein-coding region is identical to that of a PG cDNA [20]. Comparison of the cloned restriction fragments with genomic Southern data suggests that there may only be one gene for PG per haploid genome. The PG gene, which covers approximately 7 kb, is interrupted by 8 intervening sequences ranging in size from 99 bp to 953 bp. The transcription start point was identified by S1 mapping and primer extension analysis. About 1.4 kb of 5′ flanking DNA has been sequenced. This contains putative TATA and CAAT boxes and also direct repeat sequences. A transcriptional fusion has been constructed between the putative 1.4 kb promoter fragment and the chloramphenicol acetyl transferase (CAT) gene. Constructs containing this gene have been transferred to tomato using binary vectors. Regenerated transgenic plants express CAT in ripe tomato fruit, but not in unripe tomatoes, leaves, or roots.

    Notizen: Summary Derivatives of potato (Solanum tuberosum cv.'s ‘Maris Bard’ and ‘Desiree’) transformed with disarmed T-DNA from genetically engineered Agrobacterium tumefaciens strains were isolated. The transformed plants were recovered from shoot-forming tumours induced by infection of wounds with mixedcultures of shoot-inducing A. tumefaciens strains T37 and either Agrobacterium strain LBA1834(pRAL1834), (Hille et al. 1983) or LBA4404(pBIN6; pRAL4404), (Bevan 1984). Two small-scale feasibility experiments gave at least four ‘Maris Bard’ plants transformed with pRAL1834 T-DNA and two ‘Desiree’ plants with pBIN6 T-DNA. The transformed ‘Maris Bard’ plants were morphologically abnormal and highly aneuploid. This was probably an unfortunate side-effect of a tissue culture-step introduced to promote the efficiency of shoot regeneration. The transformed ‘Desiree’ plants, in contrast, were isolated without promoting additional shoot-growth. They were morphologically normal, contained 47 and the euploid 48 chromosomes per cell respectively and had improved growth on media containing kanamycin.

    Notizen: Summary A series of deletion mutants of a soybean heat shock (hs) gene promoter was generated and linked to the chloramphenicol acetyl transferase (CAT) coding sequence. These chimaeric promoter/reporter gene constructs were introduced into tobacco and thermoregulated expression of CAT activity was examined in leaf extracts. Three different types of gene fusions were tested using two differentBIN19 vector constructions: (1) translational fusion between the N-terminus of the protein coding sequence of the heat shock geneGmhsp17.3-B and CAT; (2) transcriptional fusions between the 5′ nontranslated RNA regions ofGmhsp17.3-B and CAT; and (3) promoter fusions joining the hs promoter upstream sequences to the TATA box sequence of the Δ CaMV 35S-CATter vector. Alternatively, multiple copies of a synthetic deoxyoligonucleotide with the soybean hs consensus element (HSE2) were used. Heat inducible CAT activities were detected except in plants containing a transcriptional fusion devoid of all but 18 nucleotides at the 5′ terminus of the hs gene transcript. CAT activity was detectable in these plants only during the recovery at 25° C after a hs (40° C). Overlapping HSE-like promoter sequences seem to be necessary for the induction of heat inducible transcription of linked genes; synthetic HSE2 sequences have the capacity to reconstitute a hs promoter in combination with a TATA box sequence. Effective translation during hs seems to require sequences in the 5′ nontranslated leader of the hs protein mRNA; these sequences can be functionally replaced by the 5′ leader sequence of the Δ CaMV 35S promoter.
