    Notes: Twenty eight native vegetable oils were analyzed for polycyclic aromatic hydrocarbons. Generally PAH concentrations were low, but olive oils showed significantly higher contamination by light PAH ranging from 53 to 105.6 μg/kg. Muscle and liver samples of bream from the river Elbe contained little or no detectable amounts of PAH, whereas fresh and canned oysters and mussels showed higher contaminations. Highest PAH concentrations have been determined in “smoked oysters in oil” with 75.8 μg/kg benzo[a] pyrene for the oil and 12.2 μg/kg for the oyster meat.

    Notes: The occurrence of the highly toxic non-ortho substituted (planar) chlorobiphenyls (CBs) at low levels in wildlife, (human) fat, and fish generates the need for a routine analytical procedure. The use of a fully automated system for the separation of planar and non-planar CBs, consisting of a gel permeation chromatography system, a sample preparation system employing extraction columns, and a porous graphitic carbon HPLC column is described, and tested with horse fat. Gas chromatography on capillary columns, with electron-capture or high resolution mass spectrometric detection, is used for quantitation of the CBs.

    Notes: The feasibility of using ethyl acetate for the desorption of trace pollutants from a liquid chromatographic precolumn on-line into a diphenyltetramethyldisilazane-deactivated retention gap and, subsequently analysis by means of capillary gas chromatography has been demonstrated. First 5% of methanol are added to the water sample to prevent sorption of analytes onto parts of the preconcentration system. About 1 ml of this aqueous sample is injected onto a precolumn containing a polymeric stationary phase, using water-methanol (95:5, v/v) for transport and clean-up. The precolumn is desorbed with ethyl acetate and a fraction of 75 μl is injected on-line into the retention gap; separation is then achieved on a capillary CP Sil 19 column. No breakthrough of the test compounds was observed in the preconcentration step. The recovery was quantitative and the response obtained with flame ionization detection was linear in the range 0.1-100 ng/ml. The effect of varying the sorption flow rate on the recovery was studied. The system was applied to the analysis of river water.

    Notes: The use of chloroacetonitrile as eluent additive in thermospray liquid chromatography/mass spectrometry in the negative ion mode using filament-off, filament-on and discharge ionization modes has been studied with chlorophenols and chlorinated phenoxy acids as test compounds. In the absence of chloroacetonitrile from the eluent, different base peaks were observed corresponding to [ M — H]-, [ M + H]- and [ M + CH3COOO]-, while in the presence of 2% chloroacetonitrile [M + Cl] - was the base peak in most cases when using the filament-on and discharge ionization modes and had a maximum value of 30% relative abundance in the filament-off mode. Most chloride attachment ([ M + Cl] - ion) was always observed in the filament-on mode. Other ions observed were adduct ions with the ionizing additive (ammonium acetate) such as [M + CH3COO]- and [ M + (CH3COOH). (CH3COO)]- when the eluent contained no chloroacetonitrile and [ M + (CH3COOH). Cl]- when 2% chloroacetonitrile was present. The use of chloroacetonitrile in the eluent provides additional structural information in thermospray liquid chromatography/negative ion mass spectrometry. It does not cause any appreciable loss in sensitivity during flow injection or chromatographic analysis.

    Notes: A system is described which accelerates the solvent evaporation rate in the retention gap. The evaporation is due to a saturation effect of the carrier gas stream, and a considerable increase in evaporation rate is obtained by inserting a split outlet between the retention gap and the capillary separation column in the gas chromatograph. By varying the backpressure of the spliter device, the flow rate through the retention gap can be adjusted and so too the evaporation rate. The evaporation process was monitored by inserting a dectecter in the split outlet line. The technique was applied to the on-line LC trace enrichment/GC analysis of water containing a mixture of polycyclic aromatic hydrocarbons.

    Notes: The biocompatibility of dense Silastic implants and porous implants made of Estane 5714 F1 polyether urethane, polypropylene oxide, and an HPOE/PBT segmented polyether polyester copolymer was evaluated during an induced Staphylococcus aureus middle ear infection.The middle ear response to infection seemed not to be affected by the presence of implants made of either of the polymers. Light microscopical morphometry and transmission electron microscopy showed degradation of the porous implants under study, but not of Silastic implants, which were invariably surrounded by a fibrous capsule. This finding, combined with the degree of porous implant degradation, the composition of the tissues surrounding the implants, and the tissue/biomaterial interface reactions are consistent with the results obtained in the noninfected middle ear. Round-cell infiltrates however, were predominantly associated with implants made of polypropylene oxide and HPOE/PBT copolymer; while the presence of (phagocytosed) microbial debris was associated with copolymer.The present findings indicate that with respect to implant behavior in infected surroundings Estane is the best porous material, whereas the behavior of Silastic implants did not deviate from that in noninfected ears.

    Notes: The tissue/biomaterial interface reactions of four elastomers - selected as candidates for scaffolding for tympanic membrane tissue in a total alloplastic middle ear prosthesis - were studied at the electron microscopical level after implantation in the rat middle ear. Time-dependent changes in the phagocyte/polymer interface suggested degradation of porous implants made of Estane polyether urethane, polypropylene oxide, and a poly(ethylene oxide hydantoin) and poly(tetramethylene terephthalate) segmented polyether polyester copolymer (HPOE/PBT copolymer), but not of dense Silastic silicone rubber implants. Silastic was always encapsulated in fibrous tissue. Contact between fibrous tissue and HPOE/PBT copolymer or Estane was established in the third month, but fibrous tissue was never seen close to polypropylene oxide. Bone made contact only with Estane and HPOE/PBT copolymer implants. The bone/copolymer interface showed an electron-dense layer morphologically similar to that seen between bone and hydroxyapatite ceramic, suggesting that with respect to bone HPOE/PBT copolymer behaves like a bioactive implant material. The electron-dense layer was absent at the bone/Estane interface. Estane and especially HPOE/PBT copolymer seem to be suitable as alloplastic tympanic membrane because of their interface behavior with respect to fibrous tissue and bone.

    Notes: Epithelial reactions to Silastic, Estane polyether urethane, polypropylene oxide, and a poly(ethylene oxide hydantoin) and poly(tetramethylene terephthalate) segmented polyether polyester copolymer were investigated after implantation in tympanic membranes and submucosa of noninfected and Staphylococcus aureus-infected rat middle ears. Porous implants made of Estane and polypropylene oxide were completely covered by tympanic-membrane connective tissue, epidermis, and epithelium in 2 weeks and those made of copolymer in between 2 and 4 weeks postoperatively. Silastic implants, which were dense, were not enveloped by tympanic-membrane tissue but rejected. Starting in the 6th postoperative month the proliferative activity and structure of both the tympanic membrane epithelium and epidermis became normal except for the presence of iron-containing secretory epithelium near polypropylene oxide. After initial swelling caused by the surgical trauma, neither the proliferative activity nor the composition of the epithelium covering submucosal implants was affected by the presence of any of the biomaterials. Infection of middle ears bearing implants induced epithelial reactions similar to those associated with infected middle ears without an implant.
