    Description: Pancreatic cancer is a highly lethal malignancy with few effective therapies. We performed exome sequencing and copy number analysis to define genomic aberrations in a prospectively accrued clinical cohort (n = 142) of early (stage I and II) sporadic pancreatic ductal adenocarcinoma. Detailed analysis of 99 informative tumours identified substantial heterogeneity with 2,016 non-silent mutations and 1,628 copy-number variations. We define 16 significantly mutated genes, reaffirming known mutations (KRAS, TP53, CDKN2A, SMAD4, MLL3, TGFBR2, ARID1A and SF3B1), and uncover novel mutated genes including additional genes involved in chromatin modification (EPC1 and ARID2), DNA damage repair (ATM) and other mechanisms (ZIM2, MAP2K4, NALCN, SLC16A4 and MAGEA6). Integrative analysis with in vitro functional data and animal models provided supportive evidence for potential roles for these genetic aberrations in carcinogenesis. Pathway-based analysis of recurrently mutated genes recapitulated clustering in core signalling pathways in pancreatic ductal adenocarcinoma, and identified new mutated genes in each pathway.     Description: The non-canonical NF-kappaB pathway forms a major arm of NF-kappaB signalling that mediates important biological functions, including lymphoid organogenesis, B-lymphocyte function, and cell growth and survival. Activation of the non-canonical NF-kappaB pathway involves degradation of an inhibitory protein, TNF receptor-associated factor 3 (TRAF3), but how this signalling event is controlled is still unknown. Here we have identified the deubiquitinase OTUD7B as a pivotal regulator of the non-canonical NF-kappaB pathway. OTUD7B deficiency in mice has no appreciable effect on canonical NF-kappaB activation but causes hyperactivation of non-canonical NF-kappaB. In response to non-canonical NF-kappaB stimuli, OTUD7B binds and deubiquitinates TRAF3, thereby inhibiting TRAF3 proteolysis and preventing aberrant non-canonical NF-kappaB activation. Consequently, the OTUD7B deficiency results in B-cell hyper-responsiveness to antigens, lymphoid follicular hyperplasia in the intestinal mucosa, and elevated host-defence ability against an intestinal bacterial pathogen, Citrobacter rodentium.     Description: The synthetic peptide NH2-Tyr-Pro-Phe-Pro-CONH2 (morphiceptin), which is the amide of a fragment of the milk protein beta-casein, has morphinelike activities and is highly specific for morphine (mu) receptors but not for enkephalin (delta) receptors. It is as active as morphine in the guinea pig ileum but much less active in the mouse and rat vas deferens. The discovery of this specific morphine receptor ligand substantiates the hypothesis of multiple opiate receptors.     Description: Immunoreactive corticotropin-releasing factor (CRF) and dynorphin-(I-8) were visualized in rat hypothalamus by immunohistofluorescence with specific antibodies. In brains from colchicine-treated, adrenalectomized rats, neuronal perikarya with immunoreactive CRF were observed in the paraventricular nucleus of the hypothalamus. The CRF occurred together with the dynorphin-(1-8). However, the CRF immunoreactivity occurred only in a subpopulation of the dynorphin-(1-8) immunoreactive cells.     Notes: A comparative study is reported on separation of series of mono-, di-, and trisubstituted methyl 5β-cholanates, which differ only in the position and stereochemistry of hydroxyl or keto groups at position and stereochemistry of hydroxyl or keto groups at positions C-3, C-7, and/or C-12, by reversed-phase [with chemically-bonded (C-18) silica gel] and normal-phase (silica gel) high-performance thin-layer chromatography (HPTLC). Methnol (or acetonitrile)/water systems were employed as mobile phase. Reversed-phase HPTLC found to be particulary effective for separation of the stereoisomers of di- and trisubstituted compounds whereas the less polar monosubstituted isomers are well resolved in normal-phase HPTLC.

    Notes: A theoretical study was carried out to achieve a better understanding of the oscillatory behavior of a gas bubble growing (or collapsing) in a viscoelastic liquid, by taking into account both the hydrodynamic and diffusion effects. The Zaremba-DeWitt model was chosen to represent the rheological properties of the suspending medium. The finite difference method was employed to solve the governing system equations.The computational results show that, in the case of very fast diffusion (i.e., constant bubble pressure), the oscillatory behavior of a bubble takes place only when the ratio of the initial pressure difference between the gas bubble and the liquid phase to the elastic modulus of the suspending medium is below a certain critical value. On the other hand, in the case of very slow diffusion, the oscillatory behavior of a bubble persists, regardless of the magnitude of the rheological properties of the suspending medium. Our study indicates further that the diffusivity of a gas has a profound influence on the occurrence of oscillatory behavior, that the elastic property of the suspending medium enhances oscillatory behavior while its viscosity plays the opposite role, and that even a Newtonian medium can give rise to an oscillatory pattern of bubble growth (or collapse), although it dampens out very quickly.

    Notes: The absorption rate of sulfur dioxide from a gas mixture of SO2 and N2 into aqueous acetic acid and adipic acid solutions at pH 4 to 6 has been measured at 25°C in a continuous stirred vessel with an unbroken gas-liquid interface. The experimental data have been modeled by approximate surface renewal theory with multiple equilibrium reactions. The model has been used to evaluate the relative effectiveness of eleven organic acid additives at typical limestone scrubber conditions. Adipic, sulfopropionic, sulfosuccinic, and hydroxypropionic acids appear to be the most promising alternatives.

    Notes: The dynamic pulse-response technique was used to measure effective diffusivities under nonreacting conditions in two, unsulfided, extrudate-type, hydrodesulfurization catalyst pellets of different porosities. Pore-volume distribution data showed a bidisperse pore structure with a broad pore-size range (100 μm to 3 nm).The main purpose of the study was to evaluate various methods for calculating the tortuosity factor, τ. For this particular catalyst, the most constant and unreasonable values of τ were obtained by supposing that diffusion occurred in all the pore volume (micro and macropores) and by summing the combined Knudsen and bulk contributions over each increment of pore volume.
