    Notes: A new approach for pulse labelling of plants using the short-lived positron emitting radioisotope carbon-11 (half-life: 20.4 min) as 11CO2 is reported together with its application to measuring [11C]isoprene emissions from intact leaves capturing information associated with: (1) rate of emission; (2) the relative contribution of recently fixed carbon to isoprene biosynthesis; and (3) the transit time for tracer movement through the leaf and biochemical pathways associated with isoprene biosynthesis. This approach was applied to study the response of certain Populus species to exogenous treatments of jasmonic acid (JA), a plant hormone implicated in signal transduction linked to defence response against herbivory. Twelve hours after treatment of single intact leaves of aspen (Populus tremuloides) with a 1 m m JA spray, isoprene emissions from those leaves increased 1.5 times the controls from 35.4 ± 2.2 to 53.1 ± 4.8 nmol m−2 s−1. [11C]Isoprene emissions from the same leaves, reflecting the isoprene that was derived from recently fixed carbon, increased much more, to 2.2 times the controls. This increase coincided with a change in emitted [11C]isoprene from 0.31 to 0.68% of 11C fixed in the leaf tissue, while the tracer transit time remained constant at 12.5 min. These results suggest that JA had no effect on enzyme kinetics involved in isoprene biosynthesis, but did impact the source of recent carbon feeding that pathway. Studies with poplar (Populus nigra clone NC 5271) showed similar trends in systemic emissions (from an untreated leaf on the same plant).

    Notes: Regulated expression of structural genes involved in yeast phospholipid biosynthesis is mediated by inositol/choline-responsive element (ICRE) upstream motifs, bound by the heterodimeric activator complex Ino2 + Ino4. Gene repression occurs in the presence of sufficient inositol and choline, requiring an intact Opi1 repressor which binds to Ino2. For a better understanding of interactions among regulators, we mapped an 18 aa repressor interaction domain (RID, aa 118–135) within Ino2 necessary and sufficient for binding by Opi1. By alanine scanning mutagenesis of the entire RID we were able to identify nine residues critical for Opi1-dependent repression of Ino2 function. Consequently, the corresponding dominant Ino2 variants conferred constitutive expression of an ICRE-dependent reporter gene and were no longer inhibited even by overproduction of Opi1. Interestingly, Ino2 RID partially overlaps with transcriptional activation domain TAD2. As certain mutations exclusively affect repression while others affect both repression and activation, both functions of Ino2 can be functionally uncoupled. Correspondingly, we mapped the RID–binding activator interaction domain (AID, aa 321–380) at the C-terminus of Opi1 and introduced missense mutations at selected positions. An Opi1 variant simultaneously mutated at three highly conserved positions showed complete loss of repressor function, confirming RID–AID interaction as the crucial step of regulated expression of ICRE-dependent genes.

    Notes: A procedure for non-destructive inspection by taking into account the effects of stochastic fatigue crack growth is presented. For this purpose a Markoff chain model is used to update the crack statistics, allowing an efficient computational implementation and a clear account of the involved concepts. In this context, perfect and imperfect removal of cracks, respectively, are considered, which represent idealizations defining bounds for actual situations.

    Description / Table of Contents: Summary A method is described for determining the trace elements in soils, plants, feeds, and fertilizers. The trace elements are separated from the major constituents and concentrated by shaking with complex-forming compounds, after destroying the organic material, the concentrate obtained is dissolved in acid. These solutions are brought continuously on a disk of spectrum carbon and simultaneously spectrographed in sparks. The evaluation is made by comparison with solutions of known content in the usual manner.
    Abstract: Résumé Description d'une méthode de dosage des oligoéléments dans les sols, les plantes, la nourriture et les engrais. Les éléments sont séparés de la matière à analyser, concentrés par agitation avec des substances complexantes. Après destruction de la matière organique les concentrés obtenus sont dissouts dans un acide. Ces solutions sont amenées de façon continue sur un disque en carbone pur et analysées par spectrographie d'étincelles. La détermination se fait par comparaison avec des solutions étalons suivant la méthode classique.
    Notes: Zusammenfassung Es wird eine Methode zur Bestimmung der Spurenelemente in Böden, Pflanzen, Futter- und Düngemitteln beschrieben. Die Spurenelemente werden von den Hauptbestandteilen getrennt, durch Ausschüttelung mit komplexbildenden Verbindungen angereichert, nach Zerstörung der organischen Substanz die erhaltenen Konzentrate in Säure gelöst. Diese Lösungen werden auf einer Scheibe aus spektralreiner Kohle kontinuierlich aufgebracht und gleichzeitig im Funken spektrographiert. Die Auswertung erfolgt durch Vergleich mit Lösungen bekannten Gehaltes in üblicher Weise.

    Notes: Abstract The Saccharomyces cerevisiae nuclear gene MRP-L6 was cloned by complementation of the respiratory-deficient mutant pet-ts 2523 with a library of wildtype yeast genomic DNA. The isolated gene was part of a 3.8-kb sequenced DNA fragment containing, in addition to MRP-L6, two unassigned reading frames, ORF1 and ORF2. MRP-L6 codes for a basic protein of 205 amino acids and a molecular mass of 22.8 kDa. The protein exhibits significant sequence similarity to the ribosomal protein L6 of bacteria and chloroplasts. Unlike the corresponding bacterial proteins, however, the MRP-L6 protein (MRP-L6p) contains at its N-terminus a 16 amino-acid leader sequence exhibiting the known characteristics of mitochondrial import signals. Disruption of MRP-L6 leads to the phenotype of a mitochondrial translation-defective, rho-negative yeast mutant. The results are consistent with MRP-L6p representing an essential component of yeast mitochondrial ribosomes. Expression of MRP-L6 was examined, under conditions of glucose repression and derepression, in wild-type cells and in a series of catabolite repression-defective yeast mutants. In most cases, a distinct though small influence of the carbon source on the expression of an MRP-L6/lacZ reporter construct was observed.

    Notes: Zusammenfassung In der vorliegenden Arbeit wurde erneut die Frage nach dem Zusammenhang zwischen der Farbe der Natriumeisenpyroxene und ihrer chemischen Konstitution aufgeworfen. Da die früheren Versuche, diese Frage auf analytischem Wege zu lösen, nicht zum Ziele geführt hatten, wurde hier das reine Endglied NaFeIIIISi2O6 synthetisch hergestellt; in weiteren Versuchen wurden Na und Fe3+ durch andere Ionen zu einem kleinen Teil ersetzt und der Einfluß auf die Farbe ermittelt Es wurden Versuche durchgeführt, bei denen Na durch Ca oder Cd, Fe3+ durch Mg, Mn2+, Fe2+, Co, Ni oder Zn ersetzt wurde; bei weiteren Versuchen wurde anstelle von Fe3+ zu einem kleinen Teil sowohl eines der obengenannten 2wertigen Ionen als auch Ti4+ oder Zr4+ eingebaut. Die erhaltenen Mischkristalle wurden optisch und röntgenographisch untersucht. Zur näheren Charakterisierung dieser Mischglieder wurde in einer Reihe von Schliffen das Absorptionsspektrum in verschiedenen kristallographischen Richtungen bestimmt und mit den ebenfalls aufgenommenen Absorptionskurven von natürlichen Ägirinen verglichen. Auf Grund der synthetischen Versuche werden folgende Definitionen vorgeschlagen: Die Namen Akmit und Ägirin sollen weiterhin für Natriumeisenpyroxen nebeneinander beibehalten werden, wobei die Abgrenzung gegenüber Ägirinaugit gemäß dem Vorschlag vonTröger (2) vorgenommen werden kann. Für die Unterscheidung der beiden Varietäten soll gemäß den bisherigen Gepflogenheiten die Farbe maßgebend sein. Das reine Endglied NaFeIIISi2O6 ist braun und muß deshalb als Akmit bezeichnet werden. Ferner sind mit Akmit die Mischkristalle zu bezeichnen, bei denen das Fe3+ durch Mn ersetzt ist, da auch diese Glieder braune Farbe aufweisen; die Ansicht vonGrout konnte also durch synthetische Versuche bestätigt werden. Die grüne Farbe der Ägirine in RichtungX undY wird durch die gleichzeitige Anwesenheit von 2- und 3wertigem Eisen hervorgerufen, so daß als Ägirin diejenigen Mischkristalle zu bezeichnen sind, bei denen 3wertiges Eisen durch zwertiges ersetzt ist. Der Einfluß des 2wertigen Eisens überwiegt dabei gegenüber dem Einfluß anderer Ionen. Das häufigere Auftreten des Ägirin gegenüber Akmit hat also seine Ursache darin, daß in der Natur 2wertiges Eisen praktisch stets vorhanden ist. Die Gitterkonstanten von Akmit und Ägirin sind im Bereich der hier erzielten Meßgenauigkeit gleich und wurden wie folgt bestimmt:a = 9,66 Åb = 879 Åc = 5,26 Åβ = 72,7 ° Die Arbeit wurde gefördert durch eine Spende keramischen Materials zum Bau der Versuchsöfen, wofür ich auch an dieser Stelle der Fa.     Notes: Abstract The ICL1 gene encoding the isocitrate lyase from Saccharomyces cerevisiae was cloned and sequenced. A reading frame of 557 amino acids showing significant similarity to isocitrate lyases from seven other species could be identified. Construction of icl1 null mutants led to growth defects on C2 carbon sources while utilization of sugars or C3 substrates remained unaffected. Using an ICL1-lacZ fusion integrated at the ICL1 locus, a more than 200-fold induction of β-galactosidase activity was observed after growth on ethanol when compared with glucose-repressed conditions. A preliminary analysis of the ICL1 upstream region identified a 364-bp fragment necessary and sufficient for this regulatory phenotype. Sequence motifs also present in the upstream regions of co-regulated genes were found within this region.

    Notes: Summary Fructose-1,6-bisphosphatase is a key enzyme in gluconeogenesis and the FBP1 gene is not transcribed during growth with glucose. Genetic analysis indicated a positive regulation of FBP1 expression after exhaustion of glucose. By linker-deletion analysis, two upstream activation sites (UAS1 and UAS2) were localized and the respective UAS-binding factors (DAP I and DAP II for derepression activating protein) were identified by gel retardation. UAS1 and UAS2 span about 30 bp each, and are separated by approximately 30 bp. Both UAS sites act synergistically. Although UAS1 showed some similarities to the DNA-binding consensus for the general yeast activator Rap1, competition experiments and DEAE-chromatography proved that DAP I and Rap1 correspond to different proteins. Gel retardation by DAP I depended on carbon sources and did not occur in cells growing logarithmically with glucose, whereas a strong retardation signal was obtained with ethanol-grown cells. The present results suggest that DAP I and DAP II are the final regulatory elements for glucose derepression.

    Notes: Abstract It is well known that the microstructure of metal-matrix composite materials is significantly different from that of the unreinforced matrix. Heat treatments which optimized strength values in the matrix therefore, no longer do so in the composite. It is thus beneficial to find these variations in the heat treatment process, and to pinpoint the microstructural mechanisms responsible. It is known that, in addition to a higher dislocation density, the composite also has a very fine grain size and many whisker interfacial nucleation sites. This study has found that the time required for solutionizing appears to be much shorter in the composite primarily due to the very small grain size. It was also observed that artificial ageing was not very effective in the composite since the precipitates nucleated and grew on the whisker interfaces, therefore, only natural ageing was necessary to achieve peak tensile and yield strengths.
