    Notes: Abstract Mice of three congenic resistant lines differing from C57BL/10 at theH-3, H-13, H-7, andH-8 minor histocompatibility loci were used to immunize, and were immunized with, C57BL/10. Cytotoxic cells which were capable of causing rapid lysis of cells from the immunizing strain were generated at least one-way in all combinations tested. In order for a target to be susceptible to cytolysis, it had to carry both the sameH-2 b haplotype and the same differential minor histocompatibility alleles as the immunizing strain. That is, B10.C(47N) (H-2 b ,H-7 b ) anti-C57BL/10 (H-2 b ,H-7 a ) cytotoxic cells lysed C57BL/10 targets but not B10.BR (H-2 k ,H-7 a ) targets, nor BALB.B (H-2 b ,H-7 b ) targets. Crossreactions in the cytotoxic assay suggest that theH-3, H-13 congenic pair —B10.LP and C57BL/10 —may differ in at least two more minor histocompatibility loci which have not yet been defined. Although 6 x 1066 C57BL/10 lymphoid cells primed B10.D2(57N) (H-8 b ) mice for a secondary in vitro cytotoxic response, a tenfold higher dose apparently made them tolerant. It is concluded that all minor histocompatibility differences can generate cytotoxic T cells which show specificity both for the minor and major histocompatibility alleles.

    Notes: Abstract Functional studies concerning the unique interaction between class I H-2 allodeterminants and cytolytic T lymphocyte (CTL) antigen receptors have benefitted from the development of H-2Kb mutant mouse strains and somatic H-2 variants selected with monoclonal antibody. Here, we describe the development of a novel approach to immunoselection of somatic H-2Kb variants employing a Kb-specific CTL clone as the negative selective agent. The rationale for this method is that the use of an alloreactive CTL clone as the selective agent should enable us to detect the emergence of structural Kb variants based on their loss of the relevant CTL-defined allodeterminant. Thus, these structural variants are well suited to an in-depth analysis of the functional relationship between H-2 antigens and receptor recognition by CTL. Using this approach, we successfully isolated two types of structural Kb variants, as well as numerous Kb-loss variants. The functional studies described in this paper indicate that these structural variants exhibit alterations in expression of both CTL-defined and serologically defined H-2Kb allodeterminants. The structural characterization of such variants should enable us to identify the precise amino acid residues responsible for the creation of the relevant CTL-defined Kb allodeterminants.

    Notes: Abstract We have studied the T cell-mediated lysis of two C58 lymphoma lines: R1(TL+), which bears serologically detectable H-2k and TL1,2,3 antigens, and R1(TL−), an immunoselected variant which lacks these antigens. Unlike R1(TL+) cells, the variant cells are not sensitive to specific lysis by T cells directed against either H-2k or minor H antigens of C58 mice. An injection of R1(TL+) cells into allogeneic H-2-identical or H-2-different mice primes for an excellent secondary cytotoxic response in vitro to R1(TL+); but not to R1(TL−). Immunization in vivo and in vitro with R1(TL)−) leads to little or no priming or generation of cytotoxic T cells. Both cell lines, however, are sensitive to nonspecific lysis by cytotoxic cells in the presence of PHA or Con A, although even under these conditions, R1(TL+) is killed more effectively than is R1(TL−). We conclude that R1(TL−) does not express any form of H-2 antigen which can be detected by immunization or by sensitivity to cytotoxic T cells.

    Notes: Total internal reflection microscopy is a technique for monitoring changes in the distance between a single microscopic sphere and a flat plate by measuring the intensity of light scattered by the sphere when illuminated by an evanescent wave. A histogram of scattering intensities can be used to construct the potential energy profile as a function of distance relative to the most probable distance. Thus potential energies can be measured to within a fraction of kT while changes in distance can be measured to within 1 nm. An autocorrelation of the scattering intensities can be used to deduce an average diffusion coefficient of the sphere, which is found to be only a few percent of the Stokes–Einstein value, owing to the close proximity of the plate. The analysis of the intensity-autocorrelation function presented here can be used to deduce an absolute value for the most probable separation distance, without a priori knowledge of the functional form of the PE profile and in the presence of a constant background scattering intensity. This "hydrodynamic" separation distance is found to be within a few percent of the "optical" separation distance found independently by comparing the intensity at the most probable distance with the intensity of the same particle in contact with the plate. Since the particle does not need to be brought into contact with the plate, the hydrodynamic method is well suited for determining the absolute separation distance with deformable particles like liquid droplets, vesicles or biological cells. Moreover, the hydrodynamic separation can be immediately calculated without any additional experiments. However, accurate determination of the hydrodynamic separation requires an accurate value for the particle size, which must be determined independently. © 2000 American Institute of Physics.
