    Notizen: Several techniques for investigating the multiplicity and stability of open isothermal enzymatic reactors are discussed and some of the pitfalls in previous thinking pointed out. The example which is used to illustrate these methods exhibits several interesting features. Among these is the existence of a stable oscillatory state which surrounds a unique steady state which is asymptotically stable to certain finite disturbances.

    Notizen: The optimal production of the fructan biopolymer levan by the bacterium Erwinia herbicola was investigated, including variations in nitrogen, carbon and phosphorous sources, pH, incubation time, culture yields up to 19% by weight produced based on conversion of sucrose as the carbon source when grown in a continuous culture system and processed by tangential flow filtration. Product identity was confirmed with gas chromatography (GC) and 13C nuclear magnetic resonance (NMR). Gel permeation chromatography (GPC) and low-angle laser light scattering (LALLS) determination of the molecular weight of the product showed a significant difference in molecular weight values dependent on the method of analysis. Analysis by GPC resulted in molecular weight one order of magnitude lower than LALLS independent of sample, underscoring the unusual nature of this biopolymer.

    Notizen: In this study, different mole fractions of pure Thermomonospora fusca E5 and E3, plus Trichoderma reesei CBHI were studied for reducing sugar production at 2 h, degree of synergism, and cellulose binding. In addition, the effects of introducing the Caldocellum saccharolyticum β-glucosidase into this cellulase system were investigated. The cellulases used were purified to homogeneity. Avicel PH 102 (4% w/w solution in 0.05 sodium acetate pH 5.5 buffer) was the substrate. Reactions were run at 50°C for 2 h using total cellulase concentrations of 8.3 or 12.2 μM. A bimixture of T. fusca E3 and T. reesei CBHI was very effective in hydrolyzing microcrystalline cellulose (9.1% conversion). The addition of endoglucanase E5 to the mixture only increased conversion to 9.8%. However, when both E5 and β-glucosidase were added, conversion increased to 14%. It was also observed that increasing total cellulase concentration beyond 8.3 μM did little to increase percent conversion of cellulose into glucose. The results of the binding studies indicate no competition for binding sites between the endo- and exocellulases. © 1993 John Wiley & Sons, Inc.

    Notizen: Gram-sized quantities of purified arginine, formylmethionine, glutamic acid, and phenylalanine-2 tRNAs have been prepared from pools of E. coli K-12 MO7 mixed tRNAs by reversed-phase chromatography after preliminary fractionation on DEAE-cellulose. Purified formylmethionine tRNA and partially purified arginine tRNA and glutamie acid tRNA were obtained from large-scale RPC-3 runs (4 × 36 in. column). The arginine tRNA was further purified by rechromatography on RPC-4 columns, and the gluatmic acid tRNA by rechromatography on an RPC-3 column. Two phenylalanine tRNAs were resolved on large-scale (2 × 96 in. column) RPC-3 runs; only the second phenylalanine tRNA reached a satisfactory degree of activity. About 0.88 g of arginine tRNA, 70% activity; 3.32 g of formylmethionine tRNA, 97% activity; 0.80 g of glutamic acid tRNA, 83% activity and O.92 g of phenylalanine-2 tRNA, 78% activity, were produced. The processing steps employed are reliable and reproducible and the procedure is amenable to routine production of these tRNAs.

    Notizen: The method of normalization is used to develop the J-R curve fracture toughness characterization for polymeric materials. This method can develop J-R curves directly from load vs. displacement records without a need for an on-line crack monitoring system. It was used previously to develop J-R curves for metallic materials and is applied here for the first time to polymer materials. Single edge notched bend specimens of rubber toughened nylon 6/6 and rubber toughened amorphous nylon are used in this study. The J-R curves from the method of normalization are compared with the results obtained from the multiple specimen method of ASTM Standard E813. The results show that the method of normalization gives reasonable J-R curves; both methods show agreement over the early J-R curve region. In addition the JIc values are determined for each method and compared. Based on this work it is suggested that the method of normalization could be used as a general test method to develop J-R curves for polymeric materials.

    Notizen: Mixtures of 1,1,3,3-tetrafluoroacetone and perfluorodi-n-propyl ketone have been photolyzed together over the temperature range 50° to 200°C, and the disproportionation/combination ratio for n-C3F7 and CF2H radicals has been determined to be Δ(n-C3F7, CF2H) = 0.072 ± 0.003. A reevaluation of existing data on CH3 and CF2H radicals leads to a value of Δ(CH3, CF2H) = 0.35. The large variations in Δ for the reactions of alkyl and perfluoroalkyl radicals with CF2H radicals are discussed. © John Wiley & Sons, Inc.

    Notizen: A redetermination of the disproportionation/combination ratio for n-C3F7 and C2H5 radicals gives a value of Δ(n-C3F7, C2H5) = 0.13 ± 0.01, independent of the temperature. The radicals were produced by the photolysis of n-C3F7COC2H5. The previous determinations of this ratio are discussed and are found to be largely incorrect. The values for Δ(CF3, C2H5) and Δ(C2F5, C2H5) are also re-evaluated, and the recommended values are 0.10 ± 0.02 and 0.12 ± 0.02, respectively. Systems involving perfluoroalkyl and ethyl radicals are complicated due to rapid perfluororadical addition to the ethylene formed in the disproportionation process. The extent of this reaction, and its consequences, are discussed and evaluated. The role of the propionyl (C2H5CO) radical in the room temperature photolysis is also assessed. However, it is found that the Δ values determined by the intercept method used in this work are not affected by the secondary reactions that occur. It is concluded that high cross-combination ratios are general to perfluoroalkyl-alkyl radical interactions. For C3F7 and C2H5 radicals the ratio is 2.7-2.8. Above 100°C ratios exceed 3 due to secondary reactions.

    Notizen: Ion beam mixing was used to improve the adhesion between deposited Cu film (400 Å) and polyimide (PI) substrate. Ar+ ion with the energy levels between 180 and 200 keV, and the dose between 1014 to 4 × 1016 ions/cm2 were used. The surface analyses were carried out by Rutherford Backscattering Spectroscopy (SEM). RBS analysis, using 2 MeV He+ ions, showed mixing of Cu and FI and the mixing depended on the Ar+ energy and dose. The X-ray study showed a very broad halo for deposited Cu film but the (111) peak appeared after the Ar+ implantation and the peak increased with Ar+ ion dose. Optical micrographs showed that Cu film formed circular bubbles after many thermal cycles when adhesion was poor and fracture cracks when adhesion was good.
