    Notizen: We describe the preparation and some of the properties of heparin-bound α-chymotrypsin that were obtained via activation of heparin with water-soluble carbodiimide. Immobilized enzyme has unchanged kinetic characteristics toward low-molecular-weight and macromolecular substrates. The heparin-bound enzymes could have a wide range of medical applications.

    Notizen: The preparation and some properties of microspheres composed of oxidized polysaccharides and some vinyl polymers are described. The microspheres contain immobilized enzyme and can be slowly solubilized in water solutions, thereby releasing active a enzyme into the surrounding medium. The kinetic characteristics of the immobilized enzyme bound with a fragment of matrix after complete solubilization are unchanged, but the enzyme exhibits high thermostability. These preparations could have a wide range of medical applications, e.g., to form a drug “depot” directly in an affected organ.

    Notizen: Boron and germanium δ-doped silicon samples were studied using SIMS depth profiling on a Cameca IMS-4F instrument with O2+, N2+ and Cs+ primary beams at various energies and incidence angles. The depth resolution characteristics were compared. We show that, with experimental conditions being the same, the N2+ primary beam provides for better decay lengths, while the leading edges of the profiles turn out to be significantly broadened because of the beam-induced roughness which develops at an early stage of silicon bombardment by N2+ ions. The influence of ripple-like roughness on the SIMS profile shape is considered within the present simple analytical model. The effect of a modified layer on the SIMS profile depth was experimentally investigated. For profiles obtained with an N2+ primary beam, it was shown that swelling of the modified layer produced by the implantation and trapping of primary particles shifts the profile as a whole toward the surface.

    Notizen: Nitroxide spin labels and probes were applied to measurements of cell redox activities and oxygen concentration by ESR. Current experimental methods often require large numbers of cells and monitoring of only the amplitude of the ESR signal. This can result in experimental artifacts and data misinterpretation. An improved method is described for accurate measurement of nitroxide intensity and linewidth, which in turn provides information about cell redox activities and oxygen concentration. The method is based on fitting of ESR spectra using a fast convolution least-squares algorithm suitable for inhomogeneously broadened ESR lines. All fitting parameters, including the Lorentzian component of the linewidth and the double integrated intensity, are extracted from the experimental spectra directly. The experiments were carried out at cell concentrations of 5 × 106 cell ml-1 or less to prevent cell crowding and at initial nitroxide concentration of 1 mM or 100 μM to ensure that nitroxide metabolism by the cells was not diffusion limited. Examples include experiments with cultured baby hamster kidney and adenovirus transformed hamster cells using the nitroxide radicals TEMPOL (2,2,6,6-tetramethylpiperidine-N-oxyl-4-ol) and 15N-labeled perdeuteriated TEMPONE (15NPDT) (2,2,6,6-tetramethylpiperidine-N-oxyl-4-one). Under these experimental conditions, the rates of nitroxide metabolism derived from changes in nitroxide intensity are independent of oxygen concentration.

    Notizen: A system has been developed which allows the selection of integrative transformants with replacement of the Hansenula polymorpha methanol oxidase gene (MOX) with expression cassettes carrying heterologous gene under the control of the MOX promoter. The system is convenient for comparison of the expression levels of different constructs integrated into the same locus of the H. polymorpha genome. This system was used to compare the secretion levels of human urinary plasminogen activator, the secretion of which was directed by different signal sequences.

    Notizen: Beta-amyloid peptides (Aβ peptides) form the main protein component of the amyloid deposits found in the brains of Alzheimer's disease (AD) patients. Soluble Aβ peptides, which are proteolytic fragments of the amyloid-precursor protein (APP) are constitutively secreted by cells expressing APP during normal metabolism [1] and are also present in human plasma and cerebrospinal fluid [2]. Missense mutations in Codon 717 of the APP gene are reponsible for a small percentage of inherited AD cases (FAD) and increase the amount of Aβ peptides containing additional carboxy terminal amino acids (Aβ1-42, Aβ1-43) [3, 4]. Recent findings indicate that FAD mutations in the presenilin 1 and 2 genes also increase the amount of these longer Aβ peptides [5]. Aβ1-42 polymerizes more rapidly in vitro [6] than Aβ1-40 and has been identified as the major component of the brain amyloid deposits [7-9]. We recently developed a sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) system [10] for the separation of these two peptides. Here we describe a modified version of the original SDS-PAGE procedure, which allows the separation of Aβ1-40, Aβ1-42, and Aβ1-43 for the first time. Detection of the three Aβ peptides in the lower ng and pg range is realized by optimized silver staining or immunoblot procedures. These nonradioactive methods may validate results obtained by ELISA procedures used to study the metabolic fate of APP. They may help to define the neurotoxic potential of the longer Aβ peptides in relation to their aggregation state.
