Notes: Information is presented concerning revisions of the fermentation air system equipment which improved the sterility situation in the production of monosodium glutamate.Basically, the revision were a relocation and elevation of the intake to the air compressors and installation of a retention chamber after the compressors to take advantage of the heat of compression. The extended high temperature retention ensures sterile air to the final air filter before the fermentor. Contamination losses - mainly from phage - have been materially reduced relative to the level prevailing before the change.

Notes: The recovery of copper from synthetic aqueous media circulating in a loop fluidized bed reactor operated batchwise was investigated by using the following biopolymer systems: (1) a viscous solution of sodium alginate (from kelp) dispensed directly into the reactor fluid containing dissolved copper (sulfate salt) at initial concentrations of 60-200 ppm, (2) partially coagulated calcium alginate spheres for absorbing dissolved copper at initial concentrations of 10-40 ppm, and (3) a mixture of green algae Microcystis and sodium alginate dispensed directly into the reactor fluid. The recovery of copper and cobalt, a strategic metal, from cobalt ore leachate was achieved by a two-step approach: direct dispensing of sodium alginate to absorb the bulk of metals followed by the addition of partially coagulated calcium alginate spheres to “polish” the leachate. Metal binding capacity and conditional stability constant of each biopolymer system as well as the effective diffusivity of cupric ion in the matrix of biopolymer gels are reported.

Notes: A new apparatus for continuously detecting fluorescently labeled DNA fragments is based on infrared fluorescence technology. This technology combines state-of-the-art developments in chemistry, laser technology, and detection, while achieving improved reliability, sensitivity, and flexibility for applications including DNA sequencing. DNA molecules labeled with a novel infrared fluorophore are detected during electrophoresis using a scanning infrared fluorescence microscope. The microscope consists of a laser diode for exciting the fluorophore and a silicon avalanche photodiode for detecting the infrared emission. Optimum conditions for detection and throughput are obtained by adjusting electrophoresis, scanning and imaging parameters. Typical DNA sequencing runs (test templates) allow identification of over 500 bases per sample with 〉 99% accuracy.

Description: The objectives of the Apollo 17 Lunar Sounder Experiment (ALSE) were to detect subsurface geologic structures, to generate a continuous lunar profile, and to image the moon at radar wavelengths. A three-wavelength synthetic-aperture radar (SAR) operating at 60, 20, and 2 m wavelengths was designed to attain these objectives. The design choices reflected a balance of scientific requirements versus Apollo mission and hardware constraints. The radar data from the lunar mission were recorded on photographic film in a conventional SAR format, and were returned to earth for processing. A combination of optical and digital processing and exploitation techniques was applied to the scientific interpretation of the data. Some preliminary results from the lunar mission have been obtained.

Description: A color-television display system and interactive graphics equipment on-line to an IBM 360/44 computer are used to develop a variety of interactive displays which aid in analyzing remote sensor data. These interactive displays are used to: (1) analyze data from a multispectral scanner; (2) develop automatic pattern recognition systems based on multispectral scanner measurements; and (3) analyze data from non-imaging sensors such as the infrared radiometer and microwave scatterometer.

Description: Applications using a three-dimensional embedded grid scheme are made to high angle of attack viscous flow over two bodies: a slender cone using the conical approximation and a 75 deg swept delta wing. The embedded grids are used principally to improve the numerical resolution of the separated vortical flow above the body. Detailed comparisons are made with experimental measurements of the velocity field over the delta wing. The prediction of the maximum steamwise velocity is improved using two levels of embedded grid refinement but is still less than the experimental measurements available from a laser velocimeter.

Description: Aerodynamic drag tests were performed on a conventional cab-over-engine tractor with a 45-foot trailer and five commercially available or potentially available add-on devices using the coast-down method. The tests ranged in velocity from approximately 30 miles per hour to 65 miles per hour and included some flow visualization. A smooth, level runway at Edwards Air Force Base was used for the tests, and deceleration measurements were taken with both accelerometers and stopwatches. An evaluation of the drag reduction results obtained with each of the five add-on devices is presented.

Description: A method for analysis and classification of verbal communications typically associated with manned space missions or simulations was developed. The study was carried out in two phases. Phase 1 was devoted to identification of crew tasks and activities which require voice communication for accomplishment or reporting. Phase 2 entailed development of a message classification system and a preliminary test of its feasibility. The classification system permits voice communications to be analyzed to three progressively more specific levels of detail and to be described in terms of message content, purpose, and the participants in the information exchange. A coding technique was devised to allow messages to be recorded by an eight-digit number.

Notes: Optimization methods based on the continuous maximum principle and the calculus of variations were used to calculate optimum temperature profiles for batch penicillin fermentations. These methods were first applied to several general models to develop effective techniques for the numerical solution of the equations. Subsequently, these methods were applied to two particular models, derived from experimental data, and the optimum temperature profiles were determined. The results indicated that an improvement, in penicillin yield of about 15% was possible if the optimum temperature profiles were followed.
