    Description: The current model of apoptosis holds that upstream signals lead to activation of downstream effector caspases. We generated mice deficient in the two effectors, caspase 3 and caspase 7, which died immediately after birth with defects in cardiac development. Fibroblasts lacking both enzymes were highly resistant to both mitochondrial and death receptor-mediated apoptosis, displayed preservation of mitochondrial membrane potential, and had defective nuclear translocation of apoptosis-inducing factor (AIF). Furthermore, the early apoptotic events of Bax translocation and cytochrome c release were also delayed.     Description: Proteins containing membrane attack complex/perforin (MACPF) domains play important roles in vertebrate immunity, embryonic development, and neural-cell migration. In vertebrates, the ninth component of complement and perforin form oligomeric pores that lyse bacteria and kill virus-infected cells, respectively. However, the mechanism of MACPF function is unknown. We determined the crystal structure of a bacterial MACPF protein, Plu-MACPF from Photorhabdus luminescens, to 2.0 angstrom resolution. The MACPF domain reveals structural similarity with poreforming cholesterol-dependent cytolysins (CDCs) from Gram-positive bacteria. This suggests that lytic MACPF proteins may use a CDC-like mechanism to form pores and disrupt cell membranes.     Description: Energetic young pulsars and expanding blast waves [supernova remnants (SNRs)] are the most visible remains after massive stars, ending their lives, explode in core-collapse supernovae. The Fermi Gamma-Ray Space Telescope has unveiled a radio quiet pulsar located near the center of the compact synchrotron nebula inside the supernova remnant CTA 1. The pulsar, discovered through its gamma-ray pulsations, has a period of 316.86 milliseconds and a period derivative of 3.614 x 10(-13) seconds per second. Its characteristic age of 10(4) years is comparable to that estimated for the SNR.     Description: Microquasars are accreting black holes or neutron stars in binary systems with associated relativistic jets. Despite their frequent outburst activity, they have never been unambiguously detected emitting high-energy gamma rays. The Fermi Large Area Telescope (LAT) has detected a variable high-energy source coinciding with the position of the x-ray binary and microquasar Cygnus X-3. Its identification with Cygnus X-3 is secured by the detection of its orbital period in gamma rays, as well as the correlation of the LAT flux with radio emission from the relativistic jets of Cygnus X-3.     Description: In another work, the “open safe” of the HAZUS-MH methodology was cracked to create seismic vulnerability functions that honor all HAZUS-MH methodologies and data, yet that appear in the form of tables of mean casualty rates (indoor deaths and injuries as four fractions of total occupancy) versus a structure-independent intensity measure, in particular, [Formula: see text] or [Formula: see text]. In this work, mean repair cost is tabulated against both these intensity measures, for various combinations of model building type, code design level, occupancy class, seismic environment, NEHRP site soil class, magnitude range and distance range.

    Description: The Applied Technology Council is adapting PEER's performance-based earthquake engineering methodology to professional practice. The methodology's damage-analysis stage uses fragility functions to calculate the probability of damage to facility components given the force, deformation, or other engineering demand parameter (EDP) to which each is subjected. This paper introduces a set of procedures for creating fragility functions from various kinds of data: (A) actual EDP at which each specimen failed; (B) bounding EDP, in which some specimens failed and one knows the EDP to which each specimen was subjected; (C) capable EDP, where specimen EDPs are known but no specimens failed; (D) derived, where fragility functions are produced analytically; (E) expert opinion; and (U) updating, in which one improves an existing fragility function using new observations. Methods C, E, and U are all introduced here for the first time. A companion document offers additional procedures and more examples.

    Description: The HAZUS-MH Technical Manual provides a method and data for calculating earthquake loss to ordinary buildings using in part the capacity spectrum method (CSM) of structural analysis, but it does not provide tabular results relating loss to structure-independent intensity measures such as [Formula: see text] or [Formula: see text], and no procedure for doing so is offered. It is a minor challenge to perform HAZUS-MH loss calculations outside of HAZUS-MH, owing to the sometimes iterative nature of CSM. A technique to calculate mean loss (here, casualty rates in ordinary buildings) as a function of site-soil-adjusted [Formula: see text] and [Formula: see text] is presented that honors all HAZUS-MH methods and data. The resulting seismic vulnerability functions are available at www.risk-agora.org as a resource for open risk modeling. Such vulnerability functions facilitate loss analyses by de-coupling the calculation of hazard from that of loss given hazard.
