    Notes: Summary Stimulated by the rather sparse information in the literature on cellular changes induced by EHDP, we carried out electron microscopic investigations on young bone tissue and on de novo bone formation. Cellular changes could be observed during continuous administration of EHDP. The osteoblasts demonstrated temporary storing of crystalloid structures in the mitochondria, and atypical osteocytes showed persistent changes indicative of hyperactivity. The osteoclasts exhibited varying ultrastructural features with respect to the number and appearance of nuclei, Golgi, RER, and lysosomes. These changes under the influence of EHDP could be an indication of altered activity of the osteoclast. The possible interference of EHDP with bone cell metabolism is discussed.

    Notes: Summary The expression of cytokeratin polypeptides in the different epithelia of the developing inner ear of the rat from 12 days post conception to 20 days after birth was analysed immunohistochemically, using a panel of monoclonal antibodies. Throughout the development of the complex epithelial lining of the inner ear originating from the otocyst epithelium, only cytokeratins which are typical of simple epithelia were expressed. Cytokeratins 8, 18, and 19 were detectable shortly after the formation of the otocyst from the ectoderm (12 dpc), whereas cytokeratin 7 expression was delayed and first appeared in the vestibular portion and subsequently in the developing cochlear duct. During the development of the different types of specialized cells, differentiation-dependent modulation of the cytokeratin expression patterns was observed. In the mature inner ear, the specialized cell types displayed a function-related cytokeratin expression profile, both in the cochlear and vestibular portion. Cytokeratin expression in the flat epithelium of the vestibular portion suggests a more complex composition of this epithelium than has been established from routine morphology. Remarkably, the cochlear sensory cells were apparently devoid of cytokeratins, but no final conclusion could be drawn on the presence of cytokeratins in the sensory cells of the vestibular portion, because of the difficulty to delineate the cell borders between sensory cells and supporting cells.

    Notes: Abstract When a viscous fluid is extruded from a capillary or an annular die, the thickness of the fluid jet is in general unequal to the width of the die. This phenomenon is called “die-swell” and is studied in this paper for a die made up of two parallel plates. It is assumed that no slip will occur between the fluid and the plates, and that the pressure in the space into which the fluid is emitted is constant and uniform. The fluid surface is a free streamline. Its shape is calculated with the use of complex-function theory and conformal-mapping techniques. The predicted ratio of swell is found to be in full agreement with known finite-element results.

    Notes: Abstract The injection of a viscous fluid into a mould formed by two parallel plates is considered. The flow front is supposed to move at constant speed. It is assumed that there is complete adherence between the fluid and the mould walls, and that the environmental pressure is constant. For a Newtonian fluid the problem is described in terms of two analytic complex functions. The shape of the fluid surface is calculated by means of a conformal-mapping technique, which leads to a Hilbert problem. The results are compared with known finite-element simulations.

    Notes: Summary The secretory behaviour of rat nasal glands, under normal conditions and after the application of cholinergic drugs, has been studied using morphological and radiobiochemical techniques. Autoradiography and electrophoresis provide evidence for the selective incorporation of 3H-arginine into the glycoprotein containing fraction of the nasal glandular secretion. Radiobiochemical experiments show that labelled arginine is rapidly incorporated into the acinar cells of unstimulated glands, although it takes approximately 4 h before the labelled secretory proteins leave the cells. The secretion of proteins is stimulated by the parasympathetic agonist pilocarpine, whose main action is to promote discharge. Histological sections show a depletion of secretory granules after pilocarpine treatment. The cholinergic antagonist atropine inhibits the secretion; the acinar cells are completely filled with secretory granules following this treatment. The time course of the events following atropine administration suggests that there is no feed-back system controlling glycoprotein synthesis. The techniques employed here therefore appear to be useful for studying the effects of drugs that interfere with the secretory activity of the nasal glands.

    Notes: Summary The middle ear cavity of the rat is lined with ciliated and squamous epithelium. The arrangement of the ciliated cells, interspersed with secretory cells, in distinct tracts and their continuity with the ciliated epithelium of the Eustachian tube, suggests the existence of a mucociliary transport system for cleaning the middle ear cleft. The secretory cells produce either neutral or sulphated glycoproteins, dependent on their location. In addition to these secretions, the epithelium of the lower part of the Eustachian tube is bathed with secretory products of seromucous glands. Also in the areas with squamous epithelium, numerous small secretory cells, the character of which is only identifiable with the electronmicroscope, are present. It is concluded that the middle ear lining can be considered as a locally modified respiratory epithelium. Blockade of the mucociliary transport system, supposedly a crucial aetiological factor in secretory otitis media, by obstruction of the Eustachian tube, induces pathogenic behaviour of microorganisms normally present in the middle ear. This results in either a transient or a longstanding infective middle ear disease, associated with a large variety of changes of the mucosa, especially with respect to the secretory activity. The data obtained indicate that the increased secretory activity encountered in secretory otitis media cannot be attributed to the isolated effect of tubal occlusion, but rather to an infective process.

    Notes: The biological performance of macroporous β-whitlockite implanted in the rat middle ear was evaluated. The material was studied in the non-infected middle ear and in middle ears infected by Staphylococcus aureus. β-whitlockite was quickly covered by a normal mucosa. One week post-operatively the macropores were filled with exudate, fibrous tissue, and a small quantity of bone. Six months after the operation the greater part of the macropore area was filled with bone (74%); fibrous tissue accounted for 20%, and exudate for 5%. In histological sections, the macropore area of β-whitlockite had increased by 68% after six months, indicating biodegradation. Macrophages and multinucleated cells were present in the vicinity of the implant and played a role in this biodegradation. Besides cytoplasmic vacuoles containing calcium phosphate, the cells showed smaller granules containing trace elements originally present in the implant material, such as silicon, titanium, aluminum, iron, and magnesium.
